Abstract

Provided herein are compositions and methods for the assembly of a bioluminescent complex from two or more non-luminescent (e.g., substantially non-luminescent) peptide and/or polypeptide units. In particular, bioluminescent activity is conferred upon a non-luminescent polypeptide via structural complementation with another, complementary non-luminescent peptide.

Claims (17)

1. A system comprising:

(a) a polypeptide comprising an amino acid sequence having less than 100% and greater than 40% sequence identity with SEQ ID NO: 440;

(b) a peptide comprising an amino acid sequence having less than 100% and greater than 40% sequence identity with SEQ ID NO: 2;

(c) a bioluminescent substrate having an emission spectrum; and

(d) a fluorescent dye or protein having an excitation spectrum.

2. The system of claim 1 , wherein the fluorescent dye or protein is linked to the peptide.

3. The system of claim 1 , wherein the fluorescent dye or protein is linked to the polypeptide.

The system of claim 1 , wherein a first fusion protein comprises the peptide and a first interaction element, wherein a second fusion protein comprises the polypeptide and a second interaction element, wherein the first and second interaction elements are configured to form and interaction complex, and wherein formation of the interaction complex facilitates formation of the bioluminescent complex.

5. The system of claim 1 , wherein the peptide comprises an amino acid sequence with at least 70% sequence identity to SEQ ID NO: 2.

7. The system of claim 6 , wherein the peptide comprises an amino acid sequence of SEQ ID NO: 390.

8. The system of claim 6 , wherein the peptide comprises an amino acid sequence of SEQ ID NO: 2271.

9. The system of claim 1 , wherein the polypeptide comprises an amino acid sequence with at least 70% sequence identity to SEQ ID NO: 440.

The system of claim 1 , wherein the polypeptide comprises an amino acid sequence selected from the polypeptide sequences of SEQ ID NOS: 442, 444, 446, 448, 450, 452, 454, 456, 458, 460, 462, 464, 466, 12. The system of claim 11 , wherein the peptide comprises an amino acid sequence with at least 80% sequence identity to SEQ ID NO: 2, and wherein the polypeptide comprises an amino acid sequence with at least 80% sequence identity to SEQ ID NO: 440.

13. The system of claim 12 , wherein the peptide comprises an amino acid sequence with at least 90% sequence identity to SEQ ID NO: 2, and wherein the polypeptide comprises an amino acid sequence with at least 80% sequence identity to SEQ ID NO: 440.
