Pharmaceutical composition comprising Nanog shRNA, and method of using Nanog shRNA to treat cancer
The present description relates to an inhibitory RNA molecule, comprising an oligonucleotide that selectively knocks down expression a Nanog pseudogene expressed in many human cancers, a replicating viral vector capable of encoding such inhibitory RNA molecule, pharmaceutical compositions comprising said vector, and methods of treating cancer by administration of said pharmaceutical composition.
1. A conditionally replicating viral vector, comprising an inhibitory RNA molecule comprising an oligonucleotide that knocks down expression of NanogP8 wherein the oligonucleotide comprises a sequence selected from the group consisting of
SEQ ID NO:3 (5′-GAGGCAGCAGAGACCGCTGCATGCACTTCCAGCCA-3′);
SEQ ID NO:37 (5′-GAGGCAGCAGAGACCGCTGCATGCAGTTCCAGCCG-3′);
SEQ ID NO:8 (5′-TCTGACAGGAAGTGGCTGGAAGTGCATGCAG-3′) and
SEQ ID NO:34 (5′-TCTGACAGGAAGTGGCTGGAACTGCATGCAG-3′).
2. The viral vector of claim 1 , wherein the vector induces apoptosis of cancer cells.
3. The viral vector of claim 1 , wherein the vector inhibits cancer cell proliferation.
4. The viral vector of claim 1 , wherein the vector reduces tumor mass.
5. The viral vector of claim 1 , wherein the viral vector is packaged in a coat protein that specifically binds to malignant cells that express receptors for Ad5/3 chimeric fibritins.
6. The viral vector of claim 5 , wherein the malignant cell is one of a leukemia, a sarcoma, and a carcinoma.
7. The viral vector of claim 5 , wherein the malignant cell is a colorectal carcinoma (CRC) cell.
8. The viral vector of claim 1 , wherein the viral vector overexpresses an RNA that inhibits NanogP8 expression.
9. The viral vector of claim 1 , wherein the viral vector is a conditionally replicating adenovirus (CRAd).
10. The viral vector of claim 1 , wherein the viral vector comprises a minimal promoter operatively linked to the inhibitory RNA molecule, wherein the minimal promoter comprises a portion of the adenovirus E1a gene and a portion of the NANOGP8 promoter.
11. The viral vector of claim 1 , wherein the vector is a measles virus or a herpes simplex virus (HSV) vector.
12. The viral vector of claim 8 , wherein the inhibitory RNA molecule is a double stranded siRNA.
13. A pharmaceutical composition, comprising the viral vector according to claim 1 , and a pharmaceutically acceptable carrier.
14. A method of treating colorectral cancer (CRC) in a patient comprising administering to a CRC patient in need of such treatment a therapeutically effective amount of a viral vector, selected from the group consisting of:
a) a conditionally replicating viral vector, comprising an inhibitory RNA molecule comprising an oligonucleotide that knocks down expression of either Nanog or NanogP8 wherein the oligonucleotide comprises a sequence of
SEQ ID NO:3 (5′-GAGGCAGCAGAGACCGCTGCATGCACTTCCAGCCA-3′) or
SEQ ID NO:37 (5′-GAGGCAGCAGAGACCGCTGCATGCAGTTCCAGCCG-3′), and,
b) a conditionally replicating viral vector viral vector, comprising an inhibitory RNA molecule comprising an oligonucleotide that knocks down expression of NanogP8 wherein the oligonucleotide has a sequence of SEQ ID NO:8 (5′-TCTGACAGGAAGTGGCTGGAAGTGCATGCAG-3′) or SEQ ID NO:34 (5′-TCTGACAGGAAGTGGCTGGAACTGCATGCAG-3′).
