Abstract

Use of antagonists to IL-31 are used to treat inflammation and pain by inhibiting, preventing, reducing, minimizing, limiting or minimizing stimulation in neuronal tissues. Such antagonists include antibodies and fragments, derivative, or variants thereof. Symptoms such as pain, tingle, sensitization, tickle associated with neuropathies are ameliorated.

Claims (66)

1. A method of antagonizing signal transduction in dorsal root ganglion cells in vitro, comprising admixing an antibody with the dorsal root ganglion cells wherein the antibody binds Interleukin 31 (IL-31) and whereby the signal transduction is inhibited.

2. The method of claim 1 , wherein the antibody is a monoclonal antibody.

3. The method of claim 1 , wherein the antibody is humanized.

4. The method of claim 1 , wherein the antibody is an antibody fragment.

5. The method of claim 4 , wherein the antibody fragment is selected from the group consisting of:

a) a single-chain antibody;

b) a Fab fragment;

c) a ″Fab′ fragment; and

d) a F(ab′)2 fragment.

6. The method of claim 1 , wherein the antibody is a chimeric antibody.

7. The method of claim 1 , wherein the antibody is produced by the hybridoma selected from the group consisting of:

a) the hybridoma having the ATCC Patent Deposit Designation PTA-6815;

b) the hybridoma having the ATCC Patent Deposit Designation PTA-6816;

c) the hybridoma having the ATCC Patent Deposit Designation PTA-6829;

d) the hybridoma having the ATCC Patent Deposit Designation PTA-6830;

e) the hybridoma having the ATCC Patent Deposit Designation PTA-6831;

f) the hybridoma having the ATCC Patent Deposit Designation PTA-6871;

g) the hybridoma having the ATCC Patent Deposit Designation PTA-6872;

h) the hybridoma having the ATCC Patent Deposit Designation PTA-6875; and

i) the hybridoma having the ATCC Patent Deposit Designation PTA-6873.

8. The method of claim 7 , wherein the antibody is humanized.

9. The method of claim 7 , wherein the antibody is an antibody fragment.

10. The method of claim 9 , wherein the antibody fragment is selected from the group consisting of:

a) a single-chain antibody;

b) a Fab fragment;

c) a ″Fab′ fragment; and

d) a F(ab′)2 fragment.

11. The method of claim 7 , wherein the antibody is a chimeric antibody.

12. A method of antagonizing signal transduction in dorsal root ganglion cells in vitro, comprising administering an antibody to the dorsal root ganglion cells wherein the antibody binds Interleukin 31 (IL-31) and whereby the signal transduction is inhibited.

13. The method of claim 12 , wherein the antibody is a monoclonal antibody.

14. The method of claim 12 , wherein the antibody is humanized.

15. The method of claim 12 , wherein the antibody is an antibody fragment.

16. The method of claim 15 , wherein the antibody fragment is selected from the group consisting of:

a) a single-chain antibody;

b) a Fab fragment;

c) a ″Fab′ fragment; and

d) a F(ab′)2 fragment.

17. The method of claim 12 , wherein the antibody is a chimeric antibody.

18. The method of claim 12 , wherein the antibody is produced by the hybridoma selected from the group consisting of:

a) the hybridoma having the ATCC Patent Deposit Designation PTA-6815;

b) the hybridoma having the ATCC Patent Deposit Designation PTA-6816;

c) the hybridoma having the ATCC Patent Deposit Designation PTA-6829;

d) the hybridoma having the ATCC Patent Deposit Designation PTA-6830;

e) the hybridoma having the ATCC Patent Deposit Designation PTA-6831;

f) the hybridoma having the ATCC Patent Deposit Designation PTA-6871;

g) the hybridoma having the ATCC Patent Deposit Designation PTA-6872;

h) the hybridoma having the ATCC Patent Deposit Designation PTA-6875; and

i) the hybridoma having the ATCC Patent Deposit Designation PTA-6873.

19. The method of claim 18 , wherein the antibody is humanized.

20. The method of claim 18 , wherein the antibody is an antibody fragment.

21. The method of claim 20 , wherein the antibody fragment is selected from the group consisting of:

a) a single-chain antibody;

b) a Fab fragment;

c) a ″Fab′ fragment; and

d) a F(ab′)2 fragment.

22. The method of claim 21 , wherein the antibody is a chimeric antibody.

23. A method of antagonizing signal transduction in dorsal root ganglion cells in vitro, comprising admixing a receptor with the dorsal root ganglion cells wherein the receptor binds Interleukin 31 (IL-31) and whereby the signal transduction is inhibited.

24. The method of claim 23 , wherein the receptor comprises the amino acid sequence of SEQ ID NO: 6.

25. The method of claim 23 , wherein the receptor comprises the amino acid of SEQ ID NO: 8.

26. The method of claim 23 , wherein the receptor is a soluble receptor comprising the amino acid sequence of SEQ ID NO: 10.

27. The method of claim 23 , wherein the receptor is a soluble receptor comprising the amino acid sequence of SEQ ID NO: 12.

28. A method of antagonizing signal transduction in dorsal root ganglion cells in vitro, comprising administering a receptor to the dorsal root ganglion cells wherein the receptor binds Interleukin 31 (IL-31) and whereby the signal transduction is inhibited.

29. The method of claim 28 , wherein the receptor comprises the amino acid sequence of SEQ ID NO: 6.

30. The method of claim 28 , wherein the receptor comprises the amino acid of SEQ ID NO: 8.

31. The method of claim 28 , wherein the receptor is a soluble receptor comprising the amino acid sequence of SEQ ID NO:10.

32. The method of claim 28 , wherein the receptor is a soluble receptor comprising the amino acid sequence of SEQ ID NO: 12.
