Abstract

The invention relates to compounds of Formula (I) and their pharmaceutically acceptable salts, wherein the substituents are as described herein, and their use in medicine for the treatment of diseases, disorders associated with the modulation of calcium release-activated calcium (CRAC) channel. The invention also relates to pharmaceutical compositions containing such compounds in treating diseases disorders associated with calcium release-activated calcium (CRAC) channel modulators. wherein, ring D is Formula (a) or Formula (b): A and B, which may be same or different, are independently CR 3 or N; Y is CR 3 or N; L is selected from —NR 2 C(O)—, —C(O)NR 2 — and —NR 2 CR a R b —; R a and R b are independently hydrogen, halogen or substituted or unsubstituted alkyl; ring E is selected from the Formula (i) to (vii).

Claims (259)

1. A compound of Formula (I):

wherein,

ring D is Formula (a) or Formula (b):

A and B, which may be same or different, are independently CR 3 or N;

Y is CR 3 or N;

L is selected from —NR 2 C(O)—, —C(O)NR 2 — and —NR 2 CR a R b —;

R a and R b are independently hydrogen, halogen or substituted or unsubstituted alkyl;

ring E is selected from the Formula (i) to (vii):

X is N or CR 4 ;

Z is N or CR where R is selected from hydrogen, halogen or substituted or unsubstituted alkyl;

R 1 , which may be same or different at each occurrence, is independently selected from halogen, cyano, nitro, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted haloalkoxy, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkoxy;

R 2 is hydrogen or substituted or unsubstituted alkyl;

R 3 is independently selected from hydrogen, halogen, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted haloalkoxy, substituted or unsubstituted cycloalkyl and —C(O)OR 6 ;

R 4 , which may be same or different at each occurrence, is independently selected from hydrogen, halogen, cyano, nitro, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted haloalkoxy, substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkoxy, —NR 8 R 9 , —COOR 6 and CONH 2 ;

R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted alkoxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl;

R 6 is a hydrogen or substituted or unsubstituted alkyl;

R 7 is selected from hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted alkoxyalkyl, substituted or unsubstituted cycloalkyl, and substituted or unsubstituted cycloalkylalkyl;

R 8 and R 9 are independently selected from hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, and substituted or unsubstituted cycloalkyl;

R c and R d are independently selected from hydrogen or substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkylalkyl, substituted or unsubstituted hydroxyalkyl and substituted or unsubstituted alkoxyalkyl; and

‘n’ is an integer ranging from 1 to 4, both inclusive;

or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 having the Formula (II):

wherein,

A and B, which may be same or different, are independently CR 3 or N, wherein R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —;

ring E is selected from the Formula (i) to (v):

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl and substituted or unsubstituted haloalkyl;

R 3 is selected from hydrogen, halogen, or substituted or unsubstituted alkyl;

R 4 is independently selected from hydrogen, halogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl;

R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl; and

‘n’ is an integer ranging from 1 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

3. The compound of claim 1 having the Formula (III):

wherein,

A and B, which may be same or different, are independently CR 3 or N, where R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —;

ring E is selected from the Formula (i) to (v):

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl and substituted or unsubstituted haloalkyl;

R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl;

R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl; and

‘n’ is an integer ranging from 1 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

4. The compound of claim 1 having the Formula (II):

wherein,

A and B, which may be same or different, are independently CR 3 or N, where R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —;

ring E is Formula (vi) or (vii):

X is N or CR 4 where R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl, and substituted or unsubstituted cycloalkyl;

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl and substituted or unsubstituted haloalkyl;

R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 , which may be same or different at each occurrence, is independently selected from hydrogen, halogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl;

R 7 is selected from hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl;

R c and R d are hydrogen or substituted or unsubstituted alkyl; and

‘n’ is an integer ranging from 1 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

5. The compound of claim 1 having the Formula (III):

wherein,

A and B, which may be same or different, are independently CR 3 or N, where R 3 is selected from hydrogen, halogen or substituted or unsubstituted alkyl;

L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —;

ring E is Formula (vi) or (vii):

X is N or CR 4 where R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl, and substituted or unsubstituted cycloalkyl;

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl and substituted or unsubstituted haloalkyl;

R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 , which may be same or different at each occurrence, is independently selected from hydrogen, halogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl;

R 7 is selected from hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl;

R c and R d are hydrogen or substituted or unsubstituted alkyl; and

‘n’ is an integer ranging from 1 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

6. The compound of claim 1 having the Formula (IV):

wherein,

Y is CR 3 or N, where R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

Z is CH or N;

L is —NHC(O)— or —C(O)NH—;

ring E is selected from the Formula (i) to (v):

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl and substituted or unsubstituted haloalkyl;

R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl;

R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl; and

‘n’ is an integer ranging from 1 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

7. The compound of claim 1 , wherein ring D is Formula (a) or Formula (b):

wherein A and B, which may be same or different, are independently CR 3 or N; Y is CR 3 or N; wherein each of R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl.

8. The compound of claim 1 , wherein ring E selected from the Formula (i) to (v):

wherein R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, and substituted or unsubstituted cycloalkyl; R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl.

9. The compound of claim 1 wherein ring E is Formula (vi) or (vii):

wherein X is N or CR 4 where R 4 is hydrogen, halogen or substituted or unsubstituted alkyl; R c and R d are hydrogen or substituted or unsubstituted alkyl; R 4 is selected from hydrogen, halogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, and substituted or unsubstituted cycloalkyl; and R 7 is selected from hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl, substituted or unsubstituted cycloalkyl and substituted or unsubstituted cycloalkylalkyl.

10. The compound of claim 1 having the Formula (I):

or a pharmaceutically acceptable salt thereof;

wherein L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —; Z is N or CH; R 1 is halogen or substituted or unsubstituted alkyl; ‘n’ is 1 or 2; ring D is Formula (a) or Formula (b):

A and B are independently CR 3 or N; Y is CR 3 or N; each of R 3 is independently hydrogen, substituted or unsubstituted alkyl; and

ring E is selected from (i) to (v)

R 4 is selected from hydrogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl; R 5 is selected from substituted or unsubstituted alkyl, substituted or unsubstituted haloalkyl, substituted or unsubstituted hydroxyalkyl and substituted or unsubstituted cycloalkylalkyl.

11. The compound of claim 1 having the Formula (I):

wherein L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —; Z is N or CH; each of R 1 is halogen or substituted or unsubstituted alkyl; ‘n’ is 1 or 2; ring D is Formula (a) or Formula (b):

A and B are independently CR 3 or N; Y is CR 3 or N; each of R 3 is selected from hydrogen or substituted or unsubstituted alkyl; and ring E is (vi) or (vii)

X is N or CR 4 ; R 4 is selected from hydrogen, substituted or unsubstituted alkyl and substituted or unsubstituted cycloalkyl; R c and R d are hydrogen or substituted or unsubstituted alkyl; and R 7 is hydrogen or substituted or unsubstituted alkyl.

13. A pharmaceutical composition comprising one or more compounds of Formula (I) according to claim 1 and one or more pharmaceutically acceptable excipients.

14. A method of treating, managing and/or lessening diseases or disorders, syndromes or conditions associated with the modulation of calcium release-activated calcium (CRAC) channel in a subject in need thereof wherein the method comprises administering to the subject a therapeutically effective amount of a compound of claim 1 or a pharmaceutically acceptable salt thereof.

15. The method of claim 14 , wherein the diseases, disorders, syndromes or conditions associated with the modulation of calcium release-activated calcium (CRAC) channel are selected from the group consisting of inflammatory diseases, autoimmune diseases, allergic disorders, organ transplant, cancer and cardiovascular disorders.

16. The method of claim 14 , wherein the disease is rheumatoid arthritis, multiple sclerosis and psoriasis.

17. The method of claim 15 , wherein the allergic disorders are selected from asthma, chronic obstructive pulmonary disorder (COPD) or respiratory disorders.

18. The method of claim 15 , wherein inflammatory diseases are selected from rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, psoriatic arthritis, chronic obstructive pulmonary disease (COPD), inflammatory bowel diseases, pancreatitis, peripheral neuropathy, multiple sclerosis (MS) and inflammation associated with cancer.

19. A process for the preparation of compound of Formula (I):

reacting a borate compound of Formula (1) with various halobenzamides of Formula (2) where X′ is halogen or OTf, to give compound of Formula (1) by using suitable reagents Pd(PPh 3 ) 2 Cl 2 , Pd 2 dba 3 , Pd(PPh 3 ) 4 , or Pd(OAc) 2 and suitable ligand 2,2′-bis(diphenylphosphino)-1,1′-binaphthyl (BINAP), xanthophos, or triphenylphosphine

method (B)

reacting a compound of Formula (3) where X′ is halogen or OTf, with compound of Formula (4) where P is pinacolatoboronate or stannane, to give compound of Formula (I) by using suitable reagents Pd(PPh 3 ) 2 Cl 2 , Pd 2 dba 3 , Pd(PPh 3 ) 4 , or Pd(OAc) 2 and a suitable ligand 2,2′-bis(diphenylphosphino)-1,1′-binaphthyl (BINAP), xanthophos, or triphenylphosphine

method (C), which comprising the steps of
