LXR agonists and uses thereof

1. A compound of Formula I:

wherein e is 0, 1, 2, 3, or 4;

R A is optionally substituted C 1 -C 6 acyl, optionally substituted C 1 -C 6 alkyl, optionally substituted C 2 -C 6 alkenyl, optionally substituted C 2 -C 6 alkynyl, optionally substituted C 2 -C 6 heteroalkenyl, optionally substituted C 2 -C 6 heteroalkynyl, optionally substituted C 3 -C 10 cycloalkyl, optionally substituted C 4 -C 10 cycloalkenyl, optionally substituted C 8 -C 12 cycloalkynyl, optionally substituted C 6 -C 10 aryl, optionally substituted C 6 -C 10 aryl C 1 -C 6 alkyl, optionally substituted C 2 -C 9 heteroaryl, optionally substituted C 2 -C 9 heteroaryl C 1 -C 6 alkyl, optionally substituted C 2 -C 9 heterocyclyl, 8F optionally substituted C 2 -C 9 heterocyclyl C 1 -C 6 alkyl;

each R B is independently hydroxyl, optionally substituted amino, halo, thiol, optionally substituted amino acid, optionally substituted C 1 -C 6 acyl, optionally substituted C 1 -C 6 alkyl, optionally substituted C 2 -C 6 alkenyl, optionally substituted C 2 -C 6 alkynyl, optionally substituted C 1 -C 6 heteroalkyl, optionally substituted C 2 -C 6 heteroalkenyl, optionally substituted C 2 -C 6 heteroalkynyl, optionally substituted C 3 -C 10 cycloalkyl, optionally substituted C 8 -C 12 cycloalkenyl, optionally substituted C 4 -C 10 cycloalkynyl, optionally substituted C 6 -C 10 aryl, optionally substituted C 6 -C 10 aryl C 1 -C 6 alkyl, optionally substituted C 2 -C 9 heteroaryl, optionally substituted C 2 -C 9 heteroaryl C 1 -C 6 alkyl, optionally substituted C 2 -C 9 heterocyclyl, or optionally substituted C 2 -C 9 heterocyclyl C 1 -C 6 alkyl; and

L is —O—;

a is 2 or 3;

b is 1;

c is 0;

d is 1;

each R 1 , R 2 , R 6 , R 7 , R 8 , and R 9 are independently hydrogen or optionally substituted C 1 -C 6 alkyl;

R 3 is hydrogen, halo, or optionally substituted C 1 -C 6 alkyl;

R 4 is hydrogen, optionally substituted C 1 -C 6 alkyl, optionally substituted C 3 -C 10 cycloalkyl, or optionally substituted C 6 -C 10 aryl;

R 5 is optionally substituted C 6 -C 10 aryl; and

B is optionally substituted C 6 -C 10 aryl;

or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 , wherein each R 6 and R 7 is hydrogen.

3. The compound of claim 1 , wherein R 3 is hydrogen.

4. The compound of claim 1 , wherein R 4 is hydrogen, phenyl, cyclohexyl, or methyl.

5. The compound of claim 1 , wherein R 5 is phenyl.

6. The compound of claim 1 , wherein each R 8 and R 9 is hydrogen.

7. The compound of claim 1 , wherein —(CR 1 R 2 ) a — has the structure:

9. The compound of claim 1 , wherein said compound has the structure:

10. The compound of claim 1 , wherein said optionally substituted C 6 -C 10 aryl is 2-chloro-3-trifluoromethyl-phenyl, 2-fluoro-3-trifluoromethyl-phenyl, 3-(1,1,2,2-tetrafluoroethoxy)-phenyl, 3-trifluoromethyl-phenyl, 3-trifluoromethyl-4-fluoro-phenyl, or 3-trifluoromethoxy-phenyl.

11. The compound of claim 10 , wherein B is 2-chloro-3-trifluoromethyl-phenyl, 2-fluoro-3-trifluoromethyl-phenyl, or 3-trifluoromethyl-phenyl.

12. The compound of claim 11 , wherein B is 2-chloro-3-trifluoromethyl-phenyl.

13. A compound, or pharmaceutically acceptable salt thereof, selected from:

14. The compound of claim 1 , wherein at least one of R A or R B is optionally substituted C 1 -C 6 heteroalkyl, wherein the optionally substituted C 1 -C 6 heteroalkyl is —OR V , wherein R V is:

wherein each R′ and R″ is independently hydrogen, optionally substituted C 1 -C 6 alkyl, or optionally substituted C 6 -C 10 aryl C 1 -C 6 alkyl.

15. A method of ameliorating one or more symptoms of cancer comprising administering an effective amount of a compound of claim 1 , or pharmaceutically acceptable salts thereof to a subject in need thereof.

16. A method of ameliorating one or more symptoms of cancer comprising contacting a cell with an effective amount of a compound of claim 1 , or pharmaceutically acceptable salts thereof.

17. The method of claim 15 , wherein said cancer is breast cancer, colon cancer, renal cell cancer, non-small cell lung cancer, hepatocellular carcinoma, gastric cancer, ovarian cancer, pancreatic cancer, esophageal cancer, prostate cancer, sarcoma, glioblastoma, diffuse large B-cell lymphoma, acute myeloid leukemia, multiple myeloma, or melanoma.

18. The method of claim 15 , wherein said method further comprises administration of an additional anticancer therapy.

19. The method of ameliorating one or more symptoms of a neurological disorder comprising administering an effective amount of a compound of claim 1 , or pharmaceutically acceptable salts thereof to a subject in nee thereof, wherein said neurological disorder is Alzheimer's disease.
