Abstract

This document provides methods and materials related to treating glaucoma, ocular hypertension, cardiovascular diseases, and renal diseases. For example, this document provides isolated nucleic acid molecules and viral vectors (e.g., lentiviral vectors) containing isolated nucleic acid molecules. Methods for reducing intraocular pressure as well as symptoms and progression of cardiovascular and renal diseases also are provided.

Claims (13)

1. A method for treating a mammal having glaucoma or elevated intraocular pressure, said method comprising administering nucleic acid encoding a polypeptide having cyclooxygenase-2 activity and nucleic acid encoding a polypeptide having prostaglandin F 2α receptor activity to an eye of said mammal under conditions effective to reduce intraocular pressure of said eye by at least 10 percent.

2. The method of claim 1 , wherein said mammal is a human.

3. The method of claim 1 , wherein said administering step comprises contacting said eye with a solution containing said nucleic acid encoding said polypeptide having cyclooxygenase-2 activity and said nucleic acid encoding said polypeptide having prostaglandin F 2α receptor activity.

4. The method of claim 1 , wherein said method is effective to reduce said intraocular pressure by at least 20 percent.

5. The method of claim 1 , wherein said method is effective to reduce said intraocular pressure by at least 30 percent.

6. The method of claim 1 , wherein said nucleic acid encoding said polypeptide having cyclooxygenase-2 activity is administered using a viral vector.

7. The method of claim 6 , wherein said viral vector is a lentiviral vector.

8. The method of claim 1 , wherein said nucleic acid encoding said polypeptide having prostaglandin F 2α receptor activity is administered using a viral vector.

9. The method of claim 8 , wherein said viral vector is a lentiviral vector.

10. The method of claim 1 , wherein said nucleic acid encoding said polypeptide having cyclooxygenase-2 activity and said nucleic acid encoding said polypeptide having prostaglandin F2α receptor activity are administered using a viral vector.

11. The method of claim 10 , wherein said viral vector is a lentiviral vector.

12. The method of claim 1 , wherein said polypeptide having cyclooxygenase-2 activity is a human cyclooxygenase-2 polypeptide, and wherein said nucleic acid encoding said polypeptide having cyclooxygenase-2 activity is codon-optimized for use in mammalian cells.

13. The method of claim 1 , wherein said polypeptide having prostaglandin F 2α receptor activity is a human prostaglandin F 2α receptor polypeptide, and wherein said nucleic acid encoding said polypeptide having prostaglandin F 2α receptor activity is codon-optimized for use in mammalian cells.
