Inhibiting serum response factor (SRF) to improve insulin sensitivity
Described are methods of improving glycemic control/improving insulin sensitivity by administering an inhibitor of serum response factor (SRF) activity, and methods of identifying new compounds for use in the described methods of treatment.
1. A method for improving insulin sensitivity in a subject, the method comprising:
selecting a subject in need of improved insulin sensitivity; and administering to the subject a composition comprising a therapeutically effective amount of a small molecule that reduces human serum response factor (SRF) transcriptional activity, thereby improving insulin sensitivity in the subject.
2. The method of claim 1 , wherein the small molecule is CCG-1423.
3. The method of claim 1 , wherein the small molecule is distamycin A.
4. The method of claim 1 , wherein the subject has type 2 diabetes mellitus.
5. The method of claim 4 , wherein the subject has a fasting blood glucose level of 126 mg/dL and above on more than one testing occasion.
6. The method of claim 4 , wherein the subject is not pregnant but records a score of over 200 mg/dL about two-hours after consuming a 75-gram glucose drink on more than one testing occasion, as assessed by the oral glucose tolerance test (OGTT).
7. The method of claim 1 , wherein the subject is at risk of developing type 2 diabetes.
8. The method of claim 7 , wherein the subject presents a risk factor for developing type 2 diabetes selected from the group consisting of: a body mass index (BMI) of 30 or above, age of 45 years old or greater, low birth weight, family history of diabetes, low high density lipoprotein (HDL) cholesterol and high triglycerides, high blood pressure, personal history of gestational diabetes, gave birth to a baby weighing more than nine pounds, minority group background, and low cardiorespiratory fitness.
9. The method of claim 7 , wherein the subject has pre-diabetes as identified by a fasting blood glucose level of 100-125 mg/dL or a score of 140-200 mg/dL about two-hours after consuming a 75-gram glucose drink as assessed by the OGTT.
10. The method of claim 7 , wherein the subject is not taking drugs prescribed to treat diabetes.
11. The method of claim 1 , wherein the subject is insulin resistant.
12. The method of claim 1 , wherein the subject has a metabolic syndrome.
13. The method of claim 1 , wherein the small molecule is MG132.
