Abstract

The present invention provides new compositions and methods for preventing and treating pathogen infection. In particular, the present invention provides compounds having an anchoring domain that anchors the compound to the surface of a target cell, and a therapeutic domain that can act extracellularly to prevent infection of a target cell by a pathogen, such as a virus. The present invention also comprises therapeutic compositions having sialidase activity, including protein-based compounds having sialidase catalytic domains. Compounds of the invention can be used for treating or preventing pathogen infection, and for treating and reducing allergic and inflammatory responses. The invention also provides compositions and methods for enhancing transduction of target cells by recombinant viruses. Such compositions and methods can be used in gene therapy.

Claims (33)

1. An isolated nucleic acid molecule encoding a polypeptide comprising amino acids 274 to 666 of SEQ ID NO:12 and the amino acid sequence of SEQ ID NO:7, wherein the polypeptide does not comprise amino acids 1 to 273 of SEQ ID NO:12 or amino acids 667 to 901 of SEQ ID NO:12.

2. An isolated nucleic acid molecule encoding a polypeptide comprising amino acids 274 to 667 of SEQ ID NO:12 and the amino acid sequence of SEQ ID NO:7, wherein the polypeptide does not comprise amino acids 1 to 273 of SEQ ID NO:12 or amino acids 668 to 901 of SEQ ID NO:12.

3. The nucleic acid molecule of claim 1 wherein the polypeptide comprises the amino acid sequence of SEQ ID NO:21.

4. A vector comprising the nucleic acid molecule of claim 1 .

5. The vector of claim 4 wherein the vector is an expression vector.

6. A cell harboring the expression vector of claim 5 .

7. The cell of claim 6 , wherein the cell is a eukaryotic cell.

8. The cell of claim 6 , wherein the cell is a prokaryotic cell.

9. The cell of claim 8 , wherein the prokaryotic cell is E. coli.

10. A method for producing a polypeptide comprising amino acids 274 to 666 of SEQ ID NO:12 and the amino acid sequence of SEQ ID NO:7, wherein the polypeptide does not comprise amino acids 1 to 273 of SEQ ID NO:12 or amino acids 667 to 901 of SEQ ID NO:12, the method comprising

(a) providing the cell of claim 6 ; and

(b) culturing the cell under conditions to express the polypeptide.

11. The method of claim 10 further comprising purifying the polypeptide.

12. A vector comprising the nucleic acid molecule of claim 2 .

13. The vector of claim 12 wherein the vector is an expression vector.

14. A cell harboring the expression vector of claim 13 .

15. The cell of claim 14 , wherein the cell is a eukaryotic cell.

16. The cell of claim 14 , wherein the cell is a prokaryotic cell.

17. The cell of claim 16 , wherein the prokaryotic cell is E. coli.

18. A method for producing a polypeptide comprising amino acids 274 to 667 of SEQ ID NO:12 and the amino acid sequence of SEQ ID NO:7, wherein the polypeptide does not comprise amino acids 1 to 273 of SEQ ID NO:12 or amino acids 668 to 901 of SEQ ID NO:12, the method comprising

(a) providing the cell of claim 14 ; and

(b) culturing the cell under conditions to express the polypeptide.

19. The method of claim 18 further comprising purifying the polypeptide.

20. A vector comprising the nucleic acid molecule of claim 3 .

21. The vector of claim 20 wherein the vector is an expression vector.

22. A cell harboring the expression vector of claim 21 .

23. The cell of claim 22 , wherein the cell is a eukaryotic cell.

24. The cell of claim 22 , wherein the cell is a prokaryotic cell.

25. The cell of claim 24 , wherein the prokaryotic cell is E. coli.

26. A method for producing a polypeptide comprising amino acids 274 to 667 of SEQ ID NO:12 and the amino acid sequence of SEQ ID NO:7, wherein the polypeptide does not comprise amino acids 1 to 273 of SEQ ID NO:12 or amino acids 668 to 901 of SEQ ID NO:12, the method comprising

(a) providing the cell of claim 22 ; and

(b) culturing the cell under conditions to express the polypeptide.

27. The method of claim 26 further comprising purifying the polypeptide.
