Abstract

The present invention relates to a gene that encodes a hyperactive reverse transcriptase having DNA polymerase activity and substantially reduced RNase H activity, vectors containing the gene and host cells transformed with the invention. The present invention also includes a method of producing the hyperactive reverse transcriptase, producing cDNA from mRNA using the reverse transcriptase of the invention, kits and assay templates made using the hyperactive reverse transcriptase.

Claims (9)

1. An isolated and purified nucleic acid encoding a mutant reverse transcriptase comprising SEQ ID NO:2.

2. The nucleic acid of claim 1 , wherein the mutant reverse transcriptase comprises enhanced DNA polymerase activity.

3. The nucleic acid of claim 1 , wherein the mutant reverse transcriptase comprises substantially reduced RNase H activity.

4. The nucleic acid of claim 1 , wherein the nucleic acid comprises SEQ ID NO.: 1.

5. The nucleic acid of claim 1 , wherein the nucleic acid comprises a nucleic acid segment encoding a leader sequence.

6. The nucleic acid of claim 1 , wherein the nucleic acid comprises a nucleic acid segment encoding a protein segment other than the mutant reverse transcriptase.

7. A process for making an isolated mutant reverse transcriptase comprising the steps of:

transforming a host cell with an isolated nucleic acid that encodes a mutant reverse transcriptase comprising SEQ ID NO:2; and

culturing the host cell under conditions such that the mutant reverse transcriptase is produced.
