Abstract

The present invention provides novel amino acid compounds useful in detecting and evaluating brain and body tumors. These compounds have the advantageous properties of rapid uptake and prolonged retention in tumors and can be labeled with halogen isotopes such as fluorine-18, iodine-123, iodine-124, iodine-125, iodine-131, bromine-75, bromine-76, bromine-77, bromine-82, astatine-210, astatine-211, and other astatine isotopes. These compounds can also be labeled with technetium and rhenium isotopes using known chelation complexes. The compounds disclosed herein bind tumor tissues in vivo with high specificity and selectivity when administered to a subject. Preferred compounds show a target to non-target ratio of at least 2:1, are stable in vivo and substantially localized to target within 1 hour after administration. Preferred compounds include 1-amino-2-[ 18 F]fluorocyclobutyl-1-carboxylic acid (2-[ 18 F]FACBC) and 1-amino-2-[ 18 F]fluoromethylcyclobutyl-1-carboxylic acid (2-[ 18 F]FMACBC). The labeled amino acid compounds of the invention are useful as imaging agents in detecting and/or monitoring tumors in a subject by PET or SPECT.

Claims (32)

1. An amino acid analog having Formula I:

wherein Y is (CR 5 ,R 6 )n, n=1 or 2; X is selected from the group consisting of halogen, C1-C6 haloalkyl and C1-C6 haloalkenyl, where halo or halogen in X is selected from the group consisting of F, Cl, Br, I, At, F-18, Br-76, I-123, I-124, I-125, and I-131; R 4 is H; R 5 and R 6 are independently selected from the group consisting of H, and C1-C6 alkyl; R 1 and R 2 are independently selected from the group consisting of H, and C1-C6 alkyl; R 3 is selected from the group consisting of H, and C1-C6 alkyl;

or a pharmaceutically acceptable salt thereof.

2. The amino acid analog of claim 1 wherein X is selected from the group consisting of halo, C1-C6 haloalkyl and haloalkenyl C1-C6 .

3. The amino acid analog of claim 2 wherein R 1 , R 2 , R 3 , R 5 , and R 6 are selected independent of each other from the group consisting of hydrogen, and C1-C6 alkyl.

4. The amino acid analog of claim 2 or 3 wherein X is selected from the group consisting of halogen, C1-C6 haloalkyl, and C1-C6 haloalkenyl, wherein halo is either 18 F or 123 I.

5. The amino acid analog of claim 4 wherein X is halogen or C1-C4 haloalkyl and R 1 , R 2 , R 3 , R 5 , and R 6 are independent of each other hydrogen or C1-C4 alkyl.

6. The amino acid analog of claim 4 or 5 wherein Y is CH 2 .

7. The amino acid analog of claim 6 wherein the analog is 2-[ 18 F]FACBC.

8. The amino acid analog of claim 6 wherein the analog is 2-[ 123 I]IACBC

9. The amino acid analog of claim 6 wherein the analog is 2-[ 18 F]FMACBC

10. The amino acid analog 2-[ 123 I]IVACBC

11. The amino acid analog 2-[ 123 I]IEACBC

12. A compound of the formula:

wherein Y is (CR 5 )n, n =1 or 2; X is selected from the group consisting of halogen, C1-C6 haloalkyl, and C1-C6 haloalkenyl, where halo or halogen in X is selected from the group consisting of F, Cl, Br, I, At, F-18, Br-76, I-123, I-124, I-125, and I-131; R 4 is H; R 5 and R 6 are independently selected from the group consisting of H, and C1-C6 alkyl, where halo is non-radioactive F, Cl, Br and I; R 1 and R 2 are independently selected from the group consisting of H, and C1-C6 alkyl; R 3 is selected from the group consisting of H, and C1-C6 alkyl;

or a pharmaceutically acceptable salt thereof.

13. The compound of claim 12 wherein X is selected from the group consisting of halogen, C1-C6 haloalkyl, and C1-C6 haloalkenyl.

14. The compound of claim 13 wherein R 1 , R 2 , R 3 , R 5 , and R 6 are selected independent of each other from the group consisting of hydrogen, and C1-C6 alkyl.

15. The compound claim 14 wherein X is halogen or C1-C4 haloalkyl and R 1 , R 2 , R 3 , R 5 , and R 6 are independent of each other hydrogen or C1-C4 alkyl.

16. The compound of claim 15 wherein X is halogen and R 1 , R 2 , R 3 , R 4 , R 5 , and R 6 are hydrogen, and Y is CH.

17. The compound of claim 16 wherein X is 18 F.

18. The compound of claim 16 wherein X is 123 I.

19. A method of synthesizing an amino acid analog according to claim 1 wherein the method comprises the step of reacting a compound of formula III with a halogenated potassium compound to yield the amino acid analog of formula I, wherein formula III is

wherein Y is selected from the group consisting of (CR 5 ,R 6 )n, n=1-2; R 4 is H; R 5 and R 6 are independently selected from the group consisting of H, and C1-C6 alkyl; R 1 is selected from the group consisting of H, and C1-C6 alkyl; R 3 is selected from the group consisting of H, and C1-C6 alkyl.

20. A method of synthesizing the amino acid analog 2-[ 18 F]FACBC wherein the method comprises the step of reacting a compound of formula III with a halogenated potassium compound to yield the amino acid analog 2-[ 18 F]FACBC, wherein formula III is

wherein Y is CH 2 ; R 4 is H; R 1 is H; R 3 is selected from the group consisting of H, and C1-C6 alkyl.

21. A diagnostic composition for imaging a tumor, comprising a radiolabeled amino acid analog of claim 1 , and a pharmaceutically acceptable carrier.

22. The diagnostic composition of claim 21 wherein the labeled amino acid analog is 2-[ 18 F]FACBC

23. A method of tumor imaging by positron emission tomography or single photon emission computed tomography, comprising: a) administering to a subject suspected of having a tumor an image-generating amount of a labeled amino acid analog of claim 1 ; b) allowing the labeled amino acid analog to become associated with the tumor; and c) measuring the distribution of the labeled amino acid analog in the subject by PET or SPECT.

24. The method of claim 23 wherein the labeled amino acid analog is 2-[ 18 F]FACBC

25. A method of tumor imaging using beta emitting amino acid analogs, comprising: a) administering to a subject suspected of having a tumor an image-generating amount of an amino acid analog of claim 1 labeled with a beta particle emitter; b) allowing the labeled amino acid analog to become associated with the tumor; and c) measuring the distribution of the beta particle emitting amino acid analog in the subject by PET or SPECT.
