Abstract

Provided herein are compositions, methods and uses involving antibodies that specifically bind to Programmed Death-1 (PD-1) and modulate the expression and/or activity of PD-1.

Claims (197)

1. An antibody or antigen-binding fragment thereof that binds to PD-1, wherein the antibody or antigen-binding fragment comprises:

a light chain variable region (VL) comprising:

a VL complementarity determining region 1 (CDR1) comprising SEQ ID NO:1,

a VL CDR2 comprising SEQ ID NO:2, and

a VL CDR3 comprising SEQ ID NO:3; and

a heavy chain variable region (VH) comprising:

a VH CDR1 comprising SEQ ID NO:4,

a VH CDR2 comprising SEQ ID NO:5, and

a VH CDR3 comprising SEQ ID NO:6.

2. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a VL comprising an amino acid sequence of SEQ ID NO:8.

3. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a VL comprising an amino acid sequence of SEQ ID NO:10.

4. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:11.

5. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:12.

6. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:13.

7. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:8; and

a VH comprising an amino acid sequence of SEQ ID NO:11.

8. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:8; and

a VH comprising an amino acid sequence of SEQ ID NO:12.

9. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:8; and

a VH comprising an amino acid sequence of SEQ ID NO:13.

10. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:10; and

a VH comprising an amino acid sequence of SEQ ID NO:11.

11. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:10; and

a VH comprising an amino acid sequence of SEQ ID NO:12.

12. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises

a VL comprising an amino acid sequence of SEQ ID NO:10; and

a VH comprising an amino acid sequence of SEQ ID NO:13.

13. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

14. The antibody or antigen-binding fragment of claim 7 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

15. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

16. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

17. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

18. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

19. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

20. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

21. The antibody or antigen-binding fragment of claim 7 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

22. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

23. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

24. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

25. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

26. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

27. The antibody or antigen-binding fragment of 1 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

28. The antibody or antigen-binding fragment of 7 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

29. The antibody or antigen-binding fragment of 8 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

30. The antibody or antigen-binding fragment of 9 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

31. The antibody or antigen-binding fragment of 10 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

32. The antibody or antigen-binding fragment of 11 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

33. The antibody or antigen-binding fragment of 12 , wherein the antibody or antigen-binding fragment comprises a human IgG4 Fc region, or a mutant thereof.

36. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a human IgG4P Fc region.

37. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a human IgG4P Fc region.

38. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a human IgG4P Fc region.

39. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a human IgG4P Fc region.

40. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a human IgG4P Fc region.

42. The antibody or antigen-binding fragment of claim 7 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

43. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

44. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

45. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

46. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

47. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a human IgG4PE Fc region.

48. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

49. The antibody or antigen-binding fragment of claim 7 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

50. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

51. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

52. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

53. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

54. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

55. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

56. The antibody or antigen-binding fragment of claim 7 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

57. The antibody or antigen-binding fragment of claim 8 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

58. The antibody or antigen-binding fragment of claim 9 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

59. The antibody or antigen-binding fragment of claim 10 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

60. The antibody or antigen-binding fragment of claim 11 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

61. The antibody or antigen-binding fragment of claim 12 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

62. The antibody or antigen-binding fragment of claim 48 , wherein the antibody or antigen-binding fragment further comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

63. The antibody or antigen-binding fragment of claim 49 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

64. The antibody or antigen-binding fragment of claim 50 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

65. The antibody or antigen-binding fragment of claim 51 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

66. The antibody or antigen-binding fragment of claim 52 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

67. The antibody or antigen-binding fragment of claim 53 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

68. The antibody or antigen-binding fragment of claim 54 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

69. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a light chain comprising an amino acid sequence of SEQ ID NO:31.

70. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a heavy chain comprising an amino acid sequence of SEQ ID NO:32.

71. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises:

a light chain comprising an amino acid sequence of SEQ ID NO:31; and

a heavy chain comprising an amino acid sequence of SEQ ID NO:32.

72. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a heavy chain comprising an amino acid sequence of SEQ ID NO:33.

73. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises:

a light chain comprising an amino acid sequence of SEQ ID NO:31; and

a heavy chain comprising an amino acid sequence of SEQ ID NO:33.

74. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a heavy chain comprising an amino acid sequence of SEQ ID NO:34.

75. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises:

a light chain comprising an amino acid sequence of SEQ ID NO:31; and

a heavy chain comprising an amino acid sequence of SEQ ID NO:34.

76. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a heavy chain comprising an amino acid sequence of SEQ ID NO:35.

77. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises:

a light chain comprising an amino acid sequence of SEQ ID NO:31; and

a heavy chain comprising an amino acid sequence of SEQ ID NO:35.

78. The antibody or antigen-binding fragment of claim 1 , wherein the antibody is a monoclonal antibody.

79. The antibody or antigen-binding fragment of claim 1 , wherein the antibody is a humanized antibody.

80. The antibody or antigen-binding fragment of claim 1 , wherein the antibody is a human antibody.

81. The antibody or antigen-binding fragment of claim 1 , wherein the antibody is a chimeric antibody.

82. The antibody or antigen-binding fragment of claim 79 , wherein the humanized antibody is a deimmunized antibody.

83. The antibody or antigen-binding fragment of claim 79 , wherein the humanized antibody is a composite human antibody.

84. The antibody or antigen-binding fragment of claim 1 wherein the antibody or antigen-binding fragment is a Fab, a Fab′, a F(ab′) 2 , a Fv, a scFv, a dsFv, a diabody, a triabody, a tetrabody, or a multispecific antibody formed from antibody fragments.

85. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment is conjugated to an agent.

86. The antibody or antigen-binding fragment of claim 85 , wherein the agent is selected from the group consisting of a radioisotope, a metal chelator, an enzyme, a fluorescent compound, a bioluminescent compound, and a chemiluminescent compound.

87. An antibody or antigen-binding fragment thereof that binds to PD-1, wherein the antibody or antigen-binding fragment comprises:

a VL comprising:

a VL CDR1 comprising SEQ ID NO:7,

a VL CDR2 comprising SEQ ID NO:2, and

a VL CDR3 comprising SEQ ID NO:3; and

a VH comprising:

a VH CDR1 comprising SEQ ID NO:4,

a VH CDR2 comprising SEQ ID NO:5, and

a VH CDR3 comprising SEQ ID NO:6.

88. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a VL comprising an amino acid sequence of SEQ ID NO:9.

89. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:11.

90. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:12.

91. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a VH comprising an amino acid sequence of SEQ ID NO:13.

98. The antibody or antigen-binding fragment of claim 94 , wherein the antibody or antigen-binding fragment comprises a human IgG1 Fc region, or a mutant thereof.

99. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

100. The antibody or antigen-binding fragment of claim 92 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

101. The antibody or antigen-binding fragment of claim 93 , wherein the antibody or antigen-binding fragment comprises a human IgG1-K322A Fc region.

116. The antibody or antigen-binding fragment of claim 92 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

117. The antibody or antigen-binding fragment of claim 93 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

118. The antibody or antigen-binding fragment of claim 94 , wherein the antibody or antigen-binding fragment comprises a heavy chain Fc region comprising an amino acid sequence selected from the group consisting of SEQ ID NO:36, SEQ ID NO:37, SEQ ID NO:38, SEQ ID NO:39 or SEQ ID NO:40.

119. The antibody or antigen-binding fragment of claim 87 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

120. The antibody or antigen-binding fragment of claim 92 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

121. The antibody or antigen-binding fragment of claim 93 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

122. The antibody or antigen-binding fragment of claim 94 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

123. The antibody or antigen-binding fragment of claim 115 , wherein the antibody or antigen-binding fragment further comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

124. The antibody or antigen-binding fragment of claim 116 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

125. The antibody or antigen-binding fragment of claim 117 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.

126. The antibody or antigen-binding fragment of claim 118 , wherein the antibody or antigen-binding fragment comprises a light chain constant region comprising an amino acid sequence of SEQ ID NO:41.
