Abstract

In one aspect the present invention is directed to mutant NGAL proteins that have the ability to bind to siderophores, such as enterochelin, and to chelate and transport iron, and that are excreted in the urine. Such NGAL mutants, and complexes thereof with siderophores, can be used to clear excess iron from the body, for example in the treatment of iron overload. The NGAL mutants of the invention also have antibacterial activity and can be used in the treatment of bacterial infections, such as those of the urinary tract.

Claims (19)

1. An engineered polypeptide that comprises the amino acid sequence of SEQ ID NO: 253, wherein said polypeptide is able to bind to a siderophore, and is excreted in the urine of a subject after systemic administration of said polypeptide to the subject.

2. A pharmaceutical composition comprising the polypeptide of claim 1 .

3. An engineered K3Cys-N65A protein comprising the amino acid sequence of SEQ ID NO:253, wherein said K3Cys-N65A protein is able to bind to a siderophore, and is excreted in the urine of a subject after systemic administration of said protein to the subject, and wherein the “K3Cys” refers to the substitution mutation C87S of K3 Neutrophil Gelatinase Associated Lipocalin (NGAL) protein.

4. The K3Cys-N65A protein of claim 3 , wherein the K3Cys-N65A protein has bacteriostatic activity.

5. The K3Cys-N65A protein of claim 3 , wherein the percent of the K3Cys-N65A protein that accumulates in the urine after the systemic administration of the K3Cys-N65A protein to the subject is greater than:

(a) the percent of a wild-type (WT) NGAL protein that accumulates in the urine over the same time period after the systemic administration of WT NGAL protein to the subject; or

(b) the percent of K3Cys protein that accumulates in the urine over the same time period after the systemic administration of K3Cys protein to the subject.

6. The K3Cys-N65A protein of claim 3 , wherein the percent of K3Cys-N65A protein that accumulates in the urine three hours after the systemic administration of the K3Cys-N65A protein to the subject is about 50% or more.

7. The K3Cys-N65A protein of claim 3 , wherein the percent of the K3Cys-N65A protein that accumulates in the kidney of the subject after the systemic administration of the K3Cys-N65A protein to the subject is lower than:

(a) the percent of a WT NGAL protein that accumulates in the kidney over the same time period after the systemic administration of WT NGAL protein to the subject; or

(b) the percent of K3Cys protein that accumulates in the kidney over the same time period after the systemic administration of K3Cys protein to the subject.

8. The K3Cys-N65A protein of claim 3 , wherein the percent of the K3Cys-N65A protein that accumulates in a kidney of the subject three hours after the systemic administration of the K3Cys-N65A protein to the subject is 5% or less.

9. A pharmaceutical composition comprising the K3Cys-N65A protein according to claim 3 .

10. A pharmaceutical composition comprising a complex of the K3Cys-N65A protein according to claim 3 and a siderophore.

11. The pharmaceutical composition of claim 10 , wherein the siderophore is selected from the group consisting of enterochelin, pyrogallol, carboxymycobactin, catechol, and variants thereof.

12. The pharmaceutical composition of claim 10 , wherein the siderophore binds to the K3Cys-N65A protein and iron in the urine.

13. The pharmaceutical composition of claim 10 , wherein the siderophore binds to the K3Cys-N65A protein and iron in the blood of the subject.

14. The pharmaceutical composition of claim 10 , wherein the K3Cys-N65A protein and the siderophore are present in a 1:1 molar ratio.

15. The pharmaceutical composition of claim 10 , wherein the siderophore is pH insensitive.
