Automated cell processing methods, modules, instruments, and systems
In an illustrative embodiment, automated multi-module cell editing instruments are provided to automate multiple edits into nucleic acid sequences inside one or more cells.
1. An automated stand-alone multi-module cell editing instrument comprising:
a receptacle configured to receive cells;
one or more receptacles configured to receive expression vectors, wherein the expression vectors introduce a desired DNA change to the target region and remove a proto-spacer motif (PAM) region from the target region;
a growth module for growing the cells for transformation;
a cell concentration module to concentrate the cells;
a transformation module configured to introduce nucleic acids into the cells by bead transfection;
a selection module;
a nuclease-directed editing module configured to allow the introduced nucleic acids to edit nucleic acids in the cells using an RNA-guided nuclease;
a processor configured to operate the automated multi-module cell editing instrument based on user input and/or selection of a pre-programmed script; and
an automated liquid handling system to move liquids from the receptacle to the growth module, from the growth module to the cell concentration module, from the cell concentration module to the transformation module, from the transformation module to the selection module, and from the selection module to the nuclease-directed editing module, without user intervention.
2. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the nuclease-directed editing module is also a selection module.
3. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the selection module is separate from the nuclease-directed editing module.
4. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the selection module comprises an antibiotic.
5. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the nuclease-directed editing module is also a recovery module.
6. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the nuclease-directed editing module further comprises a recovery module.
7. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the RNA-guided nuclease is MAD7.
8. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the RNA-guided nuclease editing system is an inducible system.
9. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the growth module is also the cell concentration module.
10. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the growth module is separate from the cell concentration module.
11. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the cell concentration module is also the cell transformation module.
12. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the cell concentration module is separate from the cell transformation module.
13. The automated stand-alone multi-module cell editing instrument of claim 1 , wherein the growth module, transformation module and nuclease-directed editing module are combined.
14. The automated stand-alone multi-module cell editing instrument of claim 13 , wherein the cell concentration module is combined with the growth module, transformation module and the nuclease-directed editing module.
15. An automated stand-alone multi-module cell editing instrument for RNA-guided editing of mammalian cells comprising:
a receptacle configured to receive mammalian cells;
one or more receptacles configured to receive expression vectors, wherein the expression vectors introduce a desired DNA change to the target region and remove a proto-spacer motif (PAM) region from the target region;
a growth module for growing the mammalian cells for transformation;
a cell concentration module to concentrate the mammalian cells;
a transformation module configured to introduce nucleic acids into the mammalian cells by bead transfection;
a nuclease-directed editing module configured to allow the introduced nucleic acids to edit nucleic acids in the cells using an RNA-guided nuclease;
a processor configured to operate the automated multi-module cell editing instrument based on user input and/or selection of a pre-programmed script; and
an automated liquid handling system to move liquids from the receptacle to the growth module, from the growth module to the cell concentration module, from the cell concentration module to the transformation module, and from the transformation module to the nuclease-directed editing module, without user intervention.
16. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the nuclease-directed editing module is also a selection module.
17. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the receptacle configured to receive mammalian cells is also the growth module.
18. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the nuclease-directed editing module is also a recovery module.
19. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the nuclease-directed editing module further comprises a recovery module.
20. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the RNA-guided nuclease is MAD7.
21. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the RNA-guided nuclease editing system is an inducible system.
22. The automated stand-alone multi-module cell editing instrument of claim 21 , wherein the RNA-guided nuclease editing system is a chemically-inducible system.
23. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the growth module is also the cell concentration module.
24. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the growth module is separate from the cell concentration module.
25. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the cell concentration module is also the cell transformation module.
26. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the cell concentration module is separate from the cell transformation module.
27. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the growth module, transformation module and nuclease-directed editing module are combined.
28. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the receptacle configured to receive mammalian cells is combined with the growth module, transformation module and the nuclease-directed editing module.
29. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the automated liquid handling system comprises a sipper or pipettor.
30. The automated stand-alone multi-module cell editing instrument of claim 15 , wherein the housing is configured to house all of the modules.
