Abstract

A new approach in the field of plant gums is described which presents a new solution to the production of hydroxyproline(Hyp)-rich glycoproteins (HRGPs), repetitive proline-rich proteins (RPRPs) and arabino-galactan proteins (AGPs). The expression of synthetic genes designed from repetitive peptide sequences of such glycoproteins, including the peptide sequences of gum arabic glycoprotein (GAGP), is taught in host cells, including plant host cells.

Claims (24)

1. An isolated recombinant polypeptide comprising at least a portion of an arabinogalactan protein comprising one or more repeating modules selected from the group consisting of (Ser-Pro) n , (Ala-Pro) n , and (Thr-Pro) n , and wherein n is from 7 to 50.

2. The polypeptide according to claim 1 , wherein at least one repeating module is (Ser-Pro) n , and wherein n is 7, 20, 30, or 50.

3. The polypeptide according to claim 1 , wherein at least one repeating module is (Ala-Pro) n , and wherein n is 7, 20, 30, or 50.

4. The polypeptide according to claim 1 , wherein at least one repeating module is (Thr-Pro) n , and wherein n is 7, 20, 30, or 50.

5. The polypeptide according to claim 1 , further comprising a non-gum arabic glycoprotein.

6. The polypeptide according to claim 5 , wherein the repeating module is (Ser-Pro) n , and wherein n is 7, 20, 30, or 50.

7. The polypeptide according to claim 1 , wherein the arabinogalactan protein is gum arabic glycoprotein.

8. The polypeptide according to claim 1 , further comprising at least a portion of a hydroxyproline-rich glycoprotein selected from lectins, chitinases, and plant gums.

9. The polypeptide according to claim 8 , wherein the plant gums are selected from gum Karaya, gum Tragacanth, and gum Ghatti.

10. The polypeptide according to claim 1 , further comprising an enzyme.

11. A recombinant polypeptide isolated from a plant, comprising at least a portion of an arabinogalactan protein having at least one O-glycosylated repeating module selected from (Ser-Hyp) n , (Ala-Hyp) n , and (Thr-Hyp) n , and wherein n is from 7 to 50.

12. The polypeptide according to claim 11 , wherein at least one O-glycosylated repeating module is (Ser-Hyp) n , and wherein n is 7, 20, 30, or 50.

13. The polypeptide according to claim 11 , wherein at least one O-glycosylated repeating module is (Ala-Hyp) n , and wherein n is 7, 20, 30, or 50.

14. The polypeptide according to claim 11 , wherein at least one O-glycosylated repeating module is (Thr-Hyp) n , and wherein n is 7, 20, 30, or 50.

15. The polypeptide according to claim 11 , further comprising a non-gum arabic glycoprotein.

16. The polypeptide according to claim 11 , wherein the arabinogalactan protein is gum arabic glycoprotein.

17. The polypeptide according to claim 16 , further comprising at least a portion of a hydroxyproline-rich glycoprotein selected from lectins, chitinases, and plant gums.

18. The polypeptide according to claim 17 , wherein the plant gums are selected from gum Karaya, gum Tragacanth, and gum Ghatti.

19. The polypeptide according to claim 11 , further comprising an enzyme.

20. A recombinant polypeptide isolated from a plant, comprising:

at least a portion of a gum arabic glycoprotein having at least one O-glycosylated repeating module selected from (Ser-Hyp) n , (Ala-Hyp)) n , and (Thr-Hyp) n ; and

least a portion of a hydroxyproline-rich glycoprotein selected from lectins, chitinases, and plant gums;

wherein n is from 7 to 50.

21. The polypeptide according to claim 20 , further comprising an enzyme.
