Abstract

The present invention provides mutant viruses with a decreased ability to block nuclear transport of mRNA or protein in an infected cell which are attenuated in vivo. The mutant viruses of the present invention may also be capable of triggering the anti-viral systems of normal host cells while remaining sensitive to the effects of these systems. The present invention further provides for the use of the mutant viruses in a range of applications including, but not limited to, as therapeutics for the treatment of cancer and infections, as vaccines and adjuvants, as viral vectors, and as oncolytic and cytolytic agents for the selective lysis of malignant or infected cells.

Claims (17)

1. A mutant vesicular stomatitis virus (VSV) having the mutation ΔM51 in the gene encoding the matrix (M) protein.

2. The mutant VSV according to claim 1 , further comprising one or more mutations in the gene encoding the matrix (M) protein selected from the group consisting of ΔM51-54, ΔM51-57, ΔV221-S226, V221X, S226X, or a combination thereof.

3. The mutant VSV according to claim 1 , comprising one or more mutations in the gene encoding the matrix (M) protein selected from the group consisting of: ΔM51/V221F; ΔM51-54/V221F; ΔM51-57/V221 F; ΔM51/S226R; ΔM51-54/S226R, and ΔM51-57/S226R.

4. The mutant VSV according to claim 1 , comprising one or more mutations in the gene encoding the matrix (M) protein selected from the group consisting of: ΔM51/V221F/S226R; ΔM51-54/V221F/S226R and ΔM51-57/V221F/S226R.

5. The mutant VSV according to claim 1 , wherein said mutant VSV is capable of triggering the production of one or more cytokines in an infected cell.

6. The mutant VSV according to claim 1 , further comprising a heterologous nucleic acid.

7. A vaccine vector comprising a mutant VSV having the mutation ΔM51 in the matrix (M) protein and a heterologous nucleic acid encoding one or more antigens.

8. A vaccine adjuvant comprising a mutant VSV having the mutation ΔM51 in the matrix (M) protein, said mutant VSV being capable of triggering the production of one or more cytokines in an infected cell.

9. A selective oncolytic agent comprising a mutant VSV having the mutation ΔM51 in the matrix (M) protein.

10. A pharmaceutical composition comprising a mutant VSV having the mutation ΔM51 in the matrix (M) protein.

11. An immunogenic composition comprising a mutant VSV having the mutation ΔM51 in the matrix (M) protein and a pharmaceutically acceptable carrier, said mutant VSV being capable of triggering the production of one or more cytokines in an infected cell.

12. A kit comprising one or more containers and a mutant VSV having the mutation ΔM51 in the gene encoding the matrix (M) protein.

13. A method of inducing an immune response, the method comprising administering the vaccine vector of claim 7 to an animal.

14. The method of claim 13 , wherein the animal is a human.

15. A method of inhibiting minor cell growth in an animal, the method comprising treating the animal with the oncolytic agent of claim 9 .

16. The method of claim 15 , wherein the animal is a human.

17. A method of expressing a heterologous nucleic acid by infecting a host cell with the VSV of claim 6 .
