Antibodies specific for sulfation sites of Wnt pathway proteins
Provided is a protein comprising an antibody binding site that binds to a sulfated epitope of a Wnt pathway protein that is not Wnt5A, Wnt11, or Wnt3a. In one embodiment, the Wnt pathway protein is the Dickkopf1 (Dkk1) protein. Further provided is a composition comprising a peptide less than 75 amino acids or amino acid analogs, the peptide consisting of a fragment of a Wnt pathway protein, such as a Dkk1, wherein the fragment is sulfated.
1. A composition comprising isolated polyclonal antibodies or fragments thereof that specifically bind the peptide sequence DNYQPYPCAEDE (SEQ ID NO:13) when the tyrosine is sulfated and do not substantially bind said peptide sequence when the tyrosine is not sulfated, made by a process comprising the steps of:
(i) immunizing a vertebrate with a synthetic peptide having the sequence DNYQPYPCAEDE (SEQ ID NO:13) wherein tyrosine is sulfated; and
(ii) isolating polyclonal antibodies from the immunized vertebrate.
2. The composition of claim 1 , comprising the fragments.
3. The composition of claim 2 , wherein the fragments are Fab antibody fragments or an F(ab)2 antibody fragments.
4. The composition of claim 1 , comprising the polyclonal antibodies.
5. The composition of claim 1 , wherein the vertebrate is a mouse, rabbit, sheep or goat.
