Abstract

The invention generally relates to systems and methods for mass spectrometry analysis of samples. In certain embodiments, the invention provides a mass spectrometry probe including at least one porous material connected to a high voltage source, in which the porous material is discrete from a flow of solvent.

Claims (30)

1. A method for analyzing a protein or peptide in a sample, the method comprising:

contacting a sample comprising a protein or peptide to a probe comprising a porous material that tapers to a tip, wherein the probe operates without pneumatic assistance;

applying a voltage to the porous material such that an electric field is generated at the tip of the material so as to generate ions of the protein or peptide in the sample that are expelled from the tip; and

analyzing the expelled ions, thereby analyzing the protein or peptide in the sample.

2. The method according to claim 1 , wherein the porous material is kept separate from a flow of solvent.

3. The method according to claim 2 , further comprising applying a solvent to the porous material.

4. The method according to claim 3 , wherein the solvent assists transport of the sample through the porous material.

5. The method according to claim 3 , wherein the solvent contains an internal standard.

6. The method according to claim 3 , wherein the solvent minimizes matrix effects.

7. The method according to claim 1 , wherein the porous material is filter paper.

8. The method according to claim 1 , wherein analyzing comprises providing a mass analyzer to generate a mass spectrum of the protein or peptide.

9. The method according to claim 8 , wherein the mass analyzer is enclosed within a miniature mass spectrometer.

10. The method according to claim 8 , wherein the mass analyzer is selected from the group consisting of: a quadrupole ion trap, a rectilinear ion trap, a cylindrical ion trap, a ion cyclotron resonance trap, and an orbitrap.

11. A method for analyzing a protein or peptide, the method comprising:

contacting a body fluid sample comprising a protein or peptide to a probe comprising a porous material that tapers to a tip, wherein the probe operates without pneumatic assistance;

applying a voltage to the porous material such that an electric field is generated at the tip of the material so as to generate ions of the protein or peptide in the body fluid sample that are expelled from the tip of the porous material; and

analyzing the expelled ions, thereby analyzing the protein or peptide in the body fluid sample.

12. The method according to claim 11 , wherein the body fluid is blood.

13. The method according to claim 11 , wherein the body fluid is urine.

14. The method according to claim 11 , wherein the body fluid is saliva.

15. The method according to claim 11 , wherein the porous material is kept separate from a flow of solvent.

16. The method according to claim 15 , further comprising applying a solvent to the porous material.

17. The method according to claim 11 , wherein analyzing comprises providing a mass analyzer to generate a mass spectrum of the protein.

18. A method for analyzing a protein or peptide in a sample, the method comprising:

obtaining a sample comprising a protein or peptide;

contacting the sample comprising the protein or peptide to a probe comprising a porous material without any sample preparation steps, wherein the probe operates without pneumatic assistance;

applying a voltage to the porous material such that an electric field is generated at the tip of the material so as to generate ions of the protein or peptide, wherein the ions generated are expelled from the porous material; and

analyzing the expelled ions, thereby analyzing the protein or peptide in the sample.

19. The method according to claim 18 , wherein the sample is a body fluid.

20. The method according to claim 19 , wherein the body fluid is blood.
