Abstract

A novel class of NIMA interacting proteins (PIN), exemplified by Pin1, is provided. Pin1 induces a G2 arrest and delays NIMA-induced mitosis when overexpressed, and triggers mitotic arrest and DNA fragmentation when depleted. Methods of identifying other Pin proteins and Pin-interacting proteins and identifying compositions which affect Pin activity or expression are also provided.

Claims (9)

1. An isolated nucleic acid molecule fragment of a encoding Pin 1 polypeptide consisting of amino acids 5–43 of SEQ ID NO:2 or a functional fragment thereof that binds to NIMA, or a complement of the encoding nucleic acid molecule.

2. The isolated nucleic acid molecule of claim 1 encoding amino acids 5–43 of SEQ ID NO:2 or the complement thereof.

3. The isolated nucleic acid molecule of claim 1 consisting of nucleotides 13–129 of SEQ ID NO:1.

4. An isolated nucleic acid molecule fragment of a encoding Pin 1 polypeptide consisting of amino acids 59–163 of SEQ ID NO:2 or a functional fragment thereof having peptidyl prolyl isomerase activity, or a complement of the nucleic acid molecule.

5. The isolated nucleic acid molecule of claim 4 encoding amino acids 59–163 of SEQ ID NO:2 or the complement thereof.

6. An isolated nucleic acid molecule encoding a fragment of a Pin 1 polypeptide, which Pin 1 polypeptide consists of amino acid residue 1–163 of SEQ ID NO:2, wherein the encoded fragment comprises amino acid residues 5–43 of SEQ ID NO:2 and binds to NIMA.

7. An isolated nucleic acid molecule encoding a fragment of a Pin 1 polypeptide, which Pin 1 polypeptide consists of amino acid residue 1–163 of SEQ ID NO:2, wherein the encoded fragment comprises amino acid residues 59–163 of SEQ ID NO:2 and has peptidyl prolyl isomerase activity.

8. A vector comprising the isolated nucleic acid molecule of claim 1 , 4 , 6 , or 7 .

9. An isolated host cell comprising the vector of claim 8 .
