Abstract

Disclosed are compounds of Formula (I) N-oxides, or salts thereof, wherein X is CR 1 or N; and G, A, R 1 ; and n are defined herein. Also disclosed are methods of using such compounds as inhibitors of signaling through Toll-like receptor 7, or 8, or 9, and pharmaceutical compositions comprising such compounds. These compounds are useful in treating inflammatory and autoimmune diseases.

Claims (224)

1. A compound of Formula (I)

N-oxide, or a salt thereof, wherein:

X is CR 1 or N;

each R 1 is independently H, F, Cl, —CN, C 1-3 alkyl, C 1-2 fluoroalkyl, —OCH 3 , or —S(O) 2 (C 1-3 alkyl);

G is

a 9-membered heterocyclic ring selected from:

each R 2 is independently F, Cl, —CN, —OH, C 1-3 alkyl, C 1-2 fluoroalkyl, C 1-2 cyanoalkyl, C 1-3 hydroxyalkyl, C 1-2 aminoalkyl, —(CH 2 ) 0-2 O(C 1-3 alkyl), C 3-6 cycloalkyl, —NR x R x , —(CH 2 ) 0-2 C(O)NR x R x , —(CH 2 ) 0-2 S(O) 2 (C 1-3 alkyl), —CH 2 (C 3-6 cycloalkyl), —CH 2 (phenyl), phenyl, pyrimidinyl, or triazolyl;

A is:

(i) —OCH 2 CH 2 NR y R y ;

(ii) —CR x R x R 3 , —CH 2 CH 2 NR x R 3 , —C(O)NR x R 3 , —NR x R 3 , —NR x CH 2 CH 2 R 3 , or —OR 3 ; or

(iii) R 3 ;

R 3 is C 3-6 cycloalkyl, azetidinyl, morpholinyl, piperazinyl, piperidinyl, or pyrrolidinyl, each substituted with zero to 2 R 3a ;

each R 3a is independently C 1-4 alkyl, C 1-3 cyanoalkyl, C 1-3 fluoroalkyl, C 1-4 hydroxyalkyl, —CH 2 C(O)NR x R x , —C(O)(CH 2 ) 1-3 NR x R x , —NR y R y , (CH 2 ) 1-2 S(O) 2 (C 1-2 alkyl), —(CH 2 ) 1-2 NR x S(O) 2 (C 1-2 alkyl), —NR x CH 2 C(O)NR y R y , —NR x CH 2 CH 2 S(O) 2 (C 1-2 alkyl), —NR x (C 3-6 cycloalkyl), —NR x (oxetanyl), —CH 2 (methyltriazolyl), morpholinyl, oxetanyl, pyrrolidinyl, fluoropyrrolidinyl, hydroxy-methylpyrrolidinyl, tetrahydropyranyl, 1-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.4]octanyl, 6-oxa-2-azaspiro[3.4]octanyl, 2,2-dioxo-2-thia-6-azaspiro[3.3]heptanyl, 2-oxa-7-azaspiro[4.4]nonanyl, or azetidinyl substituted with zero to 3 substituents independently selected from F, Cl, —OH, —CN, C 1-2 alkyl, C 1-2 hydroxyalkyl, C 1-2 fluoroalkyl, and C 1-3 alkoxy;

each R x is independently H or —CH 3 ;

each R y is independently H or C 1-6 alkyl;

n is zero, 1, or 2; and

p is zero, 1, or 2.

2. The compound according to claim 1 , N-oxide, or a salt thereof,

wherein:

each R 1 is independently H, —CH 3 , —CH 2 CH 3 , —CH(CH 3 ) 2 , —OCH 3 , or —S(O) 2 CH 3 ;

G is:

each R 2 is independently —CN, —CH 3 , —OCH 3 , or pyrimidinyl;

A is:

(i) —OCH 2 CH 2 N(CH 3 ) 2 ;

(ii) —CH 2 R 3 , —CH 2 CH 2 NHR 3 , —C(O)NHR 3 , —NHR 3 , —NHCH 2 CH 2 R 3 , or —OR 3 ; or

(iii) R 3 ;

R 3 is azetidinyl, cyclobutyl, cyclohexyl, piperazinyl, piperidinyl, or pyrrolidinyl, each substituted with zero to 1 R 3a ;

R 3a is —CH 3 , —CH 2 CH 2 CH 3 , —CH(CH 3 ) 2 , —CH 2 CN, —CH 2 CH 2 CN, —CH 2 CH 2 CF 3 , —CH 2 C(CH 3 ) 2 OH, —CH 2 C(O)NH 2 , —CH 2 C(O)N(CH 3 ) 2 , —C(O)CH 2 N(CH 3 ) 2 , —C(O)CH 2 CH 2 N(CH 3 ) 2 , —C(O)CH 2 CH 2 CH 2 N(CH 3 ) 2 , —NH(CH 3 ), —N(CH 3 ) 2 , —NH(CH(CH 3 ) 2 ), —CH 2 CH 2 S(O) 2 CH 3 , —CH 2 CH 2 NHS(O) 2 CH 3 , —NHCH 2 C(O)N(CH 3 ) 2 , —NHCH 2 CH 2 S(O) 2 CH 3 , —NH(cyclopropyl), —NH(oxetanyl), —N(CH 3 ) (oxetanyl), —CH 2 (methyltriazolyl), morpholinyl, oxetanyl, pyrrolidinyl, fluoropyrrolidinyl, hydroxy-methylpyrrolidinyl, tetrahydropyranyl, 1-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.4]octanyl, 6-oxa-2-azaspiro[3.4]octanyl, 2,2-dioxo-2-thia-6-azaspiro[3.3]heptanyl, 2-oxa-7-azaspiro[4.4]nonanyl, or azetidinyl substituted with zero to 2 substituents independently selected from F, —OH, —CH 3 , —CH 2 OH, —OCH 3 , —OCH 2 CH 3 , and —OCH(CH 3 ) 2 ;

n is zero or 1; and

p is zero, 1, or 2.

3. The compound according to claim 1 , N-oxide, or a salt thereof, wherein:

each R 1 is independently H, —CH 3 , —CH 2 CH 3 , —CH(CH 3 ) 2 , —OCH 3 , or —S(O) 2 CH 3 ;

G is:

each R 2 is independently —CN, —CH 3 , —OCH 3 , or pyrimidinyl;

A is:

(i) —CH 2 R 3 , —CH 2 CH 2 NHR 3 , —C(O)NHR 3 , —NHR 3 , —NHCH 2 CH 2 R 3 , or —OR 3 ; or

(ii) R 3 ;

R 3 is azetidinyl, cyclobutyl, cyclohexyl, piperazinyl, piperidinyl, or pyrrolidinyl, each substituted with zero to 1 R 3a ;

R 3a is —CH 3 , —CH 2 CH 2 CH 3 , —CH(CH 3 ) 2 , —CH 2 CN, —CH 2 CH 2 CN, —CH 2 CH 2 CF 3 , —CH 2 C(CH 3 ) 2 OH, —CH 2 C(O)NH 2 , —CH 2 C(O)N(CH 3 ) 2 , —C(O)CH 2 N(CH 3 ) 2 , —C(O)CH 2 CH 2 N(CH 3 ) 2 , —C(O)CH 2 CH 2 CH 2 N(CH 3 ) 2 , —NH(CH 3 ), —N(CH 3 ) 2 , —NH(CH(CH 3 ) 2 ), —CH 2 CH 2 S(O) 2 CH 3 , —CH 2 CH 2 NHS(O) 2 CH 3 , —NHCH 2 C(O)N(CH 3 ) 2 , —NH(CH 2 CH 2 S(O) 2 CH 3 ), —NH(cyclopropyl), —NH(oxetanyl), —N(CH 3 )(oxetanyl), —CH 2 (methyltriazolyl), morpholinyl, oxetanyl, pyrrolidinyl, fluoropyrrolidinyl, hydroxy-methylpyrrolidinyl, tetrahydropyranyl, 1-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.3]heptanyl, 2-oxa-6-azaspiro[3.4]octanyl, 6-oxa-2-azaspiro[3.4]octanyl, 2,2-dioxo-2-thia-6-azaspiro[3.3]heptanyl, 2-oxa-7-azaspiro[4.4]nonanyl, or azetidinyl substituted with zero to 2 substituents independently selected from F, —OH, —CH 3 , —CH 2 OH, —OCH 3 , —OCH 2 CH 3 , and —OCH(CH 3 ) 2 ;

n is zero or 1; and

p is zero, 1, or 2.

4. The compound according to claim 1 , N-oxide, or a salt thereof, wherein X is N.

5. The compound according to claim 1 or a salt thereof, wherein

X is CH or N;

G is:

R 2 is —CH 3 or —OCH 3 ;

A is: —OR 3 or R 3 ;

R 3 is cyclobutyl, cyclohexyl, piperazinyl, piperidinyl, or pyrrolidinyl, each substituted with zero to 1 R 3a ;

n is zero or 1; and

p is zero or 1.

6. The compound according to claim 1 or a salt thereof, wherein

X is N;

G is:

R 2 is —CH 3 or —OCH 3 .

7. The compound according to claim 1 , N-oxide, or a salt thereof, wherein said compound is:

8-methyl-6-(5-methyl-3-(piperidin-4-yl)-1H-indol-6-yl)-[1,2,4]triazolo[1,5-a]pyridine (1);

8-methyl-6-(4-methyl-3-(piperidin-4-yl)-1H-indol-6-yl)-[1,2,4]triazolo[1,5-a]pyridine (2);

8. A pharmaceutical composition comprising a compound according to claim 1 or a pharmaceutically-acceptable salt thereof; and a pharmaceutically acceptable carrier.

A method of treating an autoimmune disease or a chronic inflammatory disease, comprising administering to a mammalian patent a compound according to claim 1 or a pharmaceutically acceptable salt thereof, wherein said autoimmune disease or chronic inflammatory disease is selected from systemic lupus erythematosus (SLE), rheumatoid arthritis, multiple sclerosis (MS), and Sjögren's syndrome.
