T2R taste receptors and genes encoding same
Functional assays that detect the effect of a particular compound or compounds on the activation of at least one human T2R polypeptide are provided. These assays include e.g., assays which detect the effect of said compound on intracellular calcium, second messengers such as cAMP, cGMP, current e.g., by use of voltage-clamp or patch-clamp, techniques, fluorescence polarization or FRET assays, and the like. These assays are useful in identifying compounds that putatively elicit or modulate (inhibit or enhance) bitter taste in human subjects, e.g. bitter taste blockers. The effect of identified compounds on taste may be further evaluated in human or animal taste tests.
1. An isolated G protein coupled receptor (GPCR) polypeptide which is at least 95% identical to the polypeptide contained in SEQ ID NO:4 or which is at least 95% identical to the entire extracellular or the transmembrane region of said polypeptide.
2. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 95% identical to the polypeptide contained in SEQ ID NO:4.
3. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 95% identical to the entire extracellular region of said polypeptide.
4. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 95% identical to the entire transmembrane region of said polypeptide.
5. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 96% identical to the polypeptide contained in SEQ ID NO:4.
6. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 96% identical to the entire extracellular region of said polypeptide.
7. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 96% identical to the entire transmembrane region of said polypeptide.
8. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 97% identical to the polypeptide contained in SEQ ID NO:4.
9. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 97% identical to the entire extracellular region of said polypeptide.
10. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 97% identical to the entire transmembrane region of said polypeptide.
11. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 98% identical to the polypeptide contained in SEQ ID NO:4.
12. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 98% identical to the entire extracellular region of said polypeptide.
13. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 98% identical to the entire transmembrane region of said polypeptide.
14. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 99% identical to the polypeptide contained in SEQ ID NO:4.
15. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 99% identical to the entire extracellular region of said polypeptide.
16. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is at least 99% identical to the entire transmembrane region of said polypeptide.
17. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is identical to the polypeptide contained in SEQ ID NO:4.
18. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is identical to the entire extracellular region of said polypeptide.
19. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is identical to the entire transmembrane region of said polypeptide.
20. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is fused to another polypeptide.
21. The isolated G protein coupled receptor (GPCR) polypeptide of claim 20 wherein the other polypeptide is a detectable marker or another G protein coupled receptor (GPCR) polypeptide.
22. The isolated G protein coupled receptor (GPCR) polypeptide of claim 1 which is immobilized on a solid support.
