Abstract

Disclosed are dihydropteridinones of general formula (I) wherein the groups L and R 1 -R 5 have the meanings given in the claims and specification, the pharmaceutical compositions, isomers, intermediates thereof and processes for preparing these dihydropteridinones. The compounds are useful the treatment and/or prevention of cancer, infections, inflammatory and autoimmune diseases.

Claims (60)

1. A process for preparing a compound of formula

comprising the steps of:

(i) reacting a compound of formula

 with a compound of formula

 to provide a compound of formula

(ii) reducing and cyclizing the compound of formula

 to a compound of formula

and

(iii) methylating with a methylating agent the compound of formula

 to provide the compound of formula

2. A process for preparing a compound of formula

comprising the steps of:

(i) reacting a compound of formula

 with a compound of formula

 to provide a compound of formula

and

(ii) reducing and cyclizing the compound of formula

 to the compound of formula

3. A process for preparing a compound of formula

comprising the steps of:

(i) reducing and cyclizing a compound of formula

 to a compound of formula

and

(ii) methylating with a methylating agent the compound of formula

 to provide the compound of formula

4. A process for preparing a compound of formula

 comprising reducing and cyclizing a compound of formula

 to the compound of formula

5. A process for preparing a compound of formula

 comprising methylating with a methylating agent a compound of formula

to the compound of formula

6. A compound of formula

7. A compound of formula

8. A process for preparing a compound of formula

 comprising the steps of:

(i) A compound of formula

R1 = Me, Et

 is dissolved in an organic solvent and reacted under basic conditions together with a compound of formula

resulting in a compound of formula

(ii) The nitro compound is reduced and cyclized under acidic conditions in an exothermic reaction resulting in a compound of formula

(iii) Methylation with a methylating agent resulting in a compound of formula

9. A process for preparing a compound of formula

 comprising the steps of:

(i) A compound of formula R1 = Me, Et

 is dissolved in an organic solvent and reacted under basic conditions together with a compound of formula

 resulting in a compound of formula

(ii) The nitro compound is reduced and cyclized under acidic conditions in an exothermic reaction resulting in a compound of formula

10. A process for preparing a compound of formula

 comprising the steps of:

(i) reducing and cyclizing the nitro compound of formula

 under acidic conditions in an exothermic reaction resulting in a compound of formula

(ii) Methylation with a methylating agent resulting in a compound of formula

11. A process for preparing a compound of formula

comprising the steps of:

(i) reducing and cyclizing the nitro compound of formula

 under acidic conditions in an exothermic reaction resulting in a compound of formula

12. A process for preparing a compound of formula

 comprising the step of:

(i) Methylation with a methylating agent a compound of formula

 resulting in a compound of formula
