Abstract

A method of determining information about cell viability and other characteristics relating to cell membrane permeability is disclosed. The method involves determining the effect of a cell on current flow and relating that effect to a known standard which standard may be a known healthy cell and thereby deducing the viability of the cell being tested. The cells being tested can be subjected to different environmental conditions such as surrounding chemicals, temperature, pH and pressure to determine the effects of such conditions on cell viability and/or cell permeability. The cell being tested can be in a cell suspension, grown on s ubstarte, in tissue in vitro or in tissue in vivo. The method provides substantially instantaneous results and need not include the use of dyes or other markers.

Claims (29)

1. A method of determining cell viability, comprising the steps of:

(a) placing a first cell in an electrically conductive medium wherein said first cell blocks an opening in a material which material blocks current flow;

(b) applying a voltage across said first cell;

(c) detecting electrical current through said first cell;

(d) comparing the detected electrical current across said first cell to a standard comprising a previous reading on a second cell that has a known viability in order to determine the viability said first cell.

2. The method of claim 1 , further comprising:

(e) repeating steps (a)-(d) a plurality of times with a different first cell each time.

3. The method of claim 1 , wherein said first cell is held in a opening in a material which blocks current flow.

4. The method of claim 1 , wherein a plurality of first cells are placed in the electrically conductive medium in the opening of the material which blocks current flow.

5. A method of determining cell viability, comprising the steps of:

(a) placing a first cell in an electrically conductive medium wherein said first cell blocks an opening in a material which material blocks current flow and said first cell is held in an opening in the material;

(b) applying a voltage across said first cell;

(c) detecting electrical current through said first cell;

(d) comparing the detected electrical current across said first cell to a standard comprising a previous reading on a second cell that has a known viability in order to determine the viability of said first cell; and

(e) repeating steps (a)-(d) a plurality of times with a different first cell each time to determine viability of each first cell.

6. The method of claim 1 , wherein the voltage applied across said first cell is a direct current (DC) pulse voltage.

7. The method of claim 5 , wherein the voltage applied across said first cell is a direct current (DC) pulse voltage.

8. The method of claim 1 , wherein the previous measurement is obtained from a second cell substantially identical to said first cell of steps (a)-(c).

9. The method of claim 8 , wherein the second cell has been subjected to a condition affecting cell viability.

10. The method of claim 9 , wherein the condition is temperature, pressure, pH, radiation or exposure to a substance.

11. The method of claim 10 , wherein the condition is radiation selected from the group consisting of UV, visible and infrared.

12. The method of claim 1 , wherein the second cell is a normal cell.

13. The method of claim 1 , wherein the second cell is a diseased cell.

14. The method of claim 9 , wherein the condition is exposure to a substance selected from the group consisting of a protein, a nucleotide sequence, a bacteria, a virus, an infectious prion, a fungus and a polysaccharide.

15. The method of claim 5 , wherein the previous measurement is obtained from a second cell substantially identical to said first cell of steps (a)-(c).

16. The method of claim 15 , wherein the second cell has been subjected to a condition affecting cell viability.

17. The method of claim 7 , wherein the condition is temperature, pressure, pH, radiation or exposure to a substance.

18. The method of claim 17 , wherein the condition is radiation selected from the group consisting of UV, visible and infrared.

19. The method of claim 16 , wherein the condition is exposure to a substance selected from the group consisting of a protein, a nucleotide sequence, a bacteria, a virus, an infectious prion, a fungus and a polysaccharide.
