Abstract

Antibodies that specifically bind to toxins of C. difficile , antigen binding portions thereof, and methods of making and using the antibodies and antigen binding portions thereof are provided herein.

Claims (25)

1. An isolated nucleic acid comprising a nucleotide sequence encoding a polypeptide, wherein the polypeptide comprises SEQ ID NO:1, SEQ ID NO:2, SEQ ID NO:3, SEQ ID NO:4, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:54, or SEQ ID NO:58.

2. An isolated nucleic acid comprising a nucleotide sequence encoding an antibody variable region, wherein the antibody variable region is at least 90% identical to SEQ ID NO:1, at least 90% identical to SEQ ID NO:2, at least 96% identical to SEQ ID NO:3, at least 99% identical to SEQ ID NO:5, at least 99% identical to SEQ ID NO:6, or at least 95% identical to SEQ ID NO:54.

3. The isolated nucleic acid of claim 2 , wherein the antibody variable region is at least 90% identical to SEQ ID NO:1.

4. The isolated nucleic acid of claim 2 , wherein the antibody variable region is at least 90% identical to SEQ ID NO:2.

5. The isolated nucleic acid of claim 2 , wherein the antibody variable region is at least 96% identical to SEQ ID NO:3.

6. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises SEQ ID NO:4.

7. The isolated nucleic acid of claim 2 , wherein the antibody variable region at least 99% identical to SEQ ID NO:5.

8. The isolated nucleic acid of claim 2 , wherein the antibody variable region is at least 99% identical to SEQ ID NO:6.

9. The isolated nucleic acid of claim 2 , wherein the antibody variable region is at least 95% identical to SEQ ID NO:54.

10. An expression vector comprising the nucleic acid of claim 2 .

11. A host cell comprising the nucleic acid of claim 2 .

12. The host cell of claim 11 , wherein the host cell is selected from the group consisting of a bacterial cell, a eukaryotic cell, or and a mammalian cell.

13. An isolated nucleic acid comprising a nucleotide sequence set forth in SEQ ID NO:38, SEQ ID NO:35, SEQ ID NO:39, SEQ ID NO:36, SEQ ID NO:40, SEQ ID NO:37, SEQ ID NO:55, or SEQ ID NO:59.

14. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:38.

15. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:35.

16. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:39.

17. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:36.

18. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:40.

19. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:37.

20. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:55.

21. The isolated nucleic acid of claim 13 comprising the nucleotide sequence set forth in SEQ ID NO:59.

22. An expression vector comprising the nucleic acid of claim 13 .

23. A host cell comprising the nucleic acid of claim 13 .

24. The host cell of claim 23 , wherein the host cell is selected from the group consisting of a bacterial cell, a eukaryotic cell, and a mammalian cell.

25. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises SEQ ID NO:58.
