Abstract

Provided herein are compositions and formulations comprising an antifungal agent. Pharmaceutical compositions comprising luliconazole in an amount effective for the treatment of onychomycosis are provided. Also provided are methods for treating dermatomycoses and onychomycosis using the compositions and formulations.

Claims (228)

1. A method of penetrating a nail with luliconazole in a human individual in need thereof, comprising contacting the individual's nail with a pharmaceutical composition comprising:

from 5 weight percent to 15 weight percent luliconazole;

from 5 weight percent to 25 weight percent acetone;

from 1 weight percent to 15 weight percent propylene carbonate;

from 15 weight percent to 35 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

2. The method of claim 1 , wherein R is ethyl.

3. The method of claim 1 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

4. The method of claim 1 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

5. The method of claim 1 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

6. The method of claim 1 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

7. The method of claim 2 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

8. The method of claim 2 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

9. The method of claim 3 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

10. The method of claim 2 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

11. The method of claim 9 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

12. The method of claim 1 , wherein the composition comprises 10 weight percent luliconazole.

13. The method of claim 1 , wherein the composition comprises 9 weight percent to 12.5 weight percent luliconazole.

14. The method of claim 1 , wherein the composition comprises:

10 weight percent luliconazole;

12 weight percent acetone;

5 weight percent propylene carbonate;

25 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is ethyl; and

4 weight percent benzyl alcohol.

15. The method of claim 14 , wherein the composition further comprises a film forming agent in 1 weight percent.

16. The method of claim 15 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

17. The method of claim 15 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

18. The method of claim 14 , wherein the composition further comprises ethanol in a weight percent that provides a total weight percent of 100 for the composition.

19. The method of claim 15 , wherein the composition further comprises 43 weight percent ethanol.

20. The method of claim 1 , wherein the composition comprises:

from 9 weight percent to 12.5 weight percent luliconazole;

from 8 weight percent to 15 weight percent acetone;

from 3 weight percent to 8 weight percent propylene carbonate;

from 20 weight percent to 30 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

21. The method of claim 1 , wherein the nail is a toenail.

22. The method of claim 1 , wherein the nail is a fingernail.

23. The method of claim 1 , wherein the method comprises contacting the individual's nail with the composition once daily.

24. The method of claim 1 , wherein the individual is diabetic, is of an advanced age or is immunocompromised.

25. A method of delivering luliconazole to a nail bed of a human individual in need thereof, comprising administering to the surface of the individual's nail a pharmaceutical composition comprising:

from 5 weight percent to 15 weight percent luliconazole;

from 5 weight percent to 25 weight percent acetone;

from 1 weight percent to 15 weight percent propylene carbonate;

from 15 weight percent to 35 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

26. The method of claim 25 , wherein R is ethyl.

27. The method of claim 25 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

28. The method of claim 25 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

29. The method of claim 25 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

30. The method of claim 25 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

31. The method of claim 26 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

32. The method of claim 26 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

33. The method of claim 27 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

34. The method of claim 26 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

35. The method of claim 33 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

36. The method of claim 25 , wherein the composition comprises 10 weight percent luliconazole.

37. The method of claim 25 , wherein the composition comprises 9 weight percent to 12.5 weight percent luliconazole.

38. The method of claim 25 , wherein the composition comprises:

10 weight percent luliconazole;

12 weight percent acetone;

5 weight percent propylene carbonate;

25 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is ethyl; and

4 weight percent benzyl alcohol.

39. The method of claim 38 , wherein the composition further comprises a film forming agent in 1 weight percent.

40. The method of claim 39 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

41. The method of claim 39 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

42. The method of claim 38 , wherein the composition further comprises ethanol in a weight percent that provides a total weight percent of 100 for the composition.

43. The method of claim 39 , wherein the composition further comprises 43 weight percent ethanol.

44. The method of claim 25 , wherein the composition comprises:

from 9 weight percent to 12.5 weight percent luliconazole;

from 8 weight percent to 15 weight percent acetone;

from 3 weight percent to 8 weight percent propylene carbonate;

from 20 weight percent to 30 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

45. The method of claim 25 , wherein the nail is a toenail.

46. The method of claim 25 , wherein the nail is a fingernail.

47. The method of claim 25 , wherein the method comprises administering the composition to the surface of the individual's nail once daily.

48. The method of claim 25 , wherein the individual is diabetic, is of an advanced age or is immunocompromised.

49. A method of treating distal subungual onychomycosis in a human individual in need thereof, comprising contacting the individual's nail with a pharmaceutical composition comprising:

from 9 weight percent to 12.5 weight percent luliconazole;

from 8 weight percent to 15 weight percent acetone;

from 3 weight percent to 8 weight percent propylene carbonate;

from 20 weight percent to 30 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

50. The method of claim 49 , wherein R is ethyl.

51. The method of claim 49 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

52. The method of claim 49 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

53. The method of claim 49 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

54. The method of claim 49 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

55. The method of claim 50 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

56. The method of claim 50 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

57. The method of claim 51 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

58. The method of claim 50 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

59. The method of claim 57 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

60. The method of claim 49 , wherein the composition comprises 10 weight percent luliconazole.

61. The method of claim 49 , wherein the composition comprises 12.5 weight percent luliconazole.

62. The method of claim 49 , wherein the composition comprises:

10 weight percent luliconazole;

12 weight percent acetone;

5 weight percent propylene carbonate;

25 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is ethyl; and

4 weight percent benzyl alcohol.

63. The method of claim 62 , wherein the composition further comprises a film forming agent in 1 weight percent.

64. The method of claim 63 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

65. The method of claim 63 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

66. The method of claim 62 , wherein the composition further comprises ethanol in a weight percent that provides a total weight percent of 100 for the composition.

67. The method of claim 63 , wherein the composition further comprises 43 weight percent ethanol.

68. The method of claim 49 , wherein the nail is a toenail.

69. The method of claim 49 , wherein the nail is a fingernail.

70. The method of claim 49 , wherein the method comprises contacting the individual's nail with the composition once daily.

71. The method of claim 49 , wherein the individual is diabetic, is of an advanced age or is immunocompromised.

72. A kit comprising a pharmaceutical composition comprising:

from 5 weight percent to 15 weight percent luliconazole;

from 5 weight percent to 25 weight percent acetone;

from 1 weight percent to 15 weight percent propylene carbonate;

from 15 weight percent to 35 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent;

from 20 weight percent to 60 weight percent of ethanol; and

instructions for treating onychomycosis in a human individual in need thereof.

73. The kit of claim 72 , wherein R is ethyl.

74. The kit of claim 72 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

75. The kit of claim 72 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

76. The kit of claim 72 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

77. The kit of claim 72 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

78. The kit of claim 73 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

79. The kit of claim 73 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

80. The kit of claim 74 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

81. The kit of claim 73 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

82. The kit of claim 80 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

83. The kit of claim 72 , wherein the composition comprises 10 weight percent luliconazole.

84. The kit of claim 72 , wherein the composition comprises 9 weight percent to 12.5 weight percent luliconazole.

85. The kit of claim 72 , wherein the composition comprises:

10 weight percent luliconazole;

12 weight percent acetone;

5 weight percent propylene carbonate;

25 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is ethyl; and

4 weight percent benzyl alcohol.

86. The kit of claim 85 , wherein the composition further comprises a film forming agent in 1 weight percent.

87. The kit of claim 86 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

88. The kit of claim 86 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

89. The kit of claim 85 , wherein the composition further comprises ethanol in a weight percent that provides a total weight percent of 100 for the composition.

90. The kit of claim 86 , wherein the composition further comprises 43 weight percent ethanol.

91. The kit of claim 72 , wherein the composition comprises:

from 9 weight percent to 12.5 weight percent luliconazole;

from 8 weight percent to 15 weight percent acetone;

from 3 weight percent to 8 weight percent propylene carbonate;

from 20 weight percent to 30 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

92. The kit of claim 72 , wherein the onychomycosis is an infection of a toenail.

93. The kit of claim 72 , wherein the onychomycosis is an infection of a fingernail.

94. The kit of claim 72 , wherein the onychomycosis is distal subungual onychomycosis.

95. The kit of claim 72 , wherein the individual is diabetic, is of an advanced age or is immunocompromised.

96. A method of making a composition comprising combining

from 5 weight percent to 15 weight percent luliconazole;

from 5 weight percent to 25 weight percent acetone;

from 1 weight percent to 15 weight percent propylene carbonate;

from 15 weight percent to 35 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

97. The method of claim 96 , wherein R is ethyl.

98. The method of claim 96 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

99. The method of claim 96 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

100. The method of claim 96 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

101. The method of claim 96 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

102. The method of claim 97 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol.

103. The method of claim 97 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

104. The method of claim 98 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

105. The method of claim 97 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

106. The method of claim 104 , wherein the composition comprises from 35 weight percent to 45 weight percent ethanol.

107. The method of claim 96 , wherein the composition comprises 10 weight percent luliconazole.

108. The method of claim 96 , wherein the composition comprises 9 weight percent to 12.5 weight percent luliconazole.

109. The method of claim 96 , wherein the composition comprises:

10 weight percent luliconazole;

12 weight percent acetone;

5 weight percent propylene carbonate;

25 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is ethyl; and

4 weight percent benzyl alcohol.

110. The method of claim 109 , wherein the composition further comprises a film forming agent in 1 weight percent.

111. The method of claim 110 , wherein the film-forming agent is a maleic anhydride/methyl vinyl ether copolymer.

112. The method of claim 110 , wherein the film forming agent is a butyl ester of polyvinylmethylether maleic anhydride copolymer.

113. The method of claim 109 , wherein the composition further comprises ethanol in a weight percent that provides a total weight percent of 100 for the composition.

114. The method of claim 110 , wherein the composition further comprises 43 weight percent ethanol.

115. The method of claim 96 , wherein the composition comprises:

from 9 weight percent to 12.5 weight percent luliconazole;

from 8 weight percent to 15 weight percent acetone;

from 3 weight percent to 8 weight percent propylene carbonate;

from 20 weight percent to 30 weight percent of an ethylene glycol derivative of the formula HOCH 2 CH 2 OCH 2 CH 2 OR where R is an alkyl group having 1-6 carbon atoms;

from 0.01 weight percent to 6 weight percent benzyl alcohol;

from 0.01 weight percent to 4 weight percent of a film forming agent; and

from 20 weight percent to 60 weight percent of ethanol.

116. The method of claim 96 , wherein the method comprises

a) dissolving luliconazole in one or both of benzyl alcohol and the ethylene glycol derivative; and

b) adding ethanol, propylene carbonate, acetone and the film forming agent.

117. The method of claim 96 , wherein the method comprises

a) mixing the film forming agent, ethanol, benzyl alcohol, propylene carbonate and the ethylene glycol in a primary mixing vessel;

b) adding acetone and luliconazole; and

c) mixing the contents until all solids are visually dissolved.

118. The method of claim 117 , wherein acetone and luliconazole are added under amber lighting.

119. The method of claim 96 , wherein the composition is a pharmaceutical composition.

120. The method of claim 109 , wherein the composition is a pharmaceutical composition.

121. The method of claim 1 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

122. The method of claim 9 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

123. The method of claim 25 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

124. The method of claim 33 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

125. The method of claim 49 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

126. The method of claim 57 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

127. The kit of claim 72 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

128. The kit of claim 80 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

129. The method of claim 96 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

130. The method of claim 104 , wherein the composition comprises from 35 weight percent to 55 weight percent ethanol.

131. The method of claim 20 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol and 35 weight percent to 55 weight percent ethanol, and wherein R is ethyl.

132. The method of claim 44 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol and 35 weight percent to 55 weight percent ethanol, and wherein R is ethyl.

133. The kit of claim 91 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol and 35 weight percent to 55 weight percent ethanol, and wherein R is ethyl.

134. The method of claim 115 , wherein the composition comprises 2 weight percent to 4 weight percent benzyl alcohol and 35 weight percent to 55 weight percent ethanol, and wherein R is ethyl.
