Abstract

Disclosed are mutant CDK inhibitor (CKI) polypeptides having dominant negative antagonist activity against wild-type CKI proteins, as well as related compositions, including nucleic acids and vectors encoding the mutant CKI polypeptides and transformed host cells and transgenic plants comprising such nucleic acids and vectors. Also disclosed are related methods for using the mutant proteins to modulate cell division in cells, particularly plant cells.

Claims (19)

1. A transgenic plant comprising a transgene encoding a mutant Brassica napus Kinase Inhibitor Protein (KIP) related protein (BnKRP) comprising an amino acid sequence selected from the group consisting of SEQ ID NO: 93, SEQ ID NO: 94, SEQ ID NO: 97 and SEQ ID NO: 101.

2. The transgenic plant of claim 1 , wherein the BnKRP comprises the amino acid sequence of SEQ ID NO: 93.

3. The transgenic plant of claim 1 , wherein the BnKRP comprises the amino acid sequence of SEQ ID NO: 94.

4. The transgenic plant of claim 1 , wherein the BnKRP comprises the amino acid sequence of SEQ ID NO: 97.

5. The transgenic plant of claim 1 , wherein the BnKRP comprises the amino acid sequence of SEQ ID NO: 101.

6. The transgenic plant of claim 1 , wherein the transgenic plant is selected from the group consisting of Brassica napus, Arabidopsis thaliana, Glycine max , maize, wheat, rice, alfalfa, cotton, poplar and camelina.

7. A plant part of the transgenic plant of claim 1 .

A seed produced by the transgenic plant of claim 1 , wherein the seed comprises a nucleic acid sequence encoding a mutant Brassica napus Kinase Inhibitor Protein (KIP) related protein (BnKRP) comprising an amino acid sequence selected from the group consisting of SEQ ID NO: 93, SEQ ID NO: 94, SEQ ID NO: 97 and SEQ ID NO: 101.

A plant cell of the transgenic plant of claim 1 , wherein the plant cell comprises a nucleic acid sequence encoding a mutant Brassica napus Kinase Inhibitor Protein (KIP) related protein (BnKRP) comprising an amino acid sequence selected from the group consisting of SEQ ID NO: 93, SEQ ID NO: 94, SEQ ID NO: 97 and SEQ ID NO: 101.

10. A tissue culture of cells of the transgenic plant of claim 1 .

11. A recombinant nucleic acid comprising a nucleic acid sequence encoding a mutant Brassica napus Kinase Inhibitor Protein (KIP) related protein (BnKRP) comprising an amino acid sequence selected from the group consisting of SEQ ID NO: 93, SEQ ID NO: 94, SEQ ID NO: 97 and SEQ ID NO: 101.

12. An expression vector comprising the recombinant nucleic acid of claim 11 .

13. The expression vector of claim 12 further comprising a nucleic acid sequence encoding a promoter operably linked to the nucleic acid sequence encoding the BnKRP.

14. The expression vector of claim 13 , wherein the promoter is a constitutive promoter or an embryo specific promoter.

15. The expression vector of claim 14 , wherein the constitutive promoter is a CaMV 35S promoter and wherein the embryo specific promoter is a LFAH12 promoter.

16. A cell comprising the recombinant nucleic acid of claim 11 .

17. The cell of claim 16 wherein the cell is a bacterial cell, an insect cell or a plant cell.

18. The cell of claim 17 , wherein the cell is a plant cell.

19. The plant cell of claim 18 , wherein the plant is selected from the group consisting of Brassica napus, Arabidopsis thaliana, Glycine max , maize, wheat, rice, alfalfa, cotton, poplar and camelina.
