Abstract

The present invention relates to compositions and methods for characterizing, diagnosing, and treating cancer. In particular the invention provides the means and methods for the diagnosis, characterization, prognosis and treatment of cancer and specifically targeting cancer stem cells. The present invention provides an antibody that specifically binds to a non-ligand binding region of the extracellular domain of a human NOTCH receptor and inhibits growth of tumor cells. The present invention further provides a method of treating cancer, the method comprising administering a therapeutically effective amount of an antibody that specifically binds to a non-ligand binding region of the extracellular domain of a human NOTCH receptor protein and inhibits growth of tumor cells.

Claims (12)

1. An isolated monoclonal antibody that specifically binds a non-ligand binding region of the extracellular domain of at least two human Notch receptors.

2. The antibody of claim 1 , which specifically binds a non-ligand binding region of the extracellular domain of Notch2 and at least one additional human Notch receptor.

3. The antibody of claim 2 , wherein the at least one additional human Notch receptor is Notch1, Notch3, and/or Notch4.

4. The antibody of claim 2 , which binds Notch2 and Notch3.

5. The antibody of claim 1 , which specifically binds a non-ligand binding region of the extracellular domain of Notch1 and at least one additional human Notch receptor.

6. The antibody of claim 5 , wherein the at least one additional human Notch receptor is Notch2, Notch3, and/or Notch4.

7. The antibody of claim 1 , wherein the antibody is a chimeric antibody.

8. The antibody of claim 1 , wherein the antibody is a humanized antibody.

9. The antibody of claim 1 , wherein the antibody is a human antibody.

10. The antibody of claim 1 , wherein the non-ligand binding region comprises epidermal growth factor (EGF) repeats 1-10.

11. The antibody of claim 1 , wherein the non-ligand binding region comprises EGF repeats 13-36.

12. A pharmaceutical composition comprising the antibody of claim 1 .
