Enhancing a population of insulin releasing cells using GFR α-1 agonists
Provided herein are pharmaceutical and transplant compositions and methods related to the treatment and prevention of diabetes. More specifically, the compositions and methods are related to activation of glial derived neurotrophic factor (GDNF) receptors or overexpression of the GFR-α1/c-Ret receptor complex in insulin secreting cells so as to promote cell survival and proliferation.
1. A method of enhancing a population of pancreatic islet cells for transplantation comprising culturing the population of pancreatic islet cells with an effective amount of a GDNF polypeptide having SEQ ID NO: 9 or variants with at least 80% sequence identity to SEQ ID NO: 9.
2. The method of claim 1 , wherein the GDNF in the culture is at a concentration of at least about 10 ng/ml.
3. The method of claim 1 , wherein the GDNF concentration in the culture is at least about 100 ng/ml.
4. The method of claim 1 , wherein the GDNF concentration in the culture is at least about 200 ng/ml.
5. The method of claim 1 , wherein the pancreatic islet cells in the population are enhanced by increasing the number of viable islet cells as compared to a control.
6. The method of claim 1 , wherein the pancreatic islet cells in the population are enhanced by reducing apoptosis as compared to a control.
7. The method of claim 1 , wherein the transplant population of pancreatic islet cells releases more insulin as compared to a control.
8. A method of preparing a pancreatic islet cell population for transplantation comprising the steps of
administering an effective amount of a GDNF polypeptide having SEQ ID NO: 9 or variants with at least 80% sequence identity to SEQ ID NO: 9 to a transplant donor and
isolating a pancreatic islet cell population from the donor.
9. The method of claim 8 , further comprising culturing the isolated islet cell population with an effective amount of a GDNF polypeptide having SEQ ID NO: 9 or variants with at least 80% sequence identity to SEQ ID NO: 9.
10. The method of claim 8 , further comprising culturing the isolated islet cell population with glial cells.
