Abstract

The instant invention relates to a process for the preparation of a pharmaceutical composition in sustained-release tablet form comprising from 600 milligrams to 2400 milligrams of Pirfenidone (PFD), in such a way that the drug is bioavailable during an extended period of time of 12 hours from its administration. In this way, the anti-fibrotic and anti-inflammatory action of the drug Pirfenidone is optimized. Moreover, the instant invention offers advantages and a higher therapeutic efficacy compared to other pharmaceutical forms of Pirfenidone for oral administration and its therapeutic application in the regression of chronic renal failure secondary to primary glomerulosclerosis; it shows a better activity with regard to the reduction and/or regression of deleterious effects in breast capsular contracture observed after the surgical implantation of breast implants in humans and has an important anti-TNF-α and anti-TGF-β1 action for the treatment of hepatic fibrosis.

Claims (24)

1. A sustained-release tablet comprising pirfenidone, sodium stearyl fumarate, low viscosity hydroxypropylmethylcellulose (HPMC), high viscosity hydroxypropylmethylcellulose (HPMC), and one or more excipients, for sustained release of pirfenidone over 12 hours.

2. The tablet of claim 1 , wherein the tablet comprises 100-850 mg ±5% of pirfenidone.

3. The tablet of claim 1 , wherein the tablet comprises 600 mg ±5% of pirfenidone.

4. The tablet of claim 1 , wherein the tablet further comprises microcrystalline cellulose.

5. The tablet of claim 1 , wherein the tablet further comprises silicon dioxide.

6. The tablet of claim 1 , wherein the tablet further comprises microcrystalline cellulose and silicon dioxide.

7. The tablet of claim 1 , for oral administration.

8. The tablet of claim 6 , wherein the tablet comprises the following compression parameters: average weight of 850 mg ±5%; hardness of maximum 10 kgF; and brittleness that is lower than 1%.

9. The tablet of claim 1 , wherein the tablet comprises 100 mg ±5% of pirfenidone.

10. The tablet of claim 1 , wherein the tablet comprises 6.2 mg ±5% of sodium stearyl fumarate.

11. The tablet of claim 1 , wherein the tablet comprises 70.0 mg ±5% of low viscosity HPMC.

12. The tablet of claim 1 , wherein the tablet comprises 46.5 mg ±5% of high viscosity HPMC.

13. The tablet of claim 4 , wherein the tablet comprises 118.8 mg ±5% of microcrystalline cellulose.

14. The tablet of claim 5 , wherein the tablet comprises 8.5 mg ±5% of silicon dioxide.

15. The tablet of claim 1 , wherein the tablet comprises 100-850 mg of pirfenidone.

16. The tablet of claim 1 , wherein the tablet comprises 600 mg of pirfenidone.

17. The tablet of claim 1 , wherein the tablet comprises 200 mg of pirfenidone.

18. The tablet of claim 1 , wherein the tablet comprises 400 mg of pirfenidone.

19. The tablet of claim 1 , wherein the tablet comprises 6.2 mg of sodium stearyl fumarate.

20. The tablet of claim 1 , wherein the tablet comprises 70.0 mg of low viscosity HPMC.

21. The tablet of claim 1 , wherein the tablet comprises 46.5 mg of high viscosity HPMC.

22. The tablet of claim 4 , wherein the tablet comprises 118.8 mg of microcrystalline cellulose.

23. The tablet of claim 5 , wherein the tablet comprises 8.5 mg of silicon dioxide.

24. The tablet of claim 1 , wherein the tablet comprises 600 mg pirfenidone, 118.8 mg of microcrystalline cellulose, 70.0 mg low viscosity hydroxypropylmethylcellulose (HPMC), 46.5 mg high viscosity hydroxypropylmethylcellulose (HPMC), 8.5 mg silicon dioxide, and 6.2 mg sodium stearyl fumarate.
