Abstract

Pharmaceutical formulations exhibit a desirably low Cmax, among other properties, that contain particles of amorphous bardoxolone methyl, either in pure form or in the form of a solid dispersion, admixed with particles of a hydrophilic binding agent. Such formulations possess the advantage of higher oral bioavailability, relative to formulations based on the crystalline form of bardoxolone methyl.

Claims (16)

1. A solid dosage form comprising (A) particles consisting of amorphous bardoxolone methyl admixed with (B) particles consisting of one or more hydrophilic binders, wherein said particles (A) constitute a therapeutically effective amount of bardoxolone methyl.

2. A solid dosage form comprising (A) a solid dispersion of amorphous bardoxolone methyl in a glassy matrix admixed with (B) particles consisting of one or more hydrophilic binders.

3. The solid dosage form of claim 1 or 2 , wherein the hydrophilic binder is a cellulose-based excipient.

4. The solid dosage form of claim 2 , wherein said particles (A) are the product of a process that comprises spray drying a mixture of bardoxolone methyl and a methacrylic acid copolymer.

5. The solid dosage form of claim 4 , wherein said process comprises spray drying a 4:6 mixture of bardoxolone methyl and a methacrylic acid copolymer.

6. The solid dosage form of claim 3 , wherein the cellulose-based excipient is selected from the group consisting of C 3 -C 10 alkyl hydroxymethyl cellulose, aryl hydroxymethyl cellulose, and substituted aryl hydroxymethyl cellulose.

The solid dosage form of claim 6 , wherein the cellulose-based excipient is a C 3 -C 10 alkyl hydroxymethyl cellulose selected from the group consisting of methyl cellulose, ethyl cellulose, propyl cellulose, hydroxymethyl cellulose, hydroxypropyl cellulose, hydroxypropyl methyl cellulose, and cellulose acetate.

8. The solid dosage form of claim 1 , wherein the hydrophilic binder is a naturally occurring carbohydrate polymer.

9. The solid dosage form of claim 1 , wherein the hydrophilic binder is an anionic polymer.

10. The solid dosage form of claim 1 , wherein the proportion of the hydrophilic binder is between about 1% and about 40% (w/w) of the total formulation.

11. The solid dosage form of claim 10 , wherein the proportion of the hydrophilic binder is between about 2% and about 20% (w/w) of the total formulation.

12. The solid dosage form of claim 10 , wherein the proportion of the hydrophilic binder is between about 4% and about 10% (w/w) of the total formulation.

13. The solid dosage form of claim 10 , wherein the proportion of the hydrophilic binder is between about 5% and about 7.5% (w/w) of the total formulation.

14. The solid dosage form of claim 10 , wherein the proportion of the hydrophilic binder is between about 7% and about 7.5% (w/w) of the total formulation.

15. The solid dosage form of claim 10 , wherein the proportion of the hydrophilic binder is about 7% (w/w) of the total formulation.

16. The solid dosage form of any of claims 10 - 15 , wherein the hydrophilic binding agent is hydroxypropyl methylcellulose.
