Dialkylphenyl compounds having β
This invention relates to compounds of formula I: wherein R 1 and R 2 are as defined in the specification, or a pharmaceutically acceptable salt or solvate or stereoisomer thereof. The invention also relates to pharmaceutical compositions and combinations comprising such compounds, processes and intermediates for preparing such compounds, and methods of using such compound to, for example, treat pulmonary disorders, such as chronic obstructive pulmonary disease and asthma.
1. A compound of formula I:
wherein
R 1 is methyl or ethyl;
R 2 is methyl or ethyl;
or a pharmaceutically acceptable salt or stereoisomer thereof.
2. A compound of formula II:
wherein
R 1 is methyl or ethyl;
R 2 is methyl or ethyl;
or a pharmaceutically acceptable salt thereof.
3. A compound of formula IIa:
4. A pharmaceutically acceptable salt of a compound of formula IIa:
5. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and the compound of any one of claims 1 to 4 .
6. A process of preparing the compound of claim 1 , the process comprising deprotecting a compound of formula 6:
wherein P 1 is a hydroxyl-protecting group, to provide a compound of formula I.
7. A process of preparing the compound of claim 1 , the process comprising deprotecting a compound of formula 6a:
wherein R a , R b and R c are independently selected from C 1-4 alkyl, phenyl, —C 1-4 alkyl-(phenyl), or one of R 1a , R 1b and R 1c is —O—(C 1-4 alkyl); to provide a compound of formula I.
8. A process of preparing the compound of claim 1 , the process comprising:
(a) reacting a compound of formula 4:
with a compound of formula 5:
wherein P 1 is a hydroxyl-protecting group, in the presence of a reducing agent to provide a compound of formula 6:
and
(b) deprotecting the compound of formula 6 to provide a compound of formula I.
9. A process of preparing a pharmaceutically acceptable salt of the compound of claim 1 , the process comprising contacting a compound of formula I in free base form with a pharmaceutically acceptable acid.
10. Biphenyl-2-ylcarbamic acid 1-[2-(4-{[(R)-2-(3-formylamino-4-hydroxyphenyl)-2-hydroxyethylamino]methyl}-2,5-dimethylphenyl-carbamoyl)ethyl]piperidin-4-yl ester L-tartaric acid salt.
