Abstract

The present invention is directed toward a humanized neutralizing monoclonal antibody to hepatocyte growth factor, a pharmaceutical composition comprising same, and methods of treatment comprising administering such a pharmaceutical composition to a patient.

Claims (8)

1. A humanized monoclonal antibody (mAb) that binds and neutralizes human Hepatocyte Growth Factor comprising a mature heavy chain variable region having the amino acid sequence of SEQ ID NO:5 and a mature light chain variable region having the amino acid sequence of SEQ ID NO:8.

2. A humanized monoclonal antibody (mAb) that binds and neutralizes human Hepatocyte Growth Factor comprising a mature heavy chain variable region having the amino acid sequence of SEQ ID NO:5 except the first amino acid is replaced with Gln and a mature light chain variable region having the amino acid sequence of SEQ ID NO:8.

A humanized monoclonal antibody (mAb) that binds and neutralizes human Hepatocyte Growth Factor (HGF), the humanized antibody comprising humanized heavy and light chains, the humanized heavy chain comprising CDRs from L2G7 (ATCC PTA-5162) and a human heavy chain variable region framework provided that at least positions H29, H30, H48, H66, H67, H71, H94 are occupied by the amino acids occupying the corresponding position in the L2G7 heavy chain; the humanized light chain comprising CDRs from L2G7 and a human light chain variable region framework provided that at least positions L3 and L60are occupied by the amino acid occupying the corresponding position in the L2G7 light chain.

4. A humanized mAb of claim 1 , 2 or 3 which is of IgG1, K isotype.

5. A humanized mAb of claim 3 which neutralizes biological activities of HGF as well as L2G7 (ATCC PTA-5162) does.

6. The humanized monoclonal antibody (mAb) of claim 3 , further provided position H1 is occupied by Glu.

7. A cell line producing a mAb of claim 1 , 2 or 3 .

8. A pharmaceutical composition comprising a mAb of claim 1 , 2 or 3 .
