Abstract

Herein are described 1058 different bacterial taxa that were unique to either human, grazing mammal, or bird fecal wastes. These identified taxa can serve as specific identifier taxa for these sources in environmental waters. Two field tests in marine waters demonstrate the capacity of phylogenetic microarray analysis to track multiple sources with one test.

Claims (15)

1. A method for detecting the source of fecal bacteria contamination in an environmental sample, comprising the steps of

(a) providing an environmental sample to be tested;

(b) determining the presence of a plurality of taxa of fecal indicator bacteria in said environmental sample using a microarray comprising probes capable of detecting the source-specific taxa set forth in Table 1 (bird), Table 2 (grazer) or Table 3 (sewage); and

(c) identifying the environmental sample as having fecal contamination from bird feces, grazer feces and/or sewage if the plurality of taxa of fecal indicator bacteria determined to be present in said environmental sample in step (b) comprises at least 10% of the source-specific taxa set forth in Table 1 (bird), Table 2 (grazer) or Table 3 (sewage).

The method of claim 1 , wherein in step (c), the environmental sample is identified as having fecal contamination from bird feces, grazer feces and/or sewage if the plurality of taxa of fecal indicator bacteria determined to be present in said environmental sample in step (b) comprises at least 20% of the source-specific taxa set forth in at least one of Table 1, 2 and/or 3.

3. The method of claim 1 , wherein the step (b) determination of the presence of the plurality of taxa of fecal indicator bacteria present in said environmental sample is carried out by microarray probe hybridization.

The method of claim 1 , wherein in step (c), the environmental sample is identified as having fecal contamination from bird feces if the plurality of taxa of fecal indicator bacteria determined to be present in said environmental sample in step (b) comprises at least 20% of the source-specific taxa set forth in Table 1.

The method of claim 1 , wherein in step (c), the environmental sample is identified as having fecal contamination from grazer feces if the plurality of taxa of fecal indicator bacteria determined to be present in said environmental sample in step (b) comprises at least 20% of the source-specific taxa set forth in Table 2.

The method of claim 1 , wherein in step (c), the environmental sample is identified as having fecal contamination from human feces and/or sewage if the plurality of taxa of fecal indicator bacteria determined to be present in said environmental sample in step (b) comprises at least 20% of the source-specific taxa set forth in Table 3.

7. The method of claim 1 , wherein the environmental sample is an air sample, a water sample, a soil sample, or a mixture thereof.

8. The method of claim 1 , wherein the environmental sample is a water sample.

9. The method of claim 1 , wherein the environmental sample is a solid sample.

10. The method of claim 1 , wherein the step (b) determining the presence of a plurality of taxa of fecal indicator bacteria in said environmental sample comprises detecting a target sequence of the fecal indicator bacteria in the environmental sample.

11. The method of claim 10 , wherein the target sequence is a 16S rRNA gene sequence.

12. The method of claim 10 , wherein the step (b) comprises obtaining bacterial genomic DNA from the environmental sample.
