CD8-specific antibody constructs and compositions thereof
Disclosed herein are antibodies or antigen binding fragments thereof that specifically bind human CD8. Also disclosed are fusion proteins comprising a Henipavirus glycoprotein G and CD8 antibodies for targeting and transducing cells expressing CD8. Viral vectors and other compositions containing the fusion proteins, as well as methods of using the fusion proteins, are also disclosed.
1. A viral vector targeting CD8+ T cells, comprising:
i) an antibody fragment that binds CD8;
ii) a henipavirus envelope glycoprotein G (G protein) or a biologically active portion thereof;
iii) a henipavirus F protein molecule or biologically active portion thereof; and
iv) one or more nucleic acid sequences encoding a chimeric antigen receptor (CAR),
wherein the antibody fragment comprises three heavy chain complementarity determining regions (HCDR1, HCDR2, and HCDR3) and three light chain complementarity determining regions (LCDR1, LCDR2, and LCDR3), wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2, and LCDR3 comprise:
a) SEQ ID NOs: 1, 70, 148, 240, 294, 333, respectively;
b) SEQ ID NOs: 5, 74, 150, 242, 295, 336, respectively;
c) SEQ ID NOs: 40, 109, 189, 240, 294, 370, respectively; or
d) SEQ ID NOs: 43, 77, 196, 270, 320, 375, respectively; and
wherein the antibody fragment is attached to the C-terminus of the G protein or the biologically active portion thereof.
2. The viral vector of claim 1 , wherein the antibody fragment is attached to the G protein via a peptide linker.
3. The viral vector of claim 2 , wherein the peptide linker is (GGGGS)n (SEQ ID NO: 626), wherein n is 1 to 10.
4. The viral vector of claim 3 , wherein the peptide linker is (GGGGS)n (SEQ ID NO: 626), wherein n is 3.
5. The viral vector of claim 1 , wherein the antibody fragment is a single chain variable fragment (scFv).
6. The viral vector of claim 1 , wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2, and LCDR3 comprise SEQ ID NOs: 40, 109, 189, 240, 294, and 370, respectively.
7. The viral vector of claim 1 , wherein the antibody fragment comprises a heavy chain variable region (VH), wherein the VH is selected from SEQ ID NOs: 405, 408, 448, and 455.
8. The viral vector of claim 1 , wherein the antibody fragment comprises a light chain variable region (VL), wherein the VL is selected from 499, 503, 542, and 549.
9. The viral vector of claim 1 , wherein the antibody fragment comprises SEQ ID NO: 1044.
10. The viral vector of claim 1 , wherein the G protein is a wild-type Nipah virus G glycoprotein (NiV-G) or a functionally active variant or a biologically active portion thereof.
11. The viral vector of claim 10 , wherein the NiV-G variant or biologically active portion thereof comprises an amino acid sequence having at least 95% sequence identity to SEQ ID NO: 600, SEQ ID NO: 618, or SEQ ID NO: 628.
12. The viral vector of claim 11 , wherein the NiV-G variant or biologically active portion thereof comprises one or more amino acid substitutions corresponding to amino acid substitutions selected from E501A, W504A, Q530A and E533A with reference to the numbering set forth in SEQ ID NO: 618.
13. The viral vector of claim 11 , wherein the NiV-G variant comprises SEQ ID NO: 607 or 635.
14. The viral vector of claim 1 , wherein the F protein is a wild-type Nipah virus F (NiV-F) protein or a functionally active variant or a biologically active portion thereof.
15. The viral vector of claim 13 , wherein the NiV-F comprises an amino acid sequence having at least 95% sequence identity to SEQ ID NO: 593, SEQ ID NO: 594, SEQ ID NO: 596, SEQ ID NO: 597, SEQ ID NO: 599, SEQ ID NO: 1092, or SEQ ID NO: 1093.
16. The viral vector of claim 1 , wherein the antibody fragment attached to the C-terminus of the G protein comprises SEQ ID NO: 1055 or SEQ ID NO: 1072.
17. The viral vector of claim 1 , wherein the CAR targets a tumor antigen selected from CD19, CD20, CD22, or BCMA.
18. The viral vector of claim 17 , wherein the CAR comprises an anti-CD19 scFv having an amino acid sequence comprising SEQ ID NO: 1075.
19. The viral vector of claim 17 , wherein the CAR comprises one or more intracellular domains selected from a CD137 (4-1BB) signaling domain, a CD28 signaling domain, and a CD3zeta signaling domain.
20. The viral vector of claim 1 , wherein the viral vector is a lentiviral vector.
21. The viral vector of claim 1 , further comprising a viral protein selected from one or more of Gag, Pol, Rev, and Tat.

The viral vector of claim 1 , further comprising one or more of the following nucleic acid sequences: a 5′ LTR, a Psi packaging element, a central polypurine tract (cPPT)/central termination sequence (CTS), a Poly A tail sequence, a posttranscriptional regulatory element, a Rev response element (RRE), and a 3′ LTR.
23. A method of treating cancer, comprising administering to a subject in need thereof an effective amount of a composition comprising a lipid particle comprising an antibody fragment that binds CD8, a henipavirus envelope glycoprotein G (G protein) or a biologically active portion thereof, a henipavirus F protein molecule or biologically active portion thereof, and a chimeric antigen receptor (CAR),
wherein the antibody fragment comprises three heavy chain complementarity determining regions (HCDR1, HCDR2, and HCDR3) and three light chain complementarity determining regions (LCDR1, LCDR2, and LCDR3), wherein the HCDR1, HCDR2, HCDR3, LCDR1, LCDR2, and LCDR3 comprise:
a) SEQ ID NOs: 1, 70, 148, 240, 294, 333, respectively;
b) SEQ ID NOs: 5, 74, 150, 242, 295, 336, respectively;
c) SEQ ID NOs: 40, 109, 189, 240, 294, 370, respectively; or
d) SEQ ID NOs: 43, 77, 196, 270, 320, 375, respectively.
24. The viral vector of claim 15 , wherein the NiV-F comprises SEQ ID NO: 593, SEQ ID NO: 594, SEQ ID NO: 596, SEQ ID NO: 597, SEQ ID NO: 599, SEQ ID NO: 1092, or SEQ ID NO: 1093.
25. The viral vector of claim 1 , wherein the antibody fragment comprises a heavy chain variable region (VH) comprising SEQ ID NO: 448 and a light chain variable region (VL) comprising SEQ ID NO: 542.
26. The viral vector of claim 9 , wherein the G protein or the biologically active portion thereof comprises SEQ ID NO: 635.
27. The viral vector of claim 26 , wherein the CAR comprises an anti-CD19 scFv having a light chain variable region comprising SEQ ID NO: 1120 and a heavy chain variable region comprising SEQ ID NO: 1125.
28. The viral vector of claim 27 , wherein the anti-CD19 scFv has an amino acid sequence comprising SEQ ID NO: 1075.
29. The viral vector of claim 28 , wherein the CAR has an amino acid sequence comprising SEQ ID NO: 1231.
30. The viral vector of claim 29 , wherein the NiV-F comprises SEQ ID NO: 593, SEQ ID NO: 594, SEQ ID NO: 596, SEQ ID NO: 597, SEQ ID NO: 599, SEQ ID NO: 1092, or SEQ ID NO: 1093.
