Abstract

CRF peptide analogs that bind to CRFR1 with an affinity far greater than they bind to CRFR2. These analogs exhibit CRF agonist activity. One exemplary analog that may be made by solid-phase synthesis is (cyclo 31-34)[Ac-Pro 4 , D-Phe 12 , Nle 21,38 , Glu 31 , Lys 34 ]-r/hCRF(4-41).

Claims (14)

1. A 38-residue or 39-residue CRF cyclic peptide, or a nontoxic salt thereof, which binds to CRFR1 with an affinity substantially greater than it binds to CRFR2, which peptide has the following formula:

wherein the sidechains of Glu and Lys indicated are covalently linked; Y 1 is an acyl group having not more than 15 carbon atoms or is radioiodinated tyrosine; R 14 is CML or Leu; R 15 is CML or Leu; R 17 is Glu or CML; R 18 is Val or CML; R 19 is CML or Leu; R 27 is CML or Leu; R 32 is His or D-His; R 33 is Aib, D-Ala, D-Ser or Ser; R 36 is Lys or CML; R 37 is CML or Leu; R 40 is Ile or CML; and R 41 is Ile or CML; provided that D-β-(2-napthyl)alanine(D-2Nal) or D-Leu may be substituted for D-Phe.

2. A CRF agonist peptide, or a nontoxic salt thereof, which binds to CRFR1 with an affinity substantially greater than it binds to CRFR2, which peptide has the following formula:

wherein Y 1 is an acyl group having not more than 7 carbon atoms or is radioiodinated tyrosine, and wherein a cyclizing bond may exist between the side chains of Glu and Lys as indicated.

3. A 38-residue or 39-residue CRFR1 ligand cyclic peptide which binds to CRFR1 with an affinity substantially greater than it binds to CRFR2, which peptide has the following formula, or a nontoxic salt thereof:

wherein the side chains of Glu and R 34 are covalently linked as indicated; Y 1 is an acyl group having not more than 7 carbon atoms or is radioiodinated tyrosine; R 6 is Ile, Met or Nle; R 8 is Leu or Ile; R 11 is Thr or Ser; R 14 is CML or Leu; R 15 is Leu or CML; R 18 is Val, CML, Nle or Met; R 20 is Glu or D-Glu; R 22 is Ala or Thr; R 23 is Arg or Lys; R 25 is Asp or Glu; R 27 is Leu or CML; R 29 is Gln or Glu; R 32 is His, Aib, Ala, Gly, Leu, Gln or Glu; R 33 is Aib or an L- or D-isomer of Ser, Asn, Leu, Ala, CML or Ile; R 34 is Lys or Orn; R 36 is Lys or Leu; R 37 is CML or Leu; R 39 is Glu or Asp; R 40 is Ile, CML or Glu; and R 41 is Ala, Aib or Ile; provided that D-β-(2-napthyl)alanine(D-2Nal) or D-Leu may be substituted for D-Phe.

4. A CRF cyclic peptide according to claim 3 having the formula:

wherein Y 1 is an acyl group having not more than 7 carbon atoms; R 20 is Glu or D-Glu; R 22 is Ala or Thr; R 23 is Arg or Lys; R 29 is Gln or Glu; R 32 is His, Aib or Ala; R 36 is Lys or Leu; R 37 is Leu or CML; R 39 is Glu or Asp; R 40 is Ile, CML or Glu; and R 41 is Ile, Aib or Ala; wherein the remaining variabies are as defined in claim 3 .

5. A peptide according to claim 3 wherein R 18 is Val, R 22 is Ala, R 23 is Arg, R 25 is Glu, R 39 is Glu, and R 41 is Ile.

6. A peptide according to claim 3 having the following formula, or a nontoxic salt thereof:

wherein Y 1 is an acyl group having not more than 7 carbon atoms; R 22 is Ala or Thr; R 23 is Arg or Lys; R 27 is Leu or CML; R 32 is His; R 33 is Ser or Aib; and R 40 is Ile or CML.
