Concentrated aqueous silk fibroin solution and use thereof
The present invention provides for concentrated aqueous silk fibroin solutions and an all-aqueous mode for preparation of concentrated aqueous fibroin solutions that avoids the use of organic solvents, direct additives, or harsh chemicals. The invention further provides for the use of these solutions in production of materials, e.g., fibers, films, foams, meshes, scaffolds and hydrogels.
1. A method comprising:
providing a silk fibroin material; and
contacting the silk fibroin material with water vapor in the absence of alcohol, wherein the silk fibroin material comprises an agent selected from the group consisting of therapeutic agents, antiinfectives, anti-rejection agents, analgesics and analgesic combinations, anti-inflammatory agents, hormones, growth factors, polysaccharides, glycoproteins, lipoproteins, pharmacological materials, vitamins, sedatives, steroids, hypnotics, antibiotics, chemotherapeutic agents, prostaglandins, radiopharmaceuticals, proteins, peptides, small molecules, nucleic acids, nucleotides, carbohydrates, simple sugars, cells, genes, anti-thrombotics, anti-metabolics, growth factor inhibitor, growth promoters, anticoagulants, antimitotics, fibrinolytics, anti-inflammatory steroids, and monoclonal antibodies, and any combinations thereof.
2. The method of claim 1 , further comprising stretching the silk fibroin material following the contacting.
3. The method of claim 2 , wherein the stretching comprises mono-axial stretching, bi-axial stretching, or a combination thereof.
4. The method of claim 1 , wherein the silk fibroin material comprises a film, a fiber, a foam, a hydrogel, a mesh, a scaffold, a micropatterned foam, a porous scaffold, a coating, a yarn, a fabric, or any combination thereof.
5. The method of claim 1 , further comprising forming the silk fibroin material essentially free of an organic solvent.
