Abstract

Compounds having the formula (I), and pharmaceutically acceptable salts or prodrugs thereof, are useful as kinase inhibitors, wherein Y is —C(═O)NR 1 —, —NR 1 C(═O)—, —NR 1 C(═O)NR 1 —, —NR 1 SO 2 —, —SO 2 NR 1 —, —C(═O)—, —OC(═O)—, or —CO 2 —; B is alkyl, substituted alkyl, alkenyl, substituted alkenyl, hydroxy, alkoxy, aryl, cycloalkyl, heteroaryl, and heterocyclo; or when Y is —C(═O)NR 1 —, B also may be selected from —C(═O)R 8 , —C(═O)NR 8 R 9 , and —CO 2 R 8 ; R 1 is hydrogen, C 1-4 alkyl, or substituted C 1-4 alkyl; R 2 is hydrogen or C 1-4 alkyl; R 3 is hydrogen, methyl, perfluoromethyl, hydroxy, methoxy, halogen, cyano, NH 2 , or NH(CH 3 ); and R 4 , R 5 , R 6 , R 7 , R 8 , and R 9 are as defined in the specification.

Claims (61)

1. A compound having the formula:

or a pharmaceutically-acceptable salt or prodrug, thereof, wherein:

Y is —C(═O)NR 1 —, —NR 1 C(═O)—, —NR 1 C(═O)NR 1 —, —NR 1 SO 2 —, —SO 2 NR 1 —, —C(═O)—, —OC(═O)—, or —CO 2 —, provided, however, that when R 6 is hydrogen, Y is not —NR 1 C(═O)—;

B is selected from alkyl, substituted alkyl, alkenyl, substituted alkenyl, hydroxy, alkoxy, aryl, cycloalkyl, heteroaryl, and heterocyclo; or when Y is —C(═O)NR 1 —, B also may be selected from —C(═O)R 8 , —C(═O)NR 8 R 9 , and —CO 2 R 8 ,

R 1 is hydrogen, C 1-4 alkyl, or substituted C 1-4 alkyl;

R 2 is hydrogen or C 1-4 alkyl;

R 3 is hydrogen, methyl, perfluoromethyl, hydroxy, methoxy, halogen, cyano, NH 2 , or NH(CH 3 );

R 4 is hydrogen, alkyl, substituted alkyl, alkenyl, substituted alkenyl, aryl, cycloalkyl, heteroaryl, or heterocyclo;

R 5 is selected from hydrogen, halogen, cyano, alkyl, substituted alkyl, —C(═O)R 11 , —CO 2 R 11 , —S(═O)R 12 , —SO 2 R 12 , —SO 3 R 12 , —OR 11 , —SR 11 , —NR 11 R 12 , —C(═O)NR 11 R 12 , —NR 11 C(═O)R 12 , NR 11 CO 2 R 12 , —NR 11 SO 2 R 12 , —SO 2 NR 11 R 12 , cycloalkyl, aryl, heterocyclo and heteroaryl;

R 6 is selected from halogen, cyano, nitro, A 1 -R 13 , -A 1 -O-A 2 -R 13 , -A 1 -S-A 2 -R 13 , -A 1 -C(═O)-A 2 -R 13 , -A 1 -OC(═O)-A 2 -R 13 , -A 1 -S(═O)-A 2 -R 13 , -A 1 -SO 2 -A 2 -R 13 , -A 1 -CO 2 -A 2 -R 13 , -A 1 -NR 13 -A 2 -R 14 , -A 1 -NR 15 C(═O)-A 2 -R 13 , -A 1 -NR 15 C(═O)NR 16 -A 2 -R 13 , -A 1 -NR 15 CO 2 -A 2 -R 13 , -A 1 -NR 15 SO 2 -A 2 -R 13 , -A 1 -NR 15 SO 2 NR 16 -A 2 -R 13 , -A 1 -SO 2 NR 15 -A 2 -R 13 , and -A 1 -C(═O)NR 15 -A 2 -R 13 ;

A 1 is —(CR 17 R 18 ) r —;

A 2 is —(CR 19 R 20 ) s —;

R 8 and R 9 are selected from hydrogen, alkyl, substituted alkyl, aryl, cycloalkyl, heteroaryl, and heterocyclo, or may form together with the N atom to which these are attached, a heteroaryl or heterocyclo ring;

R 11 , R 12 , R 13 , and R 14 are selected from hydrogen, alkyl, substituted alkyl, alkenyl, substituted alkenyl, aryl, cycloalkyl, heteroaryl, and heterocyclo, or may form together with the N atom to which these are attached, a heteroaryl or heterocyclo ring, provided that when R 12 or R 13 is attached to a sulfonyl group (as in —S(═O)R 12 , —SO 2 R 12 , —SO 3 R 12 , -A 1 -S(═O)R 13 , -A 1 -SO 2 R 13 , and -A 1 -SO 3 R 13 ), R 12 and R 13 are not hydrogen;

R 15 and R 16 are selected from hydrogen, C 1-4 alkyl, hydroxy, hydroxyC 1-4 alkyl, and aminoC 1-4 alkyl;

R 17 , R 18 , R 19 , and R 20 are selected from hydrogen, C 1-4 alkyl, halogen, cyano, hydroxy, hydroxyC 1-4 alkyl, haloC 1-4 alkyl, C 1-4 haloalkoxy, amino, C 1-4 alkylamino, and aminoC 1-4 alkyl;

m is 0, 1 or 2; and

r and s are selected from 0, 1, 2, 3, and 4.

2. A compound according to claim 1 , or a pharmaceutically-acceptable salt, or prodrug thereof, having the formula (Ia),

wherein

R 5a and R 5b are independently selected from hydrogen, C 1-4 alkyl, substituted C 1-4 alkyl, halogen, trifluoromethoxy, trifluoromethyl, hydroxy, C 1-4 alkoxy, cyano, NH 2 , NH(C 1-4 alkyl), N(C 1-4 alkyl) 2 , C 3-7 cycloalkyl, five to six membered heteroaryl, four to six membered heterocyclo, phenyl, benzyl, phenyloxy, and benzyloxy.

3. A compound according to claim 2 , or a pharmaceutically-acceptable salt or prodrug thereof, in which:

Y is —C(═O)NH—, —NHC(═O)—, —NHC(═O)NH—, —NHSO 2 —, or —SO 2 NH—;

B is a hydroxy, alkoxy, cycloalkyl, heteroaryl, heterocyclo, arylalkyl, cycloalkylalkyl, heteroarylalkyl, or heterocycloalkyl;

R 4 is alkyl, substituted alkyl, aryl or heteroaryl;

R 6 is selected from hydrogen, alkyl, substituted alkyl, halogen, cyano, -A 1 -O-A 2 -R 13 , -A 1 -NR 13 R 14 , —NR 13 -A 2 R 14 , aryl, cycloalkyl, heteroaryl, and heterocyclo, provided R 6 is not hydrogen when B is —NHC(═O)—;

A 1 is —(CH 2 ) r —;

A 2 is —(CH 2 ) s —; and

R 13 and R 14 are selected from hydrogen, alkyl, substituted alkyl, cyano, hydroxy, alkoxy, aryl, cycloalkyl, heteroaryl, and heterocyclo, or may form together with the N atom to which these are attached, a heteroaryl or heterocyclo ring.

4. A compound according to claim 1 , or a pharmaceutically acceptable salt or prodrug thereof, in which R 2 is hydrogen.

5. A compound according to claim 1 , or a pharmaceutically acceptable salt or prodrug thereof, in which R 3 is hydrogen.

6. A compound according to claim 1 , or a pharmaceutically-acceptable salt or prodrug thereof, wherein R 4 is aryl or heteroaryl.

7. A compound according to claim 2 , or a pharmaceutically-acceptable salt or prodrug thereof, wherein R 5a is halogen or lower alkyl and R 5b is hydrogen.

A compound according to claim 1 , or a pharmaceutically-acceptable salt or prodrug thereof, wherein R 6 is selected from —O-A 2 -R 13 , —NR 13 R 14 , —NR 13 -A 2 R 14 , aryl, cycloalkyl, heteroaryl, and heterocyclo; and R 13 and R 14 are selected from aryl, cycloalkyl, heteroaryl, and heterocyclo, or alternatively, R 13 and R 14 taken together form a heterocyclo or heteroaryl ring.

9. A compound according to claim 1 , or a pharmaceutically-acceptable salt or prodrug thereof, wherein

R 6 is alkyl, —O—R 13 , —NR 13 R 14 , morpholinyl, piperidinyl, pyrrolidinyl, piperazinyl, or diazepinyl; and

R 13 and R 14 are selected from hydrogen, alkyl, hydroxy, hydroxyalkyl, haloalkyl, cyano, cyanoalkyl, morpholinyl, morpholinylalkyl, C 3-7 cycloalkyl, C 3-7 cycloalkylalkyl, pyrrolidinyl, pyrrolidinylalkyl, piperidinyl, piperidinylalkyl, piperazinyl, piperazinylalkyl, wherein each R 13 and R 14 group is optionally substituted with one to two groups selected from alkyl, halogen, cyano, hydroxy, C 1-4 alkoxy, amino, lower aminoalkyl, lower alkylamino, and C 1-4 alkyl substituted with one to two hydroxy, halogen, cyano, —O(C 1-4 alkyl), —O(C 2-4 alkenyl), amino, C 1-4 alkylamino, nitro, trifluoromethyl, trifluoromethoxy, —S(C 1-4 alkyl), —SO 2 C 1-4 alkyl, —CO 2 H, —CO 2 (C 1-4 alkyl), —C(═O)H, and/or —C(═O)(C 1-4 alkyl).

10. A compound according to claim 1 , or a pharmaceutically-acceptable salt or prodrug, thereof, in which Y is —C(═O)NH—.

11. A compound according to claim 1 , or a pharmaceutically-acceptable salt or prodrug thereof, wherein

B is selected from —(CH 2 ) n -cycloalkyl, —(CH 2 ) n -(heteroaryl), —(CH 2 ) n -heterocyclo, and —(CH 2 ) n -phenyl, wherein each B is optionally substituted with one to three R 7 , and in the case of a non-aromatic ring, optionally in addition to one to two R 7 , B may be substituted with a keto (═O) group;

R 7 is selected from C 1-6 alkyl, halogen, trifluoromethoxy, trifluoromethyl, cyano, hydroxy, —OC 1-4 alkyl, —SC 1-4 alkyl, —CO 2 H, —(CO 2 )(C 1-4 alkyl), —C(═O)(C 1-4 alkyl), nitro, phenyl, benzyl, phenyloxy, benzyloxy, NH 2 , NH(C 1-4 alkyl), N(C 1-4 alkyl) 2 , C 3-7 cycloalkyl, five or six membered heteroaryl or heterocycle, and C 1-4 alkyl substituted with one to two hydroxy, halogen, cyano, —O(C 1-4 alkyl), —O(C 2-4 alkenyl), amino, C 1-4 alkylamino, nitro, trifluoromethyl, trifluoromethoxy, —S(C 1-4 alkyl), —SO 2 C 1-4 alkyl, —CO 2 H, —CO 2 (C 1-4 alkyl), —C(═O)H, and/or —C(═O)(C 1-4 alkyl); and

n is 0, 1, 2 or 3.

12. A compound of formula (Ib),

or a pharmaceutically-acceptable salt or prodrug thereof, wherein

B is selected from hydroxy, alkoxy, —C(═O)R 8 , —C(═O)NR 8 R 9 , —CO 2 R 8 , —(CH 2 ) n —C 3-7 cycloalkyl, —(CH 2 ) n -heteroaryl, —(CH 2 ) n -heterocyclo, and —(CH 2 ) n -phenyl, wherein each cyclic group B is optionally substituted with one to three R 7 ;

R 5a and R 5b are independently hydrogen, C 1-4 alkyl, substituted C 1-4 alkyl, halogen, C 1-4 haloalkoxy, C 1-4 haloalkyl, hydroxy, —O(C 1-4 alkyl), or cyano;

R 6 is alkyl, —O—(CH 2 ) r —R 13 , —NR 13 R 14 , aryl, cycloalkyl, heterocyclo, or heteroaryl, wherein when R 6 is alkyl, aryl, cycloalkyl, heterocyclo, or heteroaryl, R 6 is optionally substituted with one to three groups selected from alkyl, substituted alkyl, halogen, cyano, nitro, hydroxy, alkoxy, haloalkoxy, amino, alkylamino, phenyl, benzyl, phenyloxy, and benzyloxy;

R 7 and R 21 are independently selected from C 1-6 alkyl, substituted C 1-4 alkyl, halogen, haloalkyl, haloalkoxy, cyano, nitro, hydroxy, —O(C 1-4 alkyl), phenyl, benzyl, phenyloxy, benzyloxy, NH 2 , NH(C 1-4 alkyl), N(C 1-4 alkyl) 2 , C 3-7 cycloalkyl, and five or six membered heteroaryl or heterocycle;

R 8 and R 9 are selected from hydrogen, alkyl, substituted alkyl, aryl, cycloalkyl, heteroaryl, and heterocyclo, or may form together with the N atom to which these are attached, a heteroaryl or heterocyclo ring;

R 13 and R 14 are selected from hydrogen, alkyl, substituted alkyl, cyano, aryl, cycloalkyl, heteroaryl, or heterocyclo, or together form a heterocyclo or heteroaryl ring;

r is 0, 1, 2, 3 or 4; and

n is 0, 1, 2, 3, or 4.

13. A compound according to claim 12 , or a pharmaceutically acceptable salt or prodrug thereof, in which:

R 6 is (i) —OR 13 , or —NR 13 R 14 ; or (ii) alkyl, pyrrolidinyl, morpholinyl, piperidinyl, piperazinyl, or diazepinyl optionally substituted with one to three groups selected from alkyl, hydroxyalkyl, cyanoalkyl, haloalkyl, haloalkoxy, halogen, cyano, hydroxy, alkoxy, amino, aminoalkyl, and alkylamino; and

R 13 and R 14 are selected from hydrogen, alkyl, hydroxyalkyl, cyano, cyanoalkyl, morpholinylalkyl, C 3-7 cycloalkyl, C 3-7 cycloalkylalkyl, pyrrolidinyl, pyrrolidinylalkyl, piperidinyl, piperidinylalkyl, wherein each R 13 and R 14 group in turn is optionally substituted with one to two groups selected from C 1-6 alkyl, substituted C 1-6 alkyl, halogen, cyano, hydroxy, —O(C 1-4 alkyl), amino, and C 1-6 alkylamino.

14. A compound according to claim 12 , or a pharmaceutically acceptable salt or prodrug thereof, in which B is phenyl optionally substituted with one to two R 7 ; cycloalkyl optionally substituted with keto and/or one to two R 7 ; or B is selected from one of:

R 7 is hydrogen, C 1-4 alkyl, trifluoromethyl, trifluoromethoxy, halogen, cyano, amino, —(C 1-4 alkyl)amino, hydroxy, —O(C 1-4 alkyl), phenyl, benzyl, phenyloxy, or benzyloxy, or two R 7 groups attached to adjacent carbon atoms or an adjacent carbon and nitrogen atom may join to form a fused heterocyclo or carbocyclic ring, said fused ring in turn being optionally substituted with one to two of C 1-4 alkyl, trifluoromethyl, trifluoromethoxy, halogen, cyano, amino, —(C 1-4 alkyl)amino, hydroxy, —O(C 1-4 alkyl), phenyl, benzyl, phenyloxy, and benzyloxy; and

t is 2 or 3.

15. A pharmaceutical composition comprising at least one compound according to claim 1 and a pharmaceutically-acceptable carrier or diluent.

16. A pharmaceutical composition comprising at least one compound according to claim 12 and a pharmaceutically-acceptable carrier or diluent.

17. A method of treating from rheumatoid arthritis, psoriatic arthritis, traumatic arthritis, rubella arthritis, gouty arthritis and/or osteoarthritis in a patient comprising administering to the patient a pharmaceutical composition according to claim 15 .
