Abstract

GnRH receptor antagonists are disclosed which have utility in the treatment of a variety of sex-hormone related conditions in both men and women. The compounds of this invention have the structure: wherein A, R 1 , R 2 , R 3a , R 3b , R 4 , R 5 , R 6 , and n are as defined herein, including stereoisomers, prodrugs and pharmaceutically acceptable salts thereof. Also disclosed are compositions containing a compound of this invention in combination with a pharmaceutically acceptable carrier, as well as methods relating to the use thereof for antagonizing gonadotropin-releasing hormone in a subject in need thereof.

Claims (51)

1. A compound having the following structure:

or a stereoisomer, or pharmaceutically acceptable salt thereof,

wherein:

A is N;

n is 2, 3 or 4;

R 1 and R 2 are the same or different and independently hydrogen, alkyl, substituted alkyl, aryl, substituted aryl, arylalkyl, substituted arylalkyl, heterocycle, substituted heterocycle, heterocyclealkyl, substituted heterocyclealkyl, —C(R 8 )(═NR 9 ) or —C(NR 10 R 11 )(═NR 9 );

or R 1 and R 2 taken together with the nitrogen atom to which they are attached form a heterocycle or a substituted heterocycle;

R 3a and R 3b are the same or different and, at each occurrence, independently hydrogen, alkyl, substituted alkyl, alkoxy, alkylthio, alkylamino, aryl, substituted aryl, arylalkyl, substituted arylalkyl, heterocycle, substituted heterocycle, heterocyclealkyl, substituted heterocyclealkyl, —COOR 12 or —CONR 10 R 11 ;

or R 3a and R 3b taken together with the carbon atom to which they are attached form a homocycle, substituted homocycle, heterocycle or substituted heterocycle;

or R 3a and the carbon to which it is attached taken together with R 1 and the nitrogen to which it is attached form a heterocycle or substituted heterocycle;

R 4 is arylalkyl, substituted arylalkyl, heteroarylalkyl or substituted heteroarylalkyl;

R 5 is hydrogen, alkyl, or substituted alkyl;

R 6 is aryl, substituted aryl, heteroaryl, or substituted heteroaryl;

R 7 is hydrogen, alkyl, or substituted alkyl;

R 8 is independently hydrogen, alkyl, substituted alkyl, aryl, substituted aryl, arylalkyl, substituted arylalkyl, heterocycle, substituted heterocycle, heterocyclealkyl or substituted heterocyclealkyl;

R 9 is independently hydrogen, alkyl, substituted alkyl, aryl, substituted aryl, arylalkyl, substituted arylalkyl, heterocycle, substituted heterocycle, heterocyclealkyl or substituted heterocyclealkyl;

R 10 and R 11 are the same or different independently hydrogen, alkyl, substituted alkyl, aryl, substituted aryl, arylalkyl, substituted arylalkyl, heterocycle, substituted heterocycle, heterocyclealkyl or substituted heterocyclealkyl; and

R 12 is hydrogen, alkyl, or substituted alkyl.

2. The compound of claim 1 wherein R 1 is hydrogen, alkyl, substituted alkyl, arylalkyl, substituted arylalkyl, heterocyclealkyl or substituted heterocyclealkyl.

3. The compound of claim 1 wherein R 4 is arylalkyl, substituted arylalkyl, or heteroarylalkyl.

4. The compound of claim 1 wherein R 6 is aryl, substituted aryl, or heteroaryl.

5. A pharmaceutical composition comprising a compound of claim 1 and a pharmaceutically acceptable carrier or diluent.

6. A compound according to claim 1 , wherein:

n is 2;

R 3a is hydrogen at each occurrence;

R 3b is hydrogen at one occurrence and aryl or substituted aryl at the other occurrence; and

R 1 and R 2 are independently hydrogen, alkyl or substituted alkyl.

7. A compound according to claim 6 , wherein:

R 3b is hydrogen at one occurrence and phenyl at the other occurrence; and

R 1 and R 2 are each hydrogen.

8. A compound according to claim 6 , wherein R 5 is hydrogen or alkyl.

9. A compound according to claim 8 , wherein R 5 is methyl.

10. A compound according to claim 8 , wherein R 6 is aryl or substituted aryl.

11. A compound according to claim 10 , wherein R 6 is phenyl or substituted phenyl.

12. A compound according to claim 11 , wherein R 6 is 2-fluorophenyl, 2-chlorophenyl, 2-fluoro-3-methoxyphenyl, 2-chloro-3-methoxyphenyl or 3-methoxyphenyl.

13. A compound according to claim 10 , wherein R 4 is arylalkyl or substituted arylalkyl.

14. A compound according to claim 13 , wherein R 4 is benzyl or substituted benzyl.

15. A compound according to claim 14 , wherein R 4 is 2-fluorobenzyl, 2-chlorobenzyl, 2-trifluoromethylbenzyl, 2-methylsulfonylbenzyl, 2,6-difluorobenzyl, 2,6-dichlorobenzyl, 2-fluoro-6-chlorobenzyl, 2-fluoro-6-trifluoromethylbenzyl, or 2-fluoro-6-methylsulfonylbenzyl.

16. A compound according to claim 7 , wherein:

R 4 is 2-fluorobenzyl, 2-chlorobenzyl, 2-trifluoromethylbenzyl, 2-methylsulfonylbenzyl, 2,6-difluorobenzyl, 2,6-dichlorobenzyl, 2-fluoro-6-chlorobenzyl, 2-fluoro-6-trifluoromethylbenzyl, or 2-fluoro-6-methylsulfonylbenzyl;

R 5 is methyl; and

R 6 is 2-fluorophenyl, 2-chlorophenyl, 2-fluoro-3-methoxyphenyl, 2-chloro-3-methoxyphenyl or 3-methoxyphenyl.

17. A pharmaceutical composition comprising a compound according to claim 6 and a pharmaceutically acceptable carrier or diluent.

18. A pharmaceutical composition according to claim 17 , wherein:

R 3b is hydrogen at one occurrence and phenyl at the other occurrence; and

R 1 and R 2 are each hydrogen.

19. A pharmaceutical composition comprising a compound according to claim 13 and a pharmaceutically acceptable carrier or diluent.

20. A pharmaceutical composition according to claim 19 , wherein:

R 4 is 2-fluorobenzyl, 2-chlorobenzyl, 2-trifluoromethylbenzyl, 2-methylsulfonylbenzyl, 2,6-difluorobenzyl, 2,6-dichlorobenzyl, 2-fluoro-6-chlorobenzyl, 2-fluoro-6-trifluoromethylbenzyl, or 2-fluoro-6-methylsulfonylbenzyl;

R 5 is methyl; and

R 6 is 2-fluorophenyl, 2-chlorophenyl, 2-fluoro-3-methoxyphenyl, 2-chloro-3-methoxyphenyl or 3-methoxyphenyl.
