Abstract

Protein kinase inhibitors are disclosed having utility in the treatment of protein kinase-mediated diseases and conditions, such as cancer. The compounds of this invention have the following structure: including steroisomers, prodrugs and pharmaceutically acceptable salts thereof, wherein A is a ring moiety selected from: and wherein R1, R2, R3, X, Z, L1, Cycl1, L2 and Cycl2 are as defined herein. Also disclosed are compositions containing a compound of this invention, as well as methods relating to the use thereof.

Claims (15)

1. A compound having the following structure (III-1-3):

or a stereoisomer, prodrug, tautomer or pharmaceutically acceptable salt thereof.

2. A composition comprising the compound of claim 1 in combination with a pharmaceutically acceptable excipient or carrier.

3. A composition comprising the compound of claim 1 or a stereoisomer, prodrug, tautomer or pharmaceutically acceptable salt thereof, one or more other chemotherapeutic agents and a pharmaceutically acceptable excipient or carrier.

4. The composition of claim 3 , wherein the chemotherapeutic agent is selected from the group consisting of mitotic inhibitors, alkylating agents, anti-metabolites, cell cycle inhibitors, enzymes, topoisomerase inhibitors, biological response modifiers, anti-hormones, antiangiogenic agents, anti-androgens, platinum coordination complexes, substituted ureas, methylhydrazine derivatives, adrenocortical suppressants, hormone and hormone antagonists, progestins, estrogens, antiestrogens, androgens, and aromatase inhibitors.

5. The composition of claim 3 , wherein the chemotherapeutic agent is a DNA-damaging agent.

6. A process for the manufacture of the compound of claim 1 , the process comprising reacting a compound of formula (XI):

with an appropriately activated thioamide.

7. The process of claim 6 , further comprising preparing the compound of formula (XI) by reaction of a compound of formula (X):

with piperazine.

8. The composition of claim 3 , wherein the one or more other chemotherapeutic agents are selected from signal transduction inhibitors.

9. The composition of claim 3 , wherein the one or more other chemotherapeutic agents are selected from epidermal growth factor receptor (EGFR) inhibitors.

10. The composition of claim 3 , wherein the one or more other chemotherapeutic agents are selected from Tarceva® (erlotinib) and Gleevec® (imatinib mesylate).

11. The composition of claim 3 , wherein the one or more other chemotherapeutic agents are selected from platinum coordination complexes, alkylating agents, epipodophyllotoxins and topoisomerase inhibitors.

12. The composition of claim 3 , wherein the one or more other chemotherapeutic agents are selected from cisplatin, fluorouracil (5-FU), etoposide and irinotecan.
