Abstract

Disclosed is the surprising discovery that aminophospholipids, such as phosphatidylserine and phosphatidylethanolamine, are specific, accessible and stable markers of the luminal surface of tumor blood vessels. The present invention thus provides aminophospholipid-targeted diagnostic and therapeutic constructs for use in tumor intervention. Antibody-therapeutic agent conjugates and constructs that bind to aminophospholipids are particularly provided, as are methods of specifically delivering therapeutic agents, including toxins and coagulants, to the stably-expressed aminophospholipids of tumor blood vessels, thereby inducing thrombosis, necrosis and tumor regression.

Claims (23)

1. A method for delivering a selected diagnostic agent to tumor vasculature, comprising administering to an animal having a vascularized tumor a binding ligand that comprises said selected diagnostic agent operatively attached to an antibody, or antigen-binding fragment thereof, which binds to phosphatidylserine on the luminal surface of blood vessels of the vascularized tumor.

2. A method for imaging a vascularized tumor, comprising administering to an animal having a vascularized tumor a diagnostically effective amount of a binding ligand that comprises a detectable agent operatively attached to a first antibody, or antigen-binding fragment thereof, which binds to phosphatidylserine on the luminal surface of blood vessels of the vascularized tumor, and detecting the image of the tumor so formed.

3. The method of claim 2 , wherein said first antibody, or antigen-binding fragment thereof, binds to a phosphatidylserine-protein complex on the luminal surface of blood vessels of said vascularized tumor.

4. The method of claim 2 , wherein said first antibody is an IgM antibody.

5. The method of claim 2 , wherein said first antibody is an IgG antibody.

6. The method of claim 2 , wherein said first antibody is an scFv, Fv, Fab′, Fab or F(ab′) 2 antigen-binding region of said antibody.

7. The method of claim 2 , wherein said first antibody is a recombinant antibody or antigen-binding fragment thereof.

8. The method of claim 2 , wherein said first antibody is a human, humanized or part-human chimeric antibody or antigen-binding fragment thereof.

9. The method of claim 2 , wherein said first antibody is a dimer, trimer or multimer of said antibody or antigen-binding fragments thereof.

10. The method of claim 2 , wherein said first antibody is a monoclonal antibody or antigen-binding fragment thereof.

11. The method of claim 3 , wherein said first antibody, or antigen-binding fragment thereof, binds to a phosphatidylserine and β 2 -glycoprotein I complex on the luminal surface of blood vessels of said vascularized tumor.

12. The method of claim 2 , wherein said binding ligand is formulated as a liposomal formulation.

13. The method of claim 2 , wherein said detectable agent is:

(a) the X-ray detectable compound bismuth (III), gold (III), lanthanum (III) or lead (II);

(b) the detectable radioactive ion copper 67 , gallium 67 , gallium 68 , indium 111 , indium 113 , iodine 123 , iodine 125 , iodine 131 , mercury 197 , mercury 203 , rhenium 186 , rhenium 188 , rubidium 97 , rubidium 103 , technetium 99m or yttrium90; or

(c) the detectable nuclear magnetic spin-resonance isotope cobalt (II), copper (II), chromium (III), dysprosium (III), erbium (III), gadolinium (III), holmium (III), iron (II), iron (III), manganese (II), neodymium (III), nickel (II), samarium (III), terbium (III), vanadium (II) or ytterbium (III).

14. The method of claim 2 , wherein said animal is a human patient.

A method for delivering a selected diagnostic agent to tumor vasculature, comprising administering to an animal having a vascularized tumor a binding ligand that comprises said selected diagnostic agent operatively attached to an antibody, or antigen-binding fragment thereof, which binds to phosphatidylserine on the luminal surface of tumor vascular endothelial cells of the blood vessels of said vascularized tumor.

A method for imaging tumor vasculature, comprising administering to an animal having a vascularized tumor a diagnostically effective amount of a binding ligand that comprises a detectable agent operatively attached to a first antibody, or antigen-binding fragment thereof, which binds to phosphatidylserine on the luminal surface of tumor vascular endothelial cells of the blood vessels of said vascularized tumor, and detecting the image of the tumor vasculature so formed.

17. The method of claim 16 , wherein said first antibody, or antigen-binding fragment thereof, binds to a phosphatidylserine-protein complex on the luminal surface of said tumor vascular endothelial cells of said blood vessels of said vascularized tumor.

18. The method of claim 17 , wherein said first antibody, or antigen-binding fragment thereof, binds to a phosphatidylserine and β 2 -glycoprotein I complex on the luminal surface of said tumor vascular endothelial cells of said blood vessels of said vascularized tumor.

19. The method of claim 16 , wherein said first antibody is a recombinant, human, humanized or part-human chimeric antibody or antigen-binding fragment thereof.

20. The method of claim 16 , wherein said animal is a human patient.
