Abstract

The present invention is a synthetic microfluidic microvasculature network and associated methods. The synthetic microfluidic microvasculature network mimics the structure, fluid flow characteristics, and physiological behavior of physiological microvasculature networks. Computational methods for simulating flow and particle adherence in synthetic and physiological microvascular systems and methods for determining parameters influencing particle adhesion and drug delivery are also described. The invention has many uses including the optimization of drug delivery and microvascular treatments and in describing disease mechanisms that affect the microvasculature such as inflammation, diabetes and hypertension.

Claims (12)

1. A microfluidic microvascular chip comprising:

one or more fluid inlets,

one or more fluid outlets, and

a plurality of non-linear flow channels forming a synthetic microvascular network allowing fluid flow between one or more fluid inlets and one or more fluid outlets

wherein

said non-linear flow channels forming said synthetic microvascular network possess a geometric characteristic selected from the group consisting of a variable cross-sectional shape, a variable cross-sectional area, a turn, a bend, a bifurcation, a junction, a convolution, an anastomosis, and combinations thereof.

2. The microfluidic microvascular chip of claim 1 , wherein the synthetic microvascular network is a replica of either an optically imaged physiological microvasculature network or an average of a plurality of optically imaged physiological microvasculature networks.

3. The microfluidic microvascular chip of claim 2 , wherein the physiological microvasculature network comprises at least one feature selected form the group consisting of an arteriole, a capillary, a post-capillary venule, a secretory ductule and an exocrine ductule.

4. The microfluidic microvascular chip of claim 1 , wherein the non-linear flow channels forming the synthetic microvascular network have the dimensions of at least one of an arteriole, a capillary, a post-capillary venule, a secretory ductule, and an exocrine ductule.

5. The microfluidic microvascular chip of claim 1 , wherein one or more surfaces of the synthetic microvascular network comprises cells, biomolecules, or bioactive chemicals.

6. The microfluidic microvascular chip of claim 1 , wherein one or more walls of the non-linear flow channels forming the synthetic microvascular network comprises a porous membrane.

The microfluidic microvascular chip of claim 1 , wherein said microfluidic microvascular chip is made from one or more materials selected from the group consisting of polydimethyliloxane, poly(styrene-butadiene-styrene), poly(styrene-ethylene-butadiene-styrene), polyester-ether thermoplast, thermoset polyester, polyolefin plastomer, glass, quartz, polymethylmethacrylate, polycarbonate, cyclic olefin copolymer, polystyrene, poly vinyl chloride, polyethylene terephthalate glycol, cellulose acetate, polyethylene terephthalate, polyimide, and combinations thereof.
