Abstract

The present invention relates to compounds of formula I, or pharmaceutically acceptable salts thereof, as well as compositions comprising such compounds. These compounds and compositions have uses in the treatment of psychiatric or neurological disorders. Compounds of formula I comprise at least one deuterium atom at the α-position and consequently have improved oral bioavailability relative to α-diprotic analogues.

Claims (24)

1. A compound of formula I, or a pharmaceutically acceptable salt thereof,

wherein x H is protium or deuterium,

n is selected from 1, 2, 3 or 4,

le is independently selected from —R 3 , —OR 3 , —O(CO)R 3 , —F, —Cl, —Br or —I, and

R 2 and R 3 are independently selected from C 1 -C 4 alkyl, with the proviso that when n is 1 and R 1 is 5-methoxy, x H is protium.

2. A pharmaceutical composition comprising:

a) a compound of formula I in the form of a pharmaceutically acceptable salt,

wherein x H is protium or deuterium,

n is selected from 1, 2, 3 or 4,

R 1 is independently selected from —R 3 , —O(CO)R 3 , —F, —Cl, —Br or —I, and

R 2 and R 3 are independently selected from C 1 -C 4 alkyl; and

b) a pharmaceutically acceptably excipient.

3. A composition comprising at least a first and a second compound, or pharmaceutically acceptable salts thereof, wherein the first compound is selected from a compound as defined in claim 2 , and the second compound is an undeuterated analogue of the first compound.

4. An oral dosage form comprising a compound as defined in claim 2 .

5. The pharmaceutical composition of claim 2 , wherein R 2 is methyl.

6. The pharmaceutical composition of claim 2 , wherein R 1 is independently selected from OR 3 and —O(CO)R 3 .

7. The pharmaceutical composition of claim 2 , wherein R 3 is methyl.

8. The pharmaceutical composition of claim 2 , wherein R 1 is methoxy.

9. The pharmaceutical composition of claim 2 , wherein n is 1.

10. The pharmaceutical composition of claim 9 , wherein R 1 is at the 4- or 5-position.

11. The pharmaceutical composition of claim 2 , wherein n is 1 and R 1 is 5-methoxy.

12. The pharmaceutical composition of claim 2 , wherein x H is deuterium.

13. The pharmaceutical composition of claim 2 , wherein the compound of formula I has a purity of between 99% and 100% by HPLC.

14. The pharmaceutical composition of claim 2 , wherein the pharmaceutically acceptable salt is a fumarate salt.
