Abstract

Novel phototherapeutic methods and compositions are described herein. Nanoparticle-assembled microcapsules as a new type of delivery vehicle for photosensitive compounds may be synthesized through a two-step assembly process. Charged polymer chains and counterions may be combined with a photosensitive compound to form photosensitive aggregates, and then nanoparticles may be combined with the aggregates to form the microcapsules. The shell may be composed of nanoparticles and/or polymer, and the core interior may contain the photosensitive compound. Formation occurs rapidly (on the order of seconds) and the conditions are very mild (at room temperature, in aqueous solution, and at neutral pH). The microcapsule synthesis is highly suitable as an encapsulation method, particularly for a charged photosensitive molecule like ICG.

Claims (11)

1. A method of encapsulating a photosensitive compound comprising:

a) providing an amount of a polyelectrolyte having a charge and an amount of a counterion having a valence of at least 2;

b) combining the polyelectrolyte and the counterion in a solution such that the polyelectrolyte self-assembles to form one or more aggregate cores;

c) adding the one or more photosensitive compounds to the aggregate cores to form one or more photosensitive aggregate cores; and

d) adding a coating material to the photosensitive aggregate cores such that the coating material arranges itself around the photosensitive aggregate cores to form a shell and encapsulate the photosensitive compound.

2. The method of claim 1 wherein the photosensitive compound has a negative charge.

3. The method of claim 1 wherein the photosensitive compound is indocyanine green.

4. The method of claim 1 wherein the polyelectrolyte is a polyamine and the counterion is a phosphate.

5. The method of claim 1 wherein (c) comprises adding the one or more photosensitive compound at a photosensitive compound to amine ratio ranging from about 0.001 to about 0.18.

6. The method of claim 1 wherein the charge ratio R of total charge of the counterions to the total charge of the polyelectrolyte is about 6.

7. The method of claim 1 , wherein the photosensitive compound is present in an amount of greater than about 20 wt. %.
