Abstract

Methods and devices for preparing a solid-fibrin web are provided. One method may include drawing blood from a patient, separating plasma from the blood, contacting the plasma with a calcium-coagulation activator and concurrently coagulating and axially centrifuging the plasma to form the solid-fibrin web. The solid-fibrin web may be suitable for regenerating body tissue in a living organism. Devices used in the methods may also be provided.

Claims (16)

1. A device for use in axial centrifugation, the device comprising:

a container configured for axial centrifugation, the container including

a primary chamber that can receive blood including an anticoagulant;

a secondary chamber containing a coagulator; and

a filter separating the primary chamber from the secondary chamber,

the filter substantially preventing red and white blood cells, originating from blood drawn into the primary chamber, from entering the secondary chamber under a centrifugal force of about 1000×G or greater, but substantially permitting plasma and platelets originating from the blood to flow into the secondary chamber under a centrifugal force of about 1000×G or greater.

2. The device of claim 1 , wherein the primary chamber contains an anti-coagulant.

3. The device of claim 1 , wherein the coagulator comprises calcium.

4. The device of claim 3 , wherein the coagulator comprises at least one calcium chloride, calcium fluoride, calcium carbonate, calcium gluconate, calcium fumarate, calcium pyruvate, an organic calcium salt and combinations thereof.

5. A device for use in axial centrifugation, the device comprising:

a container configured for axial centrifugation, the container including

(a) a primary chamber that can receive blood, wherein the primary chamber includes a separation medium comprising at least one of a silicone gel, a polyester gel, a thixotropic gel and a combination thereof,

(b) a secondary chamber containing a coagulator,

(c) a diaphragm that separates the primary chamber from the secondary chamber wherein the diaphragm has at least one vent that is plugged with the separation medium wherein said separation medium-plugged diaphragm prevents fluid communication between the primary and secondary chambers prior to centrifugation, wherein the separation medium moves into the primary chamber during centrifugation of about 1000×G or greater to provide fluid communication between the primary and secondary chambers through the diaphragm through the at least one vent, wherein the separation medium forms a barrier in the primary chamber that substantially prevents red and white blood cells, originating from the blood drawn into the primary chamber, from entering the secondary chamber during centrifugation and wherein the plasma and platelet enter the secondary chamber during centrifugation.

6. The device of claim 5 , wherein the separation medium substantially prevents red and white blood cells, originating from blood drawn into the primary chamber, from entering the secondary chamber after centrifugation.

7. The device of claim 5 , wherein the coagulator comprises at least one of calcium chloride, calcium fluoride, calcium carbonate, calcium gluconate, calcium fumarate, calcium pyruvate, an organic calcium salt and combinations thereof.
