Abstract

Compositions, recombinant vaccines and live attenuated pathogens comprising one or more isolated nucleic acid molecules that encode an immunogen in combination with an isolated nucleic acid molecule that encodes IL-28 or a functional fragment thereof are disclosed. Methods of inducing an immune response in an individual against an immunogen, using such compositions are disclosed.

Claims (18)

1. A recombinant immunogenic pharmaceutical composition comprising an immunogenic amount of an isolated nucleic acid construct comprising a nucleotide sequence that encodes a viral protein operably linked to regulatory elements, wherein said nucleic acid construct further comprises a nucleotide sequence that encodes an IgE signal peptide and a codon-optimized and RNA-optimized nucleotide sequence that encodes IL-28B or a functional fragment thereof, wherein said viral protein is from a virus selected from the group consisting of: influenza virus, human immunodeficiency virus, hepatitis C virus, West Nile Virus and hepatitis B virus.

2. An injectable immunogenic pharmaceutical composition comprising the composition of claim 1 .

3. The recombinant immunogenic pharmaceutical composition of claim 1 , wherein the viral protein is from an influenza virus.

4. The recombinant immunogenic pharmaceutical composition of claim 1 , wherein the viral protein is from HIV.

5. The recombinant immunogenic pharmaceutical composition of claim 4 wherein said recombinant pharmaceutical composition comprises a recombinant vaccinia vector.

A method of inducing an immune response in an individual against a viral protein from a virus selected from the group consisting of: influenza virus, human immunodeficiency virus, hepatitis C virus, West Nile Virus and hepatitis B virus comprising administering to said individual a recombinant vaccine of claim 1 .

A live attenuated virus comprising an isolated nucleic acid construct comprising a nucleotide sequence that encodes a viral protein operably linked to regulatory elements, wherein said nucleic acid construct further comprises a nucleotide sequence that encodes an IgE signal peptide and a codon-optimized and RNA-optimized nucleotide sequence that encodes IL-28B or a functional fragment thereof.

8. A method of immunizing an individual against a pathogen comprising administering to said individual the live attenuated pathogen of claim 7 .

9. The recombinant immunogenic pharmaceutical composition of claim 1 , wherein the nucleotide sequence that encodes IL-28B or a functional fragment thereof is operably linked to a promoter.

10. The recombinant immunogenic pharmaceutical composition of claim 9 , wherein the promoter is a SV40 promoter, MMTV promoter, L TR promoter, or CMV immediate early promoter.

11. The live attenuated virus of claim 7 , wherein the virus is an influenza virus.

12. The live attenuated virus of claim 7 , wherein the virus is human immunodeficiency virus, hepatitis C virus, West Nile Virus or a hepatitis B virus.

An immunogenic pharmaceutical composition comprising an immunogenic amount of a nucleic acid construct comprising a nucleotide sequence that encodes a viral protein operably linked to regulatory elements, wherein said nucleic acid construct further comprises a nucleotide sequence that encodes an IgE signal peptide and a codon-optimized and RNA-optimized nucleotide sequence that encodes IL-28B or a functional fragment thereof.

14. The immunogenic pharmaceutical composition of claim 13 , wherein the viral protein is from a virus selected from the group consisting of: influenza virus, human immunodeficiency virus, hepatitis C virus, West Nile Virus and hepatitis B virus.

15. An injectable immunogenic pharmaceutical composition comprising the pharmaceutical composition of claim 13 .

16. A recombinant immunogenic pharmaceutical composition comprising

an immunogenic amount of an isolated nucleic acid construct comprising a nucleotide sequence that encodes a viral protein operably linked to regulatory elements, wherein said viral protein is from a virus selected from the group consisting of: influenza virus, human immunodeficiency virus, hepatitis C virus, West Nile Virus and hepatitis B virus; and

an immunogenic amount of an isolated nucleic acid construct encoding an IgE signal peptide and a codon-optimized and RNA-optimized immunomodulating protein, wherein the immunomodulating protein consists of IL28B or a fragment thereof.
