Abstract

Disclosed are compounds, pharmaceutical compositions containing those compounds, and uses of the compounds and compositions as modulators of CK1, CK1γ1, CK1γ2, CK1γ3, CK2, Pim 1, Pim2, Pim3, the TGFβ pathway, the Wnt pathway, the JAK/STAT pathway, the AKT pathway, and/or the mTOR pathway. Uses are also disclosed for the treatment or prevention of a range of therapeutic indications due at least in part to aberrant physiological activity of CK1, CK1γ1, CK1γ2, CK1γ3, CK2, Pim 1, Pim2, Pim3, the TGFβ pathway, the Wnt pathway, the JAK/STAT pathway, the AKT pathway, and/or the mTOR pathway.

Claims (80)

1. A compound of formula 1 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence:

X is —N(R 7 ) 2 , —N(R 7 )(R 2 ), or —N(H)—R 3 -R 6 ;

R′ is H, methyl, (C 2 -C 4 )alkyl, or benzyl;

R 2 is H, —CH 2 O(P═O)(OR 8 ) 2 , —(C═O)OCHR 8 O(C═O)CH 3 , or —(C═O)OCH 2 O(P═O)(OH) 2 ;

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R 5 is selected from the group consisting of 1,4-cyclohexanediyl, 1,4-phenylene, 1,4-cycloheptanediyl, 1,4-cyclooctanediyl, and 1,5-cyclooctanediyl;

R 6 is selected from the group consisting of alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl;

R 7 is selected from the group consisting of H, —(C(R) 2 ) n R, alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl, or

two instances of R 7 and the nitrogen to which they are bonded taken together represent a nitrogen-containing heterocyclyl, optionally containing one additional heteroatom in the ring, wherein said additional heteroatom is selected from the group consisting of —O—, —N(R)—, and —S—;

R 8 is H, or alkyl;

R is H or (C 1 -C 4 )alkyl; and

n is 1, 2 or 3.

2. The compound of claim 1 , wherein R′ is H.

3. The compound of claim 1 , wherein R′ is methyl.

4. The compound of claim 1 , wherein R 2 is H.

5. The compound of claim 1 , wherein R 5 is 1,4-cyclohexanediyl.

6. The compound of claim 1 , wherein R 5 is 1,4-phenylene.

7. The compound of claim 1 , wherein R 6 is selected from the group consisting of alkyl, aryl, and heteroaryl.

8. The compound of claim 1 , wherein R 6 is selected from the group consisting of phenyl, biphenyl, pyridyl, pyrimidyl, naphthyl, quinolinyl, furanyl, and thienyl.

9. A compound of formula 2 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence

X is —N(R 7 ) 2 , —N(R 7 )(R 2 ), or —N(H)—R 3 -R 4 ;

R′ is H, methyl, (C 2 -C 4 )alkyl, or benzyl;

R 1 is selected from the group consisting of 1,4-cyclohexanediyl and 1,4-phenylene;

R 2 is H, —CH 2 O(P═O)(OR 8 ) 2 , —(C═O)OCHR 8 O(C═O)CH 3 , or —(C═O)OCH 2 O(P═O)(OH) 2 ;

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R 4 is selected from the group consisting of aryl and heteroaryl, either of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of halide, alkyl, perfluoroalkyl, aryl, heteroaryl, and heterocyclyl; wherein the optional aryl or heteroaryl substituent is itself optionally substituted with perfluoroalkyl or dioxolane;

R 7 is selected from the group consisting of H, —(C(R) 2 ) n R, alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl, or

two instances of R 7 and the nitrogen to which they are bonded taken together represent a nitrogen-containing heterocyclyl, optionally containing one additional heteroatom in the ring, wherein said additional heteroatom is selected from the group consisting of —O—, —N(R)—, and —S—;

R 8 is H, or alkyl;

R is H or (C 1 -C 4 )alkyl; and

n is 1, 2 or 3.

10. The compound of claim 9 , wherein R′ is H.

11. The compound of claim 9 , wherein R′ is methyl.

12. The compound of claim 9 , wherein R 1 is 1,4-cyclohexanediyl.

13. The compound of claim 9 , wherein R 1 is 1,4-phenylene.

14. The compound of claim 9 , wherein R 4 is selected from the group consisting of phenyl, pyridyl, naphthyl, quinolinyl, furanyl, and thienyl.

15. A compound of formula 3 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence

R 2 is H, —CH 2 O(P═O)(OR 8 ) 2 , —(C═O)OCHR 8 O(C═O)CH 3 , or —(C═O)OCH 2 O(P═O)(OH) 2 ;

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R is H or (C 1 -C 4 )alkyl;

n is 1, 2 or 3;

R 6 is selected from the group consisting of alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl; and

R 8 is H, or alkyl.

16. The compound of claim 15 , wherein R 2 is H.

17. The compound of claim 15 , wherein R 3 is —CH 2 —.

18. The compound of claim 15 , wherein R 6 is selected from the group consisting of alkyl, aryl, and heteroaryl.

19. The compound of claim 18 , wherein R 6 is selected from the group consisting of phenyl, biphenyl, pyridyl, pyrimidyl, naphthyl, quinolinyl, furanyl, and thienyl.

20. A compound of formula 4 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R is H or (C 1 -C 4 )alkyl;

n is 1, 2 or 3; and

R 4 is selected from the group consisting of aryl and heteroaryl, either of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of halide, alkyl, perfluoroalkyl, aryl, heteroaryl, and heterocyclyl; wherein the optional aryl or heteroaryl substituent is itself optionally substituted with perfluoroalkyl or dioxolane.

21. The compound of claim 20 , wherein R 4 is selected from the group consisting of phenyl, pyridyl, naphthyl, quinolinyl, furanyl, and thienyl.

22. A compound of formula 5 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence

R′ is H, methyl, (C 2 -C 4 )alkyl, or benzyl;

R 2 is H, —CH 2 O(P═O)(OR 8 ) 2 , —(C═O)OCHR 8 O(C═O)CH 3 , or —(C═O)OCH 2 O(P═O)(OH) 2 ;

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R is H or (C 1 -C 4 )alkyl;

n is 1, 2 or 3;

R 6 is selected from the group consisting of alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl; and

R 8 is H, or alkyl.

23. The compound of claim 22 , wherein R′ is H.

24. The compound of claim 22 , wherein R′ is methyl.

25. The compound of claim 22 , wherein R 2 is H.

26. A compound of formula 6 or a pharmaceutically acceptable salt thereof:

wherein independently for each occurrence

R′ is H, methyl, (C 2 -C 4 )alkyl, or benzyl;

R 2 is H, —CH 2 O(P═O)(OR 8 ) 2 , —(C═O)OCHR 8 O(C═O)CH 3 , or —(C═O)OCH 2 O(P═O)(OH) 2 ;

R 3 is —C(═NR)— or —(C(R) 2 ) n —;

R is H or (C 1 -C 4 )alkyl;

n is 1, 2 or 3;

R 6 is selected from the group consisting of alkyl, alkylaryl, aryl, alkylheteroaryl, and heteroaryl, any of which is optionally mono or di-substituted, and the substituents, if present, are independently selected from the group consisting of alkyl, alkylaryl, aralkyl, aryl, heteroaryl, alkoxy, hydroxy, perfluoroalkyl, trifluoromethoxy, and halide, wherein the optional aryl or heteroaryl substituent may be optionally substituted with alkyl, halide, alkoxy, perfluoroalkyl, or dioxolanyl; and

R 8 is H, or alkyl.

27. The compound of claim 26 , wherein R′ is H.

28. A pharmaceutical composition, comprising a compound of claim 1 and a pharmaceutically acceptable excipient.
