Abstract

An embodiment of the invention is directed to a microfabricated, silicon-based, Convection Enhanced Delivery (CED) device. The device comprises a silicon shank portion, at least one individual parylene channel disposed along at least a part of an entire length of the shank, wherein the channel has one or more dimensioned fluid exit ports disposed at one or more respective locations of the channel and a fluid (drug) input opening. The fluid input opening may be configured or adapted to be connected to a fluid reservoir and/or a pump and/or a meter and/or a valve or other suitable control device(s) or apparatus that supplies and/or delivers fluid (eg, a drug) to the microfabricated device. The device may have multiple channels disposed side by side or in different surfaces of the device.

Claims (45)

1. An implantable convection-enhanced-delivery (CED) device suitable for delivering a fluid directly into extracellular, interstitial tissue, comprising:

a layered composition shank portion comprising a silicon substrate, a polyimide layer, and a parylene layer, including at least one enclosed, parylene composition channel disposed in a layered surface along at least a portion of a length of the shank portion,

wherein the at least one parylene composition channel has a fluid inlet port adjacent a proximal end thereof that is adjacent a proximal end of the device and at least one fluid outlet port in a surface of the at least one enclosed, parylene composition channel adjacent a distal end thereof,

further wherein the CED device is adapted for delivering a fluid directly into extracellular, interstitial tissue regions.

2. The CED device of claim 1 , wherein the distal end of the shank portion is pointed so as to enable insertion into an interstitial region of a target tissue.

3. The CED device of claim 1 , wherein the at least one enclosed, parylene composition channel is disposed in a top surface of the shank portion.

4. The CED device of claim 1 , wherein the shank portion has a cross sectional, diametrical dimension equal to or less than 0.2 mm.

5. The CED device of claim 1 , wherein the at least one enclosed, parylene composition channel has an albumin coating.

6. The CED device of claim 1 , wherein the shank portion has a length L M of between two millimeters to 50 millimeters and a cross sectional area of between 1,000 microns squared and 20,000 microns squared.

7. The CED device of claim 1 , wherein the at least one enclosed, parylene composition channel has a height h C of between one micron to 50 microns.

8. The CED device of claim 1 , wherein the at least one enclosed, parylene composition channel has a width w C of between 10 microns to 100 microns.

9. The CED device of claim 1 , wherein the at least one enclosed, parylene composition channel has a plurality of outlet ports disposed in one or more surfaces of the layered composition shank portion.

10. The CM device of claim 1 , further comprising a handle portion connected to the shank portion.

11. The CED device of claim 1 , further comprising a fluid supply portion having a channel intermediate an outlet port thereof connected to the fluid inlet port, and an inlet port thereof connectable to a fluid supply.

12. The CED device of claim 11 , further comprising a controllable fluid supply generator connected to the fluid supply.

13. The CED device of claim 12 , wherein the controllable fluid supply generator is at least one of a programmable pressure generator and a programmable flow-rate generator.

14. The CED device of claim 12 , wherein the controllable fluid supply generator is a pump.

15. The CED device of claim 1 , further comprising an electrode embedded in the device.

16. The CED device of claim 15 , wherein the electrode is one of a stimulating and a recording electrode.

17. The CED device of claim 1 , further comprising at least one of a valve and a flow meter connected to the device.

18. The CED device of claim 1 , further comprising a sensor integrated with the device.

19. The CED device of claim 18 , wherein the sensor is an interrogatable sensor.

20. The CED device of claim 18 , wherein the sensor is a glutamate sensor.

21. The CED device of claim 18 , wherein the sensor is at least one of a pressure sensor, a pH sensor, a temperature sensor, an ion concentration sensor, a carbon dioxide sensor, an oxygen sensor, and a lactate sensor.

22. An implantable convection-enhanced-delivery (CED) device, comprising:

a body having a shank portion at a distal end thereof and a fluid inlet portion that is contiguous with the shank portion;

at least one enclosed, parylene composition channel disposed in at least a portion of a length of the body,

wherein the at least one enclosed, parylene composition channel has an inlet port adjacent a proximal end thereof that is adjacent a proximal end of the fluid inlet portion of the body and a distal end disposed in the shank portion, further wherein the at least one enclosed, parylene composition channel has at least one outlet port along a length of the shank portion,

further wherein the CED device is adapted for delivering a fluid directly into extracellular, interstitial tissue regions.

23. The CED device of claim 22 , wherein a distal end of the shank portion has an anatomically-compatible shape such that the shank portion is adapted for insertion into an interstitial region of a target tissue.

24. The CED device of claim 22 , wherein the body is insufficiently rigid to support insertion of the shank portion into an interstitial region of a target tissue.

The CED device of claim 24 , further comprising an insertion support scaffold that is attachable to the device, said support scaffold having a region configured to retain at least the shank portion and a distal end having an anatomically-compatible shape adapted for insertion into an interstitial region of a target tissue.

26. The CED device of claim 25 , wherein the insertion support scaffold is a bio-degradable thermoplastic polymer.

27. The CED device of claim 26 , wherein the bio-degradable thermoplastic polymer is one of a thermoplastic polyester and a thermoplastic polycarbonate.

28. The CED device of claim 26 , wherein the insertion support scaffold is made of poly(lactic-co-glycolic acid) (PLGA).

29. The CED device of claim 28 , wherein the controllable fluid supply generator is a pump.

30. The CED device of claim 25 , wherein the insertion support scaffold contains a quantity of a drug.

31. The CED device of claim 30 , wherein a surface of the insertion support scaffold is coated with the drug.

32. The CED device of claim 22 , further comprising a fluid supply connectable to the proximal end of the fluid inlet portion.

33. The CED device of claim 32 , further comprising a controllable fluid supply generator connected to the fluid supply.

34. The CED device of claim 33 , wherein the controllable fluid supply generator is at least one of a programmable pressure generator and a programmable flow-rate generator.

The CED device of claim 22 , wherein the at least one enclosed, parylene composition channel is adapted to conduct a drug containing fluid from the fluid inlet port under a first positive pressure P 1 and release the drug containing fluid through the respective at least one fluid outlet port under an infusion pressure P 0 into an interstitial region of a target tissue, where ΔP=P 1 −P 0 is the pressure drop between the at least one fluid outlet port and the fluid inlet port.

36. The CED device of claim 35 , wherein 0≦ΔP≦310 kPa.

The CED device of claim 22 , wherein the at least one enclosed, parylene composition channel is adapted to conduct a drug containing fluid from the fluid inlet port under a first positive pressure P 1 and release the drug containing fluid through the respective at least one fluid outlet port under an infusion pressure P 0 into an interstitial region of a target tissue, where ΔP=P 1 −P 0 is the pressure drop between the at least one fluid outlet port and the fluid inlet port.

38. The CED device of claim 37 , wherein 0≦ΔP≦310 kPa.
