Abstract

The present invention relates to optimized Fc variants, methods for their generation, and antibodies and Fc fusions comprising optimized Fc variants.

Claims (11)

1. A variant Fc region of a human parent IgG1 Fc region, wherein said variant Fc region comprises an amino acid substitution in the human parent Fc region at position 264, wherein said variant Fc region exhibits altered binding affinity to an Fcγreceptor (FcγR) as compared to a parent Fc region and wherein numbering is according to the EU index.

2. The variant Fc region of claim 1 , wherein said amino acid substitution is 264I.

3. The variant Fc region of claim 1 , wherein said amino acid substitution is 264T.

4. The variant Fc region of claim 1 , wherein said amino acid substitution is 264L.

5. The variant Fc region of claim 1 , where said Fc region comprises an engineered glycoform.

6. A pharmaceutical composition comprising the variant Fc region of claim 1 , further comprising a pharmaceutically acceptable carrier.

7. The variant Fc region of claim 1 , wherein said amino acid substitution is 264F.

8. The variant Fc region of claim 1 , wherein said amino acid substitution is 264M.

9. The variant Fc region of claim 1 , wherein said amino acid substitution is 264Y.

10. The variant Fc region of claim 1 , wherein said amino acid substitution is 264R.

11. The variant Fc region of claim 1 , wherein said amino acid substitution is 264A.
