Abstract

A gene which encodes a murine leukocyte α(1,3)fucosyltransferase capable of synthesizing the sialyl Lewis x determinant has been cloned.

Claims (10)

1. A method of preparing a sialyl Lewis x determinant, the method comprising contacting a recombinant murine α(1,3)fucosyltransferase (Fuc-TVII) enzyme catalytic domain with a GDP-fucose donor substrate and a sialyl-N-acetyl-lactosamine acceptor substrate in an enzyme bioreactor under conditions that allow the addition of an α1,3 linked fucose to the sialyl-N-acetyl-lactosamine acceptor substrate, wherein the recombinant murine Fuc-TVII enzyme comprises a catalytic domain is encoded by a nucleic acid sequence segment that is identical to a polynucleotide that is amplified using murine cDNA shown in SEQ ID NO:1 as a template by a 5′ primer as shown in SEQ ID NO:3 and a 3′ primer as shown in SEQ ID NO:4.

2. The method of claim 1 , wherein the Fuc-TVII enzyme catalytic domain is in solution.

3. The method of claim 1 , wherein the Fuc-TVII enzyme catalytic domain is immobilized on a solid phase matrix.

4. The method of claim 1 , wherein the Fuc-TVII enzyme catalytic domain is produced in a mammalian host cell.

5. The method of claim 1 , wherein the Fuc-TVII enzyme catalytic domain is produced in a baculovirus host.

6. The method of claim 1 , wherein the sialyl-N-acetyl-lactosamine acceptor is on a glycoprotein.

7. The method of claim 1 , wherein the sialyl-N-acetyl-lactosamine acceptor is on a glycolipid.

8. The method of claim 1 , wherein the sialyl-N-acetyl-lactosamine acceptor is a free oligosaccharide.

9. A recombinant murine Fuc-TVII enzyme catalytic domain encoded by a nucleic acid sequence segment that is identical to a polynucleotide that is amplified using murine cDNA shown in SEQ ID NO:1 as a template by a 5′ primer as shown in SEQ ID NO:3 and a 3′ primer as shown in SEQ ID NO:4.

10. A recombinant murine Fuc-TVII enzyme catalytic domain encoded by a nucleic acid segment consisting of residue 2194 to residue 3085 of SEQ ID NO:1.
