Abstract

The invention provides improved nucleic acid ligands that inhibit coagulation and improved modulators of the nucleic acid ligands to provide ideal modulators of coagulation. These improved nucleic acid ligands and modulators are particularly useful for inhibiting coagulation in a host undergoing a therapeutic regime such as surgery or coronary artery bypass.

Claims (17)

1. A first isolated nucleic acid comprising a nucleic acid sequence comprising SEQ ID NO:17.

2. The first nucleic acid of claim 1 , comprising a three dimensional structure comprising a first and a second stem wherein the first stem comprises five nucleotides.

3. The first isolated nucleic acid of claim 1 , wherein the first isolated nucleic acid further comprises a suicide position that becomes single stranded upon binding of an antidote oligonucleotide, wherein the antidote oligonucleotide is complementary to the first isolated nucleic acid and wherein the antidote oligonucleotide hybridizes to the first isolated nucleic acid.

4. An isolated nucleic acid wherein the isolated nucleic acid comprises SEQ ID NO:8.

5. The first isolated nucleic acid of claim 1 , wherein one or more nucleotides are modified, wherein the modification is a 2′-O-methyl modification or a 2′-O-fluoro modification.

6. The first isolated nucleic acid of claim 2 , wherein at least one guanine in the second stem comprises a hydroxyl sugar (2′-OH).

7. The first isolated nucleic acid of claim 2 , wherein at least one cytidine in the second stem comprises a 2′-fluoro modification.

8. The first isolated nucleic acid of claim 1 , wherein the first isolated nucleic acid comprises modifications as presented in the sequence selected from the group consisting of SEQ ID NOs: 34-37, SEQ ID NO:41-49, and SEQ ID NO:50-54.

9. The first isolated nucleic acid of claim 1 , wherein the first isolated nucleic acid is attached to a water soluble polymer.

10. The first isolated nucleic acid of claim 9 , wherein the water soluble polymer is polyethylene glycol.

11. A pharmaceutical composition comprising the isolated nucleic acid of claim 1 in combination with a pharmaceutically acceptable carrier.

12. A method for inhibiting coagulation in a host in need thereof, comprising administering to the host a therapeutically effective amount of a nucleic acid ligand comprising a SEQ ID NO:17.

13. The method according to claim 12 , wherein the nucleic acid ligand comprises a modified nucleotide modified by a 2′-O-methyl modification or a 2′-O-fluoro modification.

14. The method according to claim 12 , wherein the nucleic acid ligand is selected from the group consisting of SEQ ID NO:34-37, SEQ ID NO:41-49, and SEQ ID NO:50-54.

The method according to claim 12 , wherein the host is suffering from or at risk of suffering from a disorder selected from the group consisting of acute myocardial infarction (heart attack), cerebrovascular accidents (stroke), ischemia, angioplasty, CABG (coronary artery bypass grafts), cardiopulmonary bypass, thrombosis in the circuit of cardiac bypass apparatus and in patients undergoing renal dialysis, unstable angina, pulmonary embolism, deep vein thrombosis, arterial thrombosis, and disseminated intravascular coagulation.

16. The method according to claim 12 , wherein the administering is intravenous, subcutaneous, transdermal or parenteral administration.

A method of modulating coagulation in a host in need thereof comprising administering to the host a first isolated nucleic acid sequence comprising SEQ ID NO:17, wherein the first isolated nucleic acid sequence optionally includes one or more nucleotide modifications selected from a 2′-O-methyl modification or a 2′-O-fluoro modification, and administering an effective amount of a second isolated nucleic acid sequence comprising SEQ ID NO:8, wherein the administering of the second nucleic acid sequence is done after the administering the first nucleic acid sequence, and wherein the second nucleic acid sequence reverses the effects of the first nucleic acid sequence.
