Abstract

There are provided compounds of the formula wherein X, Y, R 1 , R 2 , R 3 , R 3 , R 4 , R 5 , R 6 and R 7 are as described herein and enantiomers and pharmaceutically acceptable salts and esters thereof. The compounds are useful as anticancer agents.

Claims (595)

1. A compound of the formula

wherein

X is selected from the group consisting of H, F, Cl, Br, I, cyano, nitro, ethynyl, cyclopropyl, methyl, ethyl, isopropyl, vinyl and methoxy,

Y is one to four group(s) independently selected from the group consisting of H, F, Cl, Br, I, CN, OH, nitro, lower alkyl, cycloalkyl, lower alkoxy, lower alkenyl, cycloalkenyl, lower alkynyl, aryl, hetereoaryl, hetereocycle, COOR′, OCOR′, CONR′R″, NR′COR″, NR″SO 2 R′, SO 2 NR′R″ and NR′R″ wherein

R′ and R″ is independently selected from H or substituted or unsubstituted lower alkyl, substituted or unsubstituted lower cycloalkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower cycloalkenyl, substituted or unsubstituted aryl, substituted or unsubstituted hetereoaryl or substituted or unsubstituted hetereocycle, and in the case of R′ and R″ may independently link to form a cyclic structure selected from substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heteroaryl or substituted or unsubstituted heterocycle,

one of R 1 and R 2 is selected from the group consisting of lower alkyl,

substituted lower alkyl, lower alkenyl, substituted lower alkenyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl and substituted cycloalkenyl and the other is hydrogen or lower alkyl,

R 3 is H or lower alkyl,

one of R 4 and R 5 is selected from the group consisting of lower alkyl, substituted lower alkyl, lower alkenyl, substituted lower alkenyl, aryl, substituted aryl,

heteroaryl, substituted heteroaryl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl, and substituted cycloalkenyl and the other is hydrogen,

R 6 and R 7 are selected from the group consisting of (CH 2 ) n —R′, (CH 2 ) n —NR′R″, (CH 2 ) n —NR′COR″, (CH 2 ) n —NR′SO 2 R″, (CH 2 ) n —COON, (CH 2 ) n —COOR′, (CH 2 ) n —CONR′ R″, (CH 2 ) n —OR′, (CH 2 ) n —SR′, (CH 2 ) n —SOR′, (CH 2 ) n —SO 2 R′, (CH 2 ) n —COR′, (CH 2 ) n —SO 3 H, (CH 2 ) n —SONR′ R″, (CH 2 ) n —SO 2 NR′R″, (CH 2 CH 2 O) m —(CH 2 ) n —R′, (CH 2 CH 2 O) m —(CH 2 ) n —OH, (CH 2 CH 2 O) m —(CH 2 ) n —OR′, (CH 2 CH 2 O) m —(CH 2 ) n —NR′R″, (CH 2 CH 2 O) m —(CH 2 ) n —NR′COR″, (CH 2 CH 2 O) m —(CH 2 ) n —NR′SO 2 R″, (CH 2 CH 2 O) m —(CH 2 ) n —COOH, (CH 2 CH 2 O) m —(CH 2 ) n —COOR′, (CH 2 CH 2 O) m —(CH 2 ) n —CONR′R″, (CH 2 CH 2 O) m —(CH 2 ) n —SO 2 R′, (CH 2 CH 2 O) m —(CH 2 ) n —COR′, (CH 2 CH 2 O) m —(CH 2 ) n —SONR′R″, (CH 2 CH 2 O) m —(CH 2 ) n —SO 2 NR′R″, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —R′, (CH 2 ) p — (CH 2 CH 2 O) m —(CH 2 ) n —OH, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —OR′, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —NR′R″, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —NR′COR″, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —NR′SO 2 R″, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —COOH, (CH 2 ) p —(CH 2 CH 2 O) m —(CH 2 ) n —

m, n and p are independently 0 to −6

and the pharmaceutically acceptable salts and esters thereof.

2. A compound of claim 1 of the formula

X is selected from the group consisting of H, F, Cl, Br, I, cyano, nitro, ethynyl, cyclopropyl, methyl, ethyl, isopropyl, vinyl and methoxy,

Y is one to four group(s) independently selected from the group consisting of H, F, Cl, Br, I, CN, OH, nitro, lower alkyl, cycloalkyl, lower alkoxy, lower alkenyl, cycloalkenyl, lower alkynyl, aryl, hetereoaryl, hetereocycle, COOR′, OCOR′, CONR′R″, NR′COR″, NR″SO 2 R′, SO 2 NR′R″ and NR′R″ wherein

R′ and R″ are independently selected from H or substituted or unsubstituted lower alkyl, substituted or unsubstituted lower cycloalkyl, substituted or unsubstituted lower alkenyl, substituted or unsubstituted lower cycloalkenyl, substituted or unsubstituted aryl, substituted or unsubstituted hetereoaryl or substituted or unsubstituted hetereocycle, and wherein R′ and R″ may independently link to form a cyclic structure selected from substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heteroaryl or substituted or unsubstituted heterocycle,

R 1 is selected from the group consisting of lower alkyl,

substituted lower alkyl, lower alkenyl, substituted lower alkenyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl and substituted cycloalkenyl,

R 2 is hydrogen or lower alkyl,

R 3 is H or lower alkyl,

R 5 is selected from the group consisting of lower alkyl, substituted lower alkyl, lower alkenyl, substituted lower alkenyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl and substituted cycloalkenyl,

R 4 is hydrogen,

3. The compound of claim 2 wherein

X is F, Cl or Br,

Y is one to two group(s) independently selected from the group consisting of H, F, Cl, Br, I, CN, OH, nitro, lower alkyl, cycloalkyl, lower alkoxy, lower alkenyl, lower cycloalkenyl and lower alkynyl,

R 1 is selected from the group consisting of lower alkyl,

substituted lower alkyl, lower alkenyl, substituted lower alkenyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl and substituted cycloalkenyl,

R 2 is hydrogen,

R 3 is H,

R 5 is selected from the group consisting of aryl, substituted aryl, heteroaryl and substituted heteroaryl,

R 4 is hydrogen,

and the pharmaceutically acceptable salts and esters thereof.

4. The compound of claim 2 wherein

X is F, Cl or Br,

Y is a mono-substituting group selected from H or F and

R 1 is selected from the group consisting of lower alkyl,

substituted lower alkyl, lower alkenyl, substituted lower alkenyl, heterocycle, substituted heterocycle, cycloalkyl, substituted cycloalkyl, cycloalkenyl and substituted cycloalkenyl.

5. The compound of claim 2 wherein

R 1 is a substituted lower alkyl of the formula

where R 8 , R 9 are both methyl or linked to form a cyclopropyl, cyclobutyl, cyclopentyl or a cyclohexyl group,

R 10 is (CH 2 ) m —R 11 ,

m is 0, 1 or 2,

R 11 is selected from hydrogen, hydroxyl, lower alkyl, lower alkoxy, aryl, substituted aryl, hetereoaryl, substituted heteroaryl, hetereocycle or substituted heterocycle,

R 2 is H,

R 3 is H,

R 5 is a substituted phenyl selected from

W is F, Cl or Br,

V is H or F,

R 4 is hydrogen,

one of R 6 and R 7 is hydrogen and the other is (CH 2 ) n —R′,

n is 0 or 1 and

R′ is selected from aryl, substituted aryl, hetereoaryl, substituted heteroaryl, hetereocycle or substituted heterocycle.

6. A compound of claim 1 selected from the group consisting of

rac-(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (2-morpholin-4-yl-ethyl)-amide,

(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (2-morpholin-4-yl-ethyl)-amide,

rac-(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid dimethylamide,

rac-(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid ((S)-3,4-dihydroxy-butyl)-amide,

rac-(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (2-piperazin-1-yl-ethyl)-amide,

(S)-2-{[(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carbonyl]-amino}-3-methyl-butyric acid methyl ester,

(S)-2-{[(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carbonyl]-amino}-3-methyl-butyric acid,

8. A compound of claim 1 selected from the group consisting of

(2S,3S,4S,5R)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid ((R)-1-hydroxymethyl-3-methyl-butyl)amide,

rac-(2R,3R,4R,5S)-3-(3-chloro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (3-hydroxy-propyl)-amide,

rac-(2R,3S,4R,5S)-3-(3-chloro-2-fluoro-phenyl)-4-(4-chloro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid [2-(cis-2,6-dimethyl-morpholin-4-yl)-ethyl]-amide,

4-{[(2R,3S,4R,5S)-4-(4-chloro-2-fluoro-phenyl)-3-(3-chloro-2 fluoro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carbonyl]-amino}-3-fluoro-benzoic acid,

Y is one to four group(s) independently selected from the group consisting of H, F, Cl, Br, I, CN, OH, nitro, lower alkyl, cycloalkyl, lower alkoxy, lower alkenyl, cycloalkenyl, lower alkynyl, aryl, hetereoaryl, hetereocycle, COOR′, OCOR′, CONR′R″, NR′COR″, NR″SO 2 R′, SO 2 NR′R″ and NR′R″ wherein

49. A compound of the formula:

(2R,3S,4R,5S)-4-(4-chloro-2-fluoro-phenyl)-3-(3-chloro-2-fluoro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (4-carbamoyl-3-chloro-phenyl)-amide.

50. A compound of the formula:

4-{[(2R,3S,4R,5S)-4-(4-chloro-2-fluoro-phenyl)-3-(3-chloro-2-fluoro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carbonyl]-amino}-3-chloro-benzoic acid methyl ester.

51. A compound of the formula:

rac-(2R,3S,4R,5S)-3-(3-chloro-2-fluoro-phenyl)-4-(4-chloro-2-fluoro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid ((R)-3,4-dihydroxy-butyl)-amide.

52. A compound of the formula:

rac-(2R,3S,4R,5S)-3-(3-chloro-2-fluoro-phenyl)-4-(4-chloro-2-fluoro-phenyl)-4-cyano-5-(2,2-dimethyl-propyl)-pyrrolidine-2-carboxylic acid (1-carbamoyl methyl-1H-pyrazol-3-yl)-amide.
